It was confirmed that when a piece of presumptive cephalic region of young blastoderm of the chick embryo is cultured in vitro combined with a fragment of the dorsal skin dermis of older embryo, differentiation of the lens takes place by way of a placode formation and invagination of the epiblast in spite of the absence of the optic vesicle. Immunofluorescent analysis revealed that the lens antigens are present in the lens structure induced in this way and that the synthesis of the lens proteins starts in a few cells in a thickened placode-like structure, before the full differentiation of the lens has taken place. Crystallin synthesis is always restricted to the lens rudiment and the newly formed lens, forming a striking contrast to the epiblast which remains undifferentiated.

Isolation of the Peptide

A decapeptide which had an inhibitory effect on blood coagulation was isolated from the degradation products of bovine fibrinogen by plasmin on BioGel P-10 column. The purification of the active peptide was carried out by ion-exchange chromatography using both Dowex 1×2 and Dowex 50×2 column. Amino acid composition of the active peptide was determined to be Ser 0.94, Glu 3.88, Pro 1.04, Ala 1.00, Leu 1.97 and Lys 1.03.

The amino acid sequence of a decapeptide which had an inhibitory effect on the blood coagulation was determined by Edman degradation, dansyl-Edman procedure, and hydrolysis by acid carboxypeptidase. The amino acid sequence of this peptide was determined as follows:
Ser-Gln-Leu-Gln-Glu-Ala-Pro-Leu-Glu-Lys.
