A new human cell line designated OMC-6 was established from a tumor of an endometrial stromal sarcoma. OMC-6 was successively subcultured 25 times in about 2 years.
The monolayer cultured cells appeared to be non-epithelial with prominent nucleoli and tendency to pile up without contact inhibition. Electronmicroscopically, cultured cells contained small mitochondria, dilated rough ER and abundant lysosomes. The population doubling time was 160 hours, the saturation density was 6.1×10
4 cells/cm
2, the plating efficiency was 8.5 % and the mitotic index was 3.0 %. Chromosome studies showed aneuploidy with a modal number of 40 and abnormal karyotype pattern. Transplanted tumor into the back of nude mice resembled the original tumor histologically. The OMC-6 cells produced TPA in culture media and the accumulated TPA titer in 25 days was 611 U/L. TPA was also demonstrated immunocytochemically in cultured cells.
This original TPA producing cell line (OMC-6) may be quite useful for studying biological characteristics of endometrial stromal sarcoma and for obtaining basic information for its management.
