  • The study of P-glycoprotein and combination chemotherapy with cyclophosphamide, adriamycin and cisplatin in ovarian cancer

    We examined the expression of P-glycoprotein in ovarian cancer and correlation with clinicopathological factors (age, clinical stage, histology, CAP chemotherapy effect). Tumor tissue from 29 primarily operated ovarian cancer patients with residual tumors which were evaluable for response to induction CAP chemotherapy was examined. The expression of P-glycoprotein was observed in 14 of the 29 specimens from tumor tissue obtained at primary surgery, but was not observed in 15 specimens. In the relation to clinicopathological factors, a statistically significant correlation was found between P-glycoprotein expression and response to induction CAP chemotherapy, with cases that were positive for P-glycoprotein showing a worse response (P=0.0094). Five of 18 (28%) patients showing a complete or partial response demonstrated P-glycoprotein expression, while 9 of 11 (82%) patients with no change or progressive disease had P-glycoprotein- expressing tumors, thus a strong correlation was observed (P=0.0078). No significant correlation was found between P-glycoprotein expression and age, clinical stage or histology. Five patients whose tumor tissue began to express P-glycoprotein after CAP chemotherapy did not show any response to subseqent chemotherapy, We concluded that the expression of P-glycoprotein could provide useful information in selecting chemotherapy.[Adv.
