    1) Correlation between serum ZTT and TTT reaction and serum, γ-globulin has been noticed. It was demonstrated, however, that these reactions have higher correlation with the product of relative mobility and γ-globulin level.
    2) It was suggested from the above results that contribution to the increase in serum ZTT and TTT reaction are stronger in γ-globulin with slower mobility than the faster one.
    3) The suggestion was confirmed by experiment with IgG separated from normal human sera.
    4) The above mentioned correlation was also observed in M-components of monoclonal gammopathies, especially in M-components composed of IgG.
    5) It might be reasonable to assume that the M-component did not have the character of the so-called “paraprotein.”

    A rapid disc electrophoretic method to obtain clear separation of serum lipoproteins was investigated.
    The recommended experimental condition for clearer separation into four fractions, i. e., α, β, pre-β, and chylomicron, was as follows:
    1. Concentration of the separating gel: ca. 3.5%.
    2. Method of staining: prestaining with saturated Sudan Black B for 0.5 to 1 hour, with serum to staining solution ratio of 10 to 1.
    3. Volume of sample applied: 15μl per tube.
    4. Electric current: 4mA per tube.

    Water-soluble and acid-soluble proteins were extracted successively with deionized water and dilute hydrochloric acid from the tissue residues of the peripheral nervous tissue (sciatic nerve) and the cerebral white matter of bovine, which had been preliminarily defatted with chloroform-methanol (2:1) mixture and acetone.
    1) The water-soluble proteins extracted from the peripheral nervous tissue mainly consisted of the fraction which corresponded in electrophoretic mobility to serum albumin, while, those from the central nervous tissue consisted of three fractions which corresponded to serum albumin and α- and β-globulins.
    2) The two components of the acid-soluble proteins from the peripheral nervous tissue were arbitrarily designated as L and N fractions. The electrophoretic mobilities of the L and N fractions corresponded to those of egg white lysozyme and serum albumin, respectively. The major part of the acid-soluble proteins from the cerebral white matter was arbitrarily designated as M fraction, which located in the midpoint between L and N fractions in the disc electrophoresis carried out at pH 4.2, but close to the L fraction in the thin layer polyacrylamide gel electrophoresis at pH 8.2.
    3) The L and N fractions of the peripheral nervous tissue and the M fraction of the cerebral white matter could be appreciably purified by ion exchange chromatography. Immunological activities of these fractions were estimated according to the severity of experimental allergic encephalomyelitis (EAE) and neuritis (EAN) induced in experimental animals. The M fraction showed a high potency of EAE induction, while, the L and N fractions showed a low potency.
    The N fraction actively produced the antibody, which was detectable by the double diffusion test. Antibodies against the L and M fractions were not proven by the double diffusion test, but by the complement fixation test.

    Determinations of haptoglobin types by immunoelectrophoresis and disc electrophoresis were described.
    The haptoglobin types of the normal sera by immunoelectrophoresis showed perfect agreement with the types obtained by disc electrophoresis. On the other hand, 21 of 110 pathologic sera showing markedly increased or decreased haptoglobin values were difficult to type by means of immunoelectrophoresis. In four pathologic sera the haptoglobin types as determined by immunoelectrophoresis showed disagreement with those revealed by disc electrophoresis.
    The haptoglobin types by immunoelectrophoresis were not affected by storage in the deep-freezer for at least six months nor by slight hemolysis.
    Disc electrophoresis showed apparent patterns of three haptoglobin types, and allowed accurate and convenient determination, although it was impossible to type the pathologic sera with haptoglobin values below 15mg/dl as determined by the single radial immunodiffusion method.

    Horse γ-globulin (2g) obtained from the antiserum specific to human α-fetoprotein was coupled to Sepharose 4B (settled volume 50ml) activated by the BrCN method of Porath et al.
    Ascitic fluid of a hepatoma patient was applied to the antibody column and α-fetoprotein was specifically adsorbed on the column. The adsorbed protein (50mg) was dissociated and recovered with an acidic buffer, 0.1M glycine-HCl, pH 2.8. The column could be used repeatedly more than 10 times without any loss of the adsorbing capacity. The preparation showed high homogeneity by electrophoretic and immunological analyses.
    Crystalline α-fetoprotein was obtained from the purified material in an ammonium sulfate solution.
