    Rats made diabetes through treatment with streptozotocin showed a relative mean value increase of 8.2-fold in serum alkaline phosphatase (ALP) activity at the 28th day following treatment. On the basis of electrophoretic mobility, this serum ALP was found to consist of principally small intestinal ALP isozyme.
    Next, to determine whether the ALP isozyme was of duodenal or ileal origin, the qualitative differences between the purified ALP obtained from tissues of both types were examined in detail. As a result, the serum and ileal ALP matched in terms of specific amino acid inhibition, substrate specificity, and molecular weight, while in examination of the sugar moiety, more of the fraction showing higher affinity to concanavalin A was found with serum ALP than with any of the intestinal ALPs. Serum and intestinal ALPs also differed slightly regarding isoelectric points.
    Form the above, it was suggested that the serum ALP of diabetic rats was ileal ALP, translocated into the bloodstream, of which the sugar moiety had been further modified.

    A relatively longer precipitin line in γ-globulin region was found in immunoelectrophoresis of seminal fluid using antiserum and this specific protein present in the seminal fluid was called as γ-seminoprotein (γ-Sm).
    γ-Sm was isolated and purified from seminal fluid by gel-filtration and isoelectrofocusing, and chemical property and γ-Sm level in seminal fluid were analysed.
    The results were;
    1. γ-Sm formed 5 to 6 bands in PAG electrophoresis.
    2. γ-Sm level in 15 seminal fluid samples was 500 to 1, 000mg/dl.
    3. Primary structure of γ-Sm was found to be consisted with 237 amino acid residues and possessed N-acetylglucosaminyl-asparagine linkage at 45 position.
    4. γ-Sm is one of serine proteases possessing active serine at 189 position from N-terminal.
    5. γ-Sm is an useful marker of seminal fluid in forensic medicine and also as a tumor marker for prostate cancer.
    6. γ-Sm was observed to be localized in prostatic cells by PAP method and the substance was demonstrated in perinuclear space, rough endoplasmic reticulum, Golgi apparatus, secretory granules and lysosome of prostatic epithelial cell by immunoelectron microscopic method.

    A new procedure for confirmation of focusing pattern after preparative agarose gel isoelectric focusing is described. The preparative gel was covered with a thin layer agarose gel (cover gel) from the start of focusing. A printing procedure can be omitted in this procedure since the cover gel is stained after focusing. The focused cover gel pattern is more clear than the printed cellulose acetate membrane. When the dry cover gel was used, the transcription volume was a little and the recovery of protein from the preparative gel was similar to the printing procedure with the membrane. In spite of removing carbonic acid gas with the utmost care, skewed protein bands were sometimes observed and its direction of skew was irregular. Therefore, it was assumed that there are some factors caused skewed protein bands besides carbonic acid gas.

    Antitumor effects and changes in electrophoretic mobility pattern of splenocytes and thymocytes were studied in sarcoma 180 or plasmacytoma X5563-bearing mice treated with Mitomycin C (MMC) and/or Krestin (PSK). The percentages of the inhibition of tumor growth were 80∼90% in the MMC-treated group and further improved in the MMC+PSK-treated group. In splenocyte electrophoretic pattern, intermediate mobility cells locating between high mobility cells (HMC: T cells) and low mobility cells (LMC: B cells) increased in tumor-bearer but recovered by treatment with antitumor drugs. On the other hand, in thymocyte electrophoretic pattern, LMC (immature thymocytes) which decreased by tumor-bearing were further reduced in MMC-treated group, but its reduction was suppressed by the combination with PSK. These results suggest that the combination therapy of MMC and PSK is more effective from the viewpoints of antitumor activity and antiinfectious activity because PSK restores immature thymocytes damaged by MMC.

    Quantitative analysis of protein components in the rat serum was done by a SDS-PAG electrophoretic method. The protein components were quantified by measuring the areas under peaks of the densitometric diagram obtained by scanning the electrophoretic lane in the gel. No systematic changes in the peak areas were found among lanes in the same gel and scanning positions in the same lane. Relations between the amounts of samples and the peak areas were linear in all protein components. Variations between the two measurements made twice at a 10 day interval were relatively great. It is concluded that relative amounts of proteins could be measured fairly accurately in the same gel, that the amount of a protein component could be estimated by comparing the unknown sample with a standard one, and that it would be necessary to make corrections using a standard sample to compare proteins among different gels.
