    Radionuclide therapy depends for success on the critical relationship between the amount of radionuclide in the target tissue and in critical normal tissue. The successful implementation of radioimmunotherapy for treatment of cancer has proven to be considerably more difficult than initially anticipated. Several groups have tried the magnetic guidance of antiplastic and antibacterial agents as well as X-ray-and MRI-contrast substances by means of magnetic particles. We propose to use magnetic nanospheres for the directed delivery of radionuclide to the tumor after intravenous administration of the biodegradable colloidal suspension. Using carriers of this size range the circulation time in the blood, the biodistribution and more or less the specific targeting are determined by uniformity of size, chemical composition, surface modification and the electric surface charge. The electrophoretic mobility is an important parameter for the prediction of the usefulness of the prepared particle, modified by chemical and biological molecules.

    Affinophoresis is a type of affinity electrophoresis in which an affinophore, a conjugate of an affinity ligand and a multiply charged soluble matrix, causes a mobility change in proteins that have specific affinity for the ligand. This technique enables, not only the separation of a specific molecule, but also the analysis of specific interaction between biomolecules. The affinity constant between fluorescence-labeled pea lectin and affinophores bearing mannoside as an affinity ligand, was determined by capillary electrophoresis using laser-induced-fluorescence detection. Although the labeling of the protein caused microheterogeneity as evidenced by electrophoretic mobility, the affinity constant determined for the main peak in the electropherogram of the labeled lectin agreed with those obtained for unlabeled lectin by the same type of analysis using ultraviolet-absorption detection. As a result of the use of the fluorescence detection, the consumption of the protein sample is greatly reduced to several pg per run.

    Separax 膜と反応するIgM-κ型M蛋白を見いだし検討を加えた. ジアセチルセルロースとトリアセチルセルロースを等量混合し油性成分を含まないセ・ア膜を用い電気泳動を行ったところ, 明瞭なM蛋白帯は認めず原点のα1からβ位に残る幅広い異常蛋白帯が観察された. 水酸基の多い電気浸透除去剤であるヒドロキシプロピルセルロースを含有した緩衝液によるSeparax 膜電気泳動では, Separax-SP膜と同様, 明瞭なM蛋白帯が観察され, 従来のセ・ア膜のアセチル基を水酸基に変えたミクロフィルターFR 100でも明瞭なM蛋白帯が観察された. 患者M蛋白と Separax 膜のアセチル基との反応が考えられ, セルロース膜による血液透析などの既往歴がまったくないことから, その反応様式は免疫学的な交差反応に基づいている可能性が考えられた.
  • IV. Anionic isotacophoresis of ampholytes using a common component which migrates faster than the leading component

    Our computer simulation procedure was applied to an anionic isotachophoretic system of ampholytes in which a component having a migration velocity faster than the leading component of the system was added to the catholyte. After a certain time of electrophoresis depending on the initial condition, isotachophoretic migration resulted even in the presence of the faster component in the catholyte. The isotachophoretic concentration of the ampholytes and the potential gradient were lower in the presence than those in the absence of the faster component. The lower potential gradient is preferable from the standpoint of Joule's heat so that the proposed isotachophoretic system seems to be useful for slab gel isotachophoretic experiments where thinning of the gel occurrs in the region of high potential gradient.

    We examined the prevalence of amylase-linked immunoglobulin complexes, known as macroamylase, in the sera of 146 patients with ulcerative colitis (UC). Six of them were found to have this complex. Combining this group with the group of 59 patients with UC examined previously in a similar fashion, provided a prevalence rate of 7 in 205 (3.4%). Of the 7 patients the binding immunoglobulin was IgG in 4, IgA in 2 and both in the remaining patient. This distribution of the immunoglobulins contrasts strikingly with a preponderance of IgA in patients with a variety of diseases. The results make definite the association between UC and circulating enzyme-linked IgG complex, and provide support for the hypothesis that autoimmune mechanisms are involved in the pathogenesis of UC.
