    The changes in dermis in the course of experimental Basal cell carcinoma and Squamous cell carcinoma on rat skin were observed histologically and histochemically.
    Remarkable accumulations of hyaluronic acid and sulfated mucopolysaccharides with the fibrous or mucinous connective tissue were noted in the superficial corium before the formation of experimental Basal cell carcinoma developed by painting 2-Anthramine. Therefore, we assume that the proliferation of tumor cells in Basal cell carcinoma were induced by not only carcinogenic action to epidermal cells but also such an acid mucopolysaccharides rich environment.
    Otherwise, much remarkable accumulation of hyaluronic acid and sulfated mucopolysaccharides in the localized area of invasiveness on experimental Squamous cell carcinoma were noticed. We concluded that squamous cell could be developed malignant tumorby painting 7, 12-Dimethylbenz-(α) anthracene, then stromal deffensive changes such as the accumulation of acid mucopolysaccharides and the connective tissue fibrosis would stand against malignant invasiveness. Therefore, malignant tumor cell probably would be able to invade by secreting mucolytic enzymes including hyaluronidase and others.

    In addition to the previously reported 8 cases of skin tumors, consisting of 2 cases of large genitoanal condyloma acuminatum, 2 cases of multiple Bowen's disease and 2 cases of solitary basal cell epithelioma of the face (Skin Research, 11: 404-435, 1969), a case of solitary basal cell epithelioma of the left ala nasi and a case of multiple basal cell epithelioma of the face in xeroderma pigmentosum were treated with 5% 5-fluorouracil cream under occlusive dressing technique (ODT). The results were quite encouraging as a new type of therapy especially for superficial skin cancers.

    The mechanism of bleaching effects hydroquinone and its derivatives in melanin formation was investigated with manometric and spectrometric procedures. Tyrosinase was prepared from Harding-Passey mouse melanoma and either L-tyrosine or L-dopa was used as a substrate. Surprisingly, it was found with a manometric procedure that hydroquinone accelerated the oxygen consumption in a dopa-tyrosinase system, while it inhibited O2-uptake in tyrosine-tyrosinase system. However, the final solution of the dopa-tyrosinase system with hydroquinone or its derivatives did not darken, and remained reddish brown in color, showing inhibitory effects of hydroquinone on melanogenesis. Moreover, a spectrometric procedure revealed that hydroquinone itself was oxidized through p-quinone to another compound by tyrosinase. There is a general agreement that the tyrosinase has two different activities, that is, cresolase and catecholase. These contradictory results, thus, would be interpreted that hydroquinone blocked the tyrosine hydroxylase phase of tyrosinase, coupling with tyrosinase directory in a tyrosine-tyrosinase system, while it did not block the dopa oxidase phase of tyrosinase, competing with L-dopa in a dopa-tyrosinase system.

  • Part 1: The Effect of the Irradiation of Ultraviolet Light on Sulfanilamide Solution

    1) Following the irradiation of ultraviolet light, an squeous solution of sulfanilamide elicits purple, accompanied by changes of maximal absorption spectrum from 260mu to 560mu in wavelength.
    2) No spot is found with the paperchromatography of the concentrated purple solution in n-butanol-acetic asid-water system (4:1:2), while a purple spot at the original point in a water-saturated n-butanol. It is negative in Bratton-Marshall color reaction.
    3) These changes are not observed in the other sulfa drugs than sulfanilamide. The purple substance is distinguished from p-hydroxylaminobenzene sulfonamide, a photoxidation product of sulfanilamide, in physicochemical characteristics.

  • Part 2: Dynamic Patterns in the Blood after an oral Administration of Sulfanilamide

    1) Blood levels of sulfanilamide were determined quantitatively after an oral administration of 2gm. of the drug in normal adults. The blood level was attained in a peak two hours after the administration, and gradually decreased in all of them. It was found that the conjugated form of sulfanilamide decreased more slowly than the free form. Thus, the concentration ratios of the conjugated from to the free form increased as the time elapsed.
    2) Plasma paperelectrophoresis was performed to investigate the binding of sulfanilamide with the plasma protein in vivo. A small amount of the compound bound with the albumin fraction was detected six hours after the administration.
    3) Binding of sulfanilamide with human serum albumin in vitro is very small in quantity, while in the case of p-hydroxylaminobenzene sulfonamide, a photoxidation product of sulfanilamide, this metabolite binds relatively strong with the albumin, increasing as the time elapsed.

  • Besonders zur Beziehung zwischen dem Vorgang des Autolyse-Phänomen und dem Mechanismus der Blasenbildung

    1) Die bei 37°C im destillierten Wasser aufbewahrte Achselhaut, die bei radikaler Operation der Osmidrosis axillae steril entgenommen wurde, zeigte selbst zuerst die histologischen Veranderungen der Pyknose mit perinucleärer Vacuolenbildung und der Chromatinverklumpung im Stratum Malpighi, später danach im Verlauf der Zeit konnten dazu subepidermale Spaltungen deutlich gefunden wurden, und endlich nach 9 tagiger Aufbewahrung das Phänomen der Acantholyse aufgetreten wurde.
    2) Durch Normalisierung des osmotischen Drucks sind die oben beschriebenen histologischen Veränderungen sich gelinden worden. Bei höherem osmotischen Druck fandman subcorneale und subepidermale Spaltungen ohne Veränderungen der Malpighi-Zellen, die bei nidrigem osmotischen Druck aufgetreten wurden.
    3) Durch die Beimischung in mäβiger Konzentration von verschiedenen Stoffwechselinhibitoren, z. B. Azid, Arsenit, Fluorid, Monoiodoacetat, Malonat und 2, 4-Dinitrophenol in der Locke-Lösung wurde der Vorgang des Autolyse-Phänomen zugenommen aber ebenf alls in 0.2%iger Konzentration von Traubenzucker dazu beimischten Lösung wurden die histologischen Veränderungen im allgemeinen gelinden.
    4) Durch die Wirkung der Hitze und des proteolytischen Enzyms (α-Chymotrypsin) wurde das Autolyse-Phänomen befördert, und besonders in hoherer Konzentration von α-Chymotrypsin nach lang-zeitiger Aufbewahrung sind die Veranderungen zu Epidermis sondern auch zu Kutis gekommen worden.
    5) Auf Grund dieser Untersuchungen kann angenommen werden, daβ der Vorgang des Autolyse-Phanomens sich auf dem Stoffwechsel des Gewebes viel bezohgen ist.
    6) Das Autolyse-Phanomen wurde von verschiedenen intrinsischen und extrinsischen Faktoren eingeflüβt, und Beziehungen zwischen Autolyse-Phänomen und Mechanismus der Blasenbildung sind diskutiert.

    In order to clarify the alteration of D-glucuronate to D-xylulose in the skin of rat, guinea pig and man, the four enzymes of this pathway, i. c. glucuronate reductase, Lgulonate dehydrogenase, L-xylulose reductase and D-xylulose reductase were determined.
    The evidences showed that, first, this pathway actually existed in those cutaneous tissues, second, in rat skin, the behaviour of this pathway would competite to ascorbic acid synthesis similar to rat liver, third, in experiments of pathological human cutaneous tissues, the higher activities of these enzymes than normal were obtained in malignant skin tumors, erythrodermas and chronic eczemas, but lower activities showed in ichthyosis vulgares and fixed drug eruption, however, in the lesions of psoriasis the deviation of the activities of each enzymes were significant.

    1) A case of amelanotic malignant melanoma on the fronto-parietal region of a 65-year-old man was reported.
    2) The tumor was amelanotic in almost of all regions.
    3) In light microscopical observation, two types of cells, a cuboidal and a fusiform type, could be recognized, and dopa reaction was strongly positive.
    4) Ultrastructure of the tumor cells revealed atypical premelanosomes in the cytoplasma. The dopa reaction was positive in premelanosomes, Golgi sacs and Golgi vesicles.
    5) On autopsy findings, numerous metastatic tumors could be seen in the lung and the liver, a few were seen in the heart, the brain, the kidney, the adrenal, the peritoneum and the spleen.

    Es wurde uber einen Fall von 64-jährigem Psoriatiker berichtet, an dessen Rücken ein groβer Stachelzellenkrebs und einige sog. chronische Radiodermatitis nach der fünf jahr-langen Grenzstrahlentherapie (Einfalldosis: 180γ, 0.3mA, 15KV, HWS: 0.4-0.5mm, und FHA: 10cm, wöchentlich) aufgetreten waren.
    Da diese Veranderungen von Überdosierung über 20000γ von Grenzstrahlen verursacht wurden, man muβ vorsichtig sein, Grenz-Strahlen zu benützen, obwohl Einfalldosis sowie auch Halbwertschicht richtig gewesen sind.

  • Developing from the Burning Scar with the Skull Invasion

    Two patients (case 1; sixty six years old, male, case2;thirty four years old, male), who were suffered from the squamous cell carcinoma at the skin of the scalp developing from burning scars got in childhood, were reported.
    Invasive changes of the skull were observed in both cases by X-ray examination. Case 1 was mainly treated Bleomycin injection, however, it showed no effectiveness. Case 2 had the total resection of the tumor including the invasive region of the skull, X-ray radiation therapy and the surgical transplantation of the skin, thereafter, he was completely cured.

    It was reported that a man, sixty seven years old who was suffered from an elevated pigmented macula on his left epigastral lesion, has presented rapid growth of the eruption in a year. Dermatohistopathologically, it was observed that tumor cell nests consisting of basaloid cells and prickle cells were existed and proliferated to the dermis besides the characteristic appearance of Bowen's disease. It was also noted that the above cells were shown to be rather atypical, and the basement membrane was partially disrupted.
