  • 飼育によるふ化幼生の浮游期間の推定

    This paper deals with the duration of the planktonic larval stages of the Tanner Crab, Chionoecetes opilio, estimated from rearing experiments of the larvae at different temperatures. Ovigerous female crabs were collected in moter trawl net which was operated in Wakasa Bay in the Japan Sea on January 11, 1969. The specimens were kept in a tank with approximately 500 liters of sea water at 7-13°C. The larvae hatched on January 27, 1969. As food, rotifers and newly hatched brine shrimp nauplii were given during the first and second stages of the larvae, and grown brine shrimp of the size from 5 to 10mm in total length during the megalopa stage. The rearing of the larvae was attempted at six different grades of temperature, ranging from 4 to 20°C, and the water temperature was regulated by the use of cold water and a heater on one or each side of the containers (Fig. 1). The highest temperature to keep the larvae alive for a long time was about 18°C. It was found that between the temperatures of 4 and 18°C, the warmer the water the shorter becomes the duration for one molting stage. The following relationship was observed between the number of days required for one molting (y) and the mean temperature of the water used for rearing (x); y=300.89/x1.1117 in the first stage zoea, y=367.45/x1.1619 in the second stage zoea. The integral temperatures for an intermolt were found to be as follows; 215-255 day•°C in the first stage zoea, 217-299 day·°C in the second stage zoea and 262-442 day•°C in the megalopa stage (Table 1). In a suspected duration of hatching season for this species of crab, the water temperature throughout the surface to the depth of 150m in the fishing grounds of Wakasa Bay was 11-13°C (Fig. 3). It was estimated from the relations above that the total duration of the planktonic larval stages in this stratum may be 63-66 days. In other words, the duration might be limited to within three months even though the larvae migrate up from or down to a stratum of cold water deeper than 150m when they rise to the surface in the first stage zoea, or when they sink to the sea-bottom in the megalopa stage or first adult type stage.
    As the carapace width (2.6-2.9mm) and the external appearance in the first molting stage in the specimens of adult form obtained in these experiments are nearly the same as those reported in the previous paper, it is considered that those reported previously were also specimens equivalent to this first molting stage (Fig. 2).

    An attempt was made to detect mercury in the body of fish killed by inorganic mercury in solution (HgCl2). The total mercury was determined by the Dithizone-Carbontetrachloride method. The 48 hour median tolerance limit of HgCl2 for gold-fish (Carassius auratus) lay between 1-0.5 ppm Hg and 5 ppm Hg was found to be fatal to gold-fish in about 10 hours (Table 1).
    Mercury content of control fish ranged 0.14-0.36γ/g Hg. The bodies of fish killed by various concentrations of HgC12 showed values higher than 2.3γ/g Hg, while those living in 0.5 ppm Hg solution for 48 hours showed 1.2-1.8γ/g Hg. Gold-fish died in 0.5 ppm Hg solution within 6 or 7 days, and 2.3-4.6γ/g Hg was detected from the dead fish.
    When the body of fish killed by mercury poisoning was washed with running tap water for 24 hours after death, the mercury content of the body decreased to about 18.2-50.7% (Table 2).
    The distribution of mercury in seven parts of fish body was examined in two cases. In the 1st case, fish was killed by Hg in solution, and in the 2nd case, fish was soaked in Hg solution after death from suffocation. In the former, gill always showed the highest value and kidney came next (Table 3). In the latter, however, mercury content showed low values in every part examined. It is interesting to note that mercury was scarcely detected in spleen and kidney in the latter case.
    Some species of aquatic animals in the Abukuma and the Hirose-Natori Rivers were tested for mercury content to find the distribution of mercury in the natural aquatic environment (Table 6). The maximum value of mercury content was 3.72γ/g Hg in Zacco platypus. Almost all species, however, showed values lower than 1γ/g Hg. This means that there was no possibility of mercury pollution in both rivers.

    Actomyosin was extracted from the dorsal muscle of carps.
    The protein solution was kept frozen at -20°C or freeze-dried with sugar (sorbitol or sucrose).
    Attempts were made to determine the required ratio proportions for protecting the actomyosin from denaturation by measuring the changes in ATP-ase activity and solubility against 0.6M KCl, pH 7.0.
    It was observed that the sugars added 3 times by weight of actomyosin markedly protected the carp actomyosin from denaturation for longer period of storage at -20°C or by the freeze drying process. No special effect with sodium polyphosphate was found when added at 0.3 times the weight of actomyosin.

    Fundamental studies on the viscosity of carp muscular proteins were made to evaluate their ATP-sensitivity.
    Although the viscosity of carp actomyosin and myosin in 0.6 M KCI, pH6.8, varied slightly from preparation to preparation, it was found that there was a proportionality between the protein concentration (C) and the logarithms of relative viscosity (log ηr) before and after the addition of ATP and MgC12.
    Because the viscosity numbers (log ηr/C), with and without ATP, were independent of their protein concentrations, namely a constant independent of concentration, the ATP-sensitivity was evaluated.
    It was, therefore, recognized that the characterization of actomyosin and myosin by means of the ATP-sensitivity test can successfully be applied to carp muscle.
    The ATP-sensitivity is defined as r-r, ATP/Zη, ATP×100 by PORTZEHL et al. where Zηr and Zηr, ATP are the viscosity numbers before and after the addition of ATP respectively.

    The acid-soluble nucleotides, nucleosides and bases were extracted from the squid liver, Ommastrephes sloani pacificus (STEENSTRUP) with cold perchloric acid. They were separated by anion exchange chromatography (Fig. 1) and paper chromatography, and identified by using ultraviolet spectra, paper chromatography and by determinations of phosphate and ribose as described previously. The compounds detected were as follows: AMP, ADP, ATP, UMP, UDP, UDP-N-acetylglucosamine, UDP-N-acetylgalactosamine, IMP, CMP, GMP, GDP, CDP-derivative, NAD, NADP, ADP-ribose, ATP-ribose, nicotinamide mononucleotide, adenine, adenosine, uracil, uridine, hypoxanthine, inosine, xanthine, guanosine, nicotinamide and nicotinic acid. The contents of total adenine nucleotides, uracil nucleotides and pyridine nucleotides were 0.5-0.6, 0.3-0.6 and 0.1 μmoles per g of wet weight, respectively (Table 2). The UDP-N-acetylhexosamine predominate in uracil nucleotides, while UDP-hexose and UDP-uronic acid were not detected in the squid liver. The nucleotides pattern on the chromatogram was similar to that of the hepatopancreas of abalone.

    Feeding experiments lasting 30 days have been carried out with diets containing varying levels of casein to see the utilization of dietary protein by young carp. The composition of the diets used is shown in Table 1. Within a range of 0.4 to 55.0% of crude protein in the diet, the increase in body weight was in proportion to the content of protein, whereas the accumulation of protein in the body was found to reach the definite value when the crude protein level in the diet was close to 38% (Figs. 1, 2 and Table 2). From these results it is considered that the mode of weight gain and protein accumulation by carp is considerably different at higher levels of protein in the diet. The protein accumulation rate (protein accumulated/protein consumed) showed its highest value at near 21% of protein level, while the protein utilization value (protein accumulation rate × protein content of diet) attained its highest value at 38% of protein level (Table 3). These results show that the requirement by young carp for dietary protein is about 38% of protein in the diet when casein is used as the sole protein source. The relationships between the protein content of diet and the conversion factor, feed efficiency, protein efficiency ratio (PER) and net protein utilization (NPU) were determined and the results are shown in Figs. 2, 4 and 5.
