    The experiments were carried out using model nets of different construction. The effect of time, from the laying of the net till the start of pursing (waiting time), and that of the direction of current on the working depth were observed.
    The results are summarized as follows:
    1) The bottom margin of the net achieved maximum working depth at the 4/10 or 5/10 stage (e=0.4-0.5) of the pursing operation. To sink the leadline to maximum depth, pursing should be started within a certain amount of time after setting the net.
    2) The working depth of the leadline was found to be deepest when there was no current, and while under current, the leadline was deeper in a loose set than in a tight set.
    3) The model nets which were constructed with webbing of small resistances, or made of heavy twine, and tightly hanged showed fairly good sinking performances under the current conditions.

    The drag coefficient of a plane net pulled parallel to the direction of motion was estimated.
    Results obtained are summarized as follows:
    1) The drag coefficient, D8 of the plane net pulled parallel to the direction of movement is formularized; the parameters of which were determined by the least square method using the experimental results and is as derived from the following equation.
    D8=0.37(D/L)-0.58(D/L)3/2. where D is the diameter of a twine, and L is the length of a bar.
    2) The drag of plane net pulled parallel to the direction of movement is reduced due to the wakes formed behind the bars.

    The gut often gets cut when a fish is hooked during trolling operation. Accordingly, for trolling gear design it is necessary to investigate the strength of the looped part of the gut against impact load. The present study was carried out using an experimen-tal apparatus and dealt with the relation between impact load and tension of trolling line. Relationship between impact load (Wv) and tension (T) can be roughly expressed as
    T= 1.97Wv+4.5.
    The impulsive strength of the samples was about 25% loss as compared with the tensile strength of the same samples.

    It is important in the design of trolling gear to measure the pulling force it will have on the fish (skipjack and frigate mackerel) when caught.
    The present experiments were carried out using the trolling gears off shionomisaki during the months of May 1969 and 1970.
    The results obtained are summarized as follows:
    1) There was no relationship between the pulling force shown by the fish caught (frigate mackerel) and the length of the troll line in “Subiki”.
    2) There was no relationship between the pulling force by fish caught (skipjack) and the trolling speed.
    3) The following equation was delived: Tmax=3.31W+0.51
    where Tmax is a regressive value of maximum tension and W is body weight of fish.

    In some species of squids, it had been found out that the exhaustion of their generative activities after copulation and spawning leads to death. With the intention of expressing numericaly the maturation and exhaustion of the common squid, Todarodes pacificus STEENSTRUP, the present report deals with weight changes of body, liver, and genital organs in relation to the maturity condition that was proposed in the first report. The materials used were composed of autumn-spawned squids caught in the southwestern part of the Japan Sea, during November 1968 to October 1970. The results obtained are summarized as follows:
    1. Exhaustion could be first seen by the loss of liver weight and subsequently by the loss of body weight. The emaciation of genital organs could be noticed at the final stage of exhaustion.
    2. In males, following copulation loss in liver weight was substantial and the livers in the spent stage (0.85 in maturity condition) were exhausted to the extent that their weights were less than a quater of those in the immature stage (0.15 in M). Females also showed reduction in their liver weights after copulation (0.25 in M) but they soon regained it (0.35 in M). Nevertheless, spawning made them lose liver weights once again (0.75 in M).
    3. The influence of copulation and spawning on body weight was so severe that the consumption of reserved nutrients in the liver and other body substances was hardly adequate and led the squid to death by general prostration, while retaining some spermatophores in Needham's sac (male) or eggs in oviducts (female) not spawned out.

    A new underwater telemetry apparatus was tested successfully in the field to record the track of a swimming fish and the temperature of its surrounding water.
    Responding to the 75KHz signals (once per second) from the base, the transponder tagged on the fish sends back responses of 50KHz, which contain temperature information, to the three receivers at the base.
    The position of the fish is calculated by the computer in the apparatus and plotted by a X-Y recorder. Simultaneously, the depth and surrounding temperature of the tag are recorded.
    The field tests in a lake showed that the accuracy of position recording was about ±5% and that the effective distance and angle to search a tag were ca. 600m and ca. 50° respec-tively.
    Some examples of fish tracking in the field and its problems are described.

    The conformation of the protein in “Kamaboko” was investigated by infrared absorption and X-ray diffraction studies, and the following results were obtained.
    1) Infrared absorption spectrum of “Kamaboko” showed an amide I band at 1680-1630cm-1 and an amide II band at 1550-1520cm-1.
    2) X-Ray diffraction pattern of Kamaboko showed two reflections at 2θ=28.5° and 18-19°, where θ is the angle of diffraction.
    On the basis of these results, it is proposed that there exists a β-chain of protein, in addition to the α-helical and random coils, in “Kamaboko”.

    Kombu, a palatable seaweed belonging to Laminariaceae, has often been used as a seasoning material for soups or vegetables in authentic Japanese cookery. The soup is especially rich in nutrients for bacterial growth. Therefore studies on the food hygiene of kombu extract were undertaken.
    The growth of bacteria adhering to the surface of kombu was observed in an extract of the plant.
    The following food poisoning bacteria, Salmonella enteritidis, Staphylococcus aureus, Vibrio parahaemolyticus, Clostridium perfringens and Cl. botulinum, were inoculated in kombu extract or artificial kombu medium.
    When the bacterial counts of cultures in natural kombu medium were compared with those in artificial medium, no remarkable differences in growth were observed in any of the bacterial species used.
    Judging from the experimental results, attention should be paid to contamination with S. enteritidis and St. aureus. With respect to the other three food poisoning bacterial species, the extract was considered to be relatively safe in food hygiene under the experimental conditions.

    In the previous report, the authors clarified the distribution of nitrogen fixing bacteria in the sea water of coastal regions. The present paper deals with the occurrence of the bacteria in the water of open oceans, and in the bottom sediments of sea water environments.
    The standing crop of nitrogen fixing bacteria was quite small in the surface waters of the Mid-Pacific Ocean and the Japan Sea, i.e. at most 200 cells/L and 400 cells/L, re-spectively. Nitrogen fixing bacteria occur more abundantly in the bottom sediments than in the water, namely 1-10 cells per 1g of wet mud in offshore regions and open sea such as Suruga Bay, Sagami Bay and East China Sea.

    In the previous papers, the authors have reported the distribution of nitrogen fixing bacteria in natural water environments. This paper concerns the occurrence of the bacteria in fish culture ponds, in which the concentration of nutritional salts is extremely high, in addition to the large population of total heterotrophic bacteria, i.e. 104-106 cells/mL in the water and 106-107 cells/g in the bottom sediments.
    Even in such eutrophic water regions, the standing crop of nitrogen fixing bacteria was not larger than that in the other water environments, namely 0.1-10 cells in 1mL of the water. A diurnal change in the number of the bacteria was not observed in spite of the considerable fluctuation of the physical environment in these ponds. In the bottom sedi-ments of the culture ponds, the bacterial standing crop was 102-103 cells/g, which was com-parable with that in natural eutrophic ponds.

    By using the SUTO medium, effects of trace metals on the growth and nitrogen contents of Porphyra yezoensis were examined. The fronds were reared for 3-5 days and analyzed for total nitrogen, protein nitrogen and carbohydrate.
    1) When kept in a medium lacking trace metals, the fronds showed very low increase rates of solids, protein nitrogen and total nitrogen, and a higher carbohydrate content.
    2) Zinc was found to be essential for the growth and increase of protein nitrogen of the fronds. The optimum concentration was approximately 0.03 ppm.
    3) For the growth and formation of protein, the optimum quantity of the Pl solution (trace metal solution) to be added to the SUTO medium was found to be 2ml/l for the young buds ranging from 0.5 to 1cm in length and 1ml/l for the fronds rang-ing from 5 to 7cm, respectively.

    An apparatus for wet ashing of algal materials was devised to prevent the loss of selenium. The recovery was 93 to 97%, when 1.0g of the dried laver containing 1.0 to 5.0μg of selenium was analyzed with apparatus. The selenium contents of 17 species of air-dried marine algae, which had been kept for 2 years, ranged from 0.1 to 1.1ppm. In 6 fresh specimens of 4 species of marine algae, the values ranged from 0.35 to 1.3ppm and were nearly proportional to their nitrogen contents. Approximately 90% of selenium in Porphyra yezoensis was found in the fractions insoluble in both hot 70% ethanol and hot water, and 77% of it in the protein fraction precipitated at 70% ethanolic concentration from the cold water extract.
    The optimum concentration of selenium in the medium was found to be 0.003ppm for the growth and nitrogen accumulation in P. yezoensis.

    An 0.5% KCl extract of carp muscle was assayed for cathepsin A, B and C activities and the existence of these cathepsins in carp muscle was recognized. In comparison with a proteinase (cathepsin D) which hydrolyzes protein substrate at about pH 3.0, these cathepsins act in a higher (more physiological) pH range.

    A blue substance could be isolated from an alkaline extract of the blue meat of crab muscle by adjusting the pH of the extract to various values. It was shown that the blue substance contained a high level of copper and showed a positive haemocyanin-like reaction. Moreover, the chemical nature of the copper in the substance, as indicated by reaction with imidazole reagent in the presence of EDTA, was similar to that of king crab haemocyanin. From the present findings, it is suggested that the causative sub-stance of the blue discoloration of canned crab meat is a haemocyanin derivative.

    The haemocytes of haemolymph from living crabs agglutinated with the passage of time. During agglutination many granules appeared in the cytoplasm after which the cell membranes ruptured. A gelatinous clot of plasma around the agglutinated haemocytes was found 15-20min after the onset of clotting, The gelatinous clot was also found in crab muscle tissue. Haemocyanin held in the network structure of the gelatinous clot may well be responsible for the blue discoloration. In squid haemolymph, however, only agglutination of haemocytes without subsequent gelation of the plasma was observed.

    Utilization of the dietary sterols by prawn, Penaeus japonicus, was studied.
    After 30 day-feeding on the diets containing only cholesterol, ergosterol, stigmasterol, and β-sitosterol, the content and composition of sterols in the tissues of the prawn were analyzed. In the prawn fed on the sterol-supplemented diets, the sterol content of the body tissues was similar to that of the prawn before feeding though in the prawn fed on the sterolfree diet, the sterol content decreased. In the prawn fed on several sterol-supplemented diets, 96-99% of the sterol content was composed of cholesterol.
    From these results, it may be assumed that the dietary ergosterol, stigmasterol, and β-sitosterol were digested by the prawn, and converted to cholesterol.
