Whale nasal cartilage was liquefied by Pronase digestion, and mucopolysaccharides were prepared by Dowex 50W-X 1 treatment and precipitation with six volumes of ethanol in the presence of 0.1M sodium acetate.
The behaviors of chondroitin sulfate (CS), hyaluronic acid (HA), and keratosulfate (KS) in the fractional precipitation of the mucopolysaccharides with ethanol and in the ion exchange chromatography of each fraction on Dowex 1 were examined, the following results being obtained.
1) The mucopolysaccharides were fractionally precipitated at ethanol concentrations of 36, 39, 44, 50, 60, 70, 80, and 85% in the presence of 2M sodium acetate. Thus, Frs. 1-8 were obtained.
2) Frs. 1 and 2 were composed of CS-A and a small amount of CS-C; Fr. 3, CS-A and small amounts of CS-C and HA; Frs. 4 and 5, CS-A and small amounts of CS-C, HA and KS; Fr. 6, KS and a small amount of CS-A; and Frs. 7 and 8, almost KS.
3) A large portion of HA in Fr. 3 was separated from CS by the chromatography on Dowex 1. But only half of KS in Fr. 5 was separated from CS, and the separation of KS from CS in Fr. 6 was difficult.
