    Two species of righteye flounders, Limanda herzensteini (173-243mm in length) and L. yokohamae (211-271mm in length), were reared under various water temperature conditions, and the effects of temperature on daily feeding rate and their survival were examined. Although L. herzensteini did not feed at temperatures below 0.7°C, no deaths were observed at the lowest rearing temperature (-0.9 ?? -1.3°C). L. yokohamae could also tolerate long-term exposure (approximately three weeks) to subzero temperatures (up to-0.5°C), but they ceased to feed at 1.6-1.9°C. At temperatures below 18-19°C, the daily Feeding rate (%wet body weight per day) of L. herzensteini increased with increasing temperature and began to decrease remarkably at about 19°C, while in L. yokohamae the temperature at when the daily feeding rate began to decrease was about 25°C. In addition, heat resistance experiments suggested that the upper lethal limits of temperature of L. herzensteini and L. yokohamae lay in 25-26°C and 28-30°C, respectively.
    From these results, it Became evident that the thermal resistance of these two species which often occur sympatrically in the coastal waters of Japan differs considerably, that is, L. herzensteini is superior in cold resistance, but is inferior in heat resistance to L. yokohamae.

    Morphological characteristics of four spawning groups of Japanese jack mackerel Trachurus japonicus, spring and summer spawning groups of the Kuroshio Current System and winter and spring-summer spawning groups of the East China Sea and Tsushima Current System were com-pared.
    Although there are differences of the allometric growth among three length ranges, in general the spring and spring-summer spawning groups are larger than the summer and winter spawning groups respectively in head length and upper jaw length, etc. The larger fish of the summer spawning group is larger than the spring spawning group in head length and upper jaw length, etc. and smaller in body depth and pectoral fin length, etc.
    Morphological differences seen between spring and summer spawning groups and spring-summer and winter spawning groups were also discussed with the differences between 'kiaji' and 'kuroaji' formerly studied around Wakasa Bay.

    Larger individuals of Trachurus japonicus of the summer spawnlng group in the Sagami Bay region have been distinguished into two types, 'nodoguro' and 'maaji' by fishermen, etc. These two types were compared morphologically.
    There are no significant differences between them for each of nine meristic characters (t-test, p>0.05).
    Sixteen morphometric characters and the fork length were examined by covariance analysis. In head length, eye diameter, upper jaw length, length from the snout to the origin of unpaired fins, head depth, pectoral fin length and height of anterior scutes, nodoguro type is larger than maaji. While in body depth, length of fin bases and length of posterior part of lateral line, maaji type is larger than nodoguro type.

    The longitudinal distribution of average temperature, salinity and transparency in the Seto Inland Sea were described based on the existing data obtained during a period with large meander of the Kuroshio (August, 1975-July, 1980) and during a period with no meander (1966-1974). respectively.
    It became clear that average salinity during the meander period showed lower value than that during no meander period in the western part of the Seto Inland Sea, contrary to the eastern pazt where average salinity was comparatively high when the meander was present. These dif-ferences in salinity could not be explained well by the differences in precipitation and river dis-charge.
    The salinity difference between the Kuroshio meander and no meander periods was large in the eastern Bisan-Seto where the lowest salinity was generally observed, and westward shift of the water with salinity lower than 31‰ during the meander present seemed to be closely related to the above salinity differences. It was suggested that a certain change occurred in the advective con-dition of the Seto Inland Sea water due to the Kuroshio meander.

    Acquisition of auditory discrimination and orientation response in carps was investigated under several reinforcement conditions. In the same-side condition, the sound-producing speaker and the reinforcer of food or electric shock were adjacent. The fish in this condition learned to obtain food by swimming toward the sound-producing speaker and to avoid electric shock by swimming in the direction away from the sound-producing speaker. In the opposite-side condi-tion, the reinforcer of food or electric shock was opposite to the active speaker. The fish in this condition had a great difficulty in acquiring acoustic orientation, though the rate of learning in the food reinforced initial training and “go-no go” discrimation was similar to that for the same-side group. Another experiment, in which fish were trained to approach the correct feeding place by discriminating two kinds of the sound signal, showed that the fish was able to associate the two sound signals with the respective correct feeding place. These results were discussed in relation to utilization of cues in the orientation response in carp.

    The distribution, external surface morphology and ultrastructure of taste buds (TB) in the minnow, Pseudorasboraparva, were studied using scanning (SEM) and transmission (TEM) electron microscopy. SEM analysis revealed that TBs were widely dispersed on the skin of the head and lips as well as the mucosa within the oropharyngeal cavity, esophagus and branchial apparatus. The density of TBs was especially high on the lips, gill rakers, and palatal organ, reaching over 140 per mm2. Each TB showed two kinds of processes on the apical surface: thick rod shaped processes and microvilli.
    TEM analysis showed the presence of three distinct cell types in each TB: tubular (t), filament-ous (f) and basal (b) cells. Their general morphology appears to be homologous to that of other species of fishes. The most striking find of this study, compared to previous studies of the mor-phology of fish TBs, is that a rod shaped process of t-cells contains tubules. Adirect opening ofthe tubules to the outer surface was sometimes observed at the base of the process. Typical synaptic contacts were seldom found, but somewhat thickened pazallel membranes with a few or no vesicular accumulation were sometimes found between nerves and either t-or b-cells.

    A mathematical model is presented for describing the behavior of fish in a water tank. The state equation includes nonlinear terms representing the main causes for fish motion, and has five non-measurable parameters. The parameters are estimated by using the observation data of a water tank experiment and by applying the least squares algorithm. The validity of the model is in-vestigated by testing the whiteness of the residual of the state equation, and also by comparing the simulation result with the experimental result. Consequently, the modeling procedure becomes clearer than the previous one, and a detailed part of the model is improved. Further, it is found from the result of parameter estimation that about a thousand points of time series data are needed for the convergence of the estimation process.

    Life cycle in adult stage of the Sakhalin surf clam Spisula sachalinensis in Sendai Bay was studied based on the interrelation between reproductive and feeding cycles. The reproductive cycle is inversely related to the water temperature. Reproduction (gonad growth and spawning) occurs primarily under the low temperature regime and the reproductive activity stops after spawn-ing for about six months during summer-autumn. Seasonal change in weight of the digestive organs is caused by the active feeding in winter-spring.
    The life cycle of surf clam may comprise of two phases: phase of high activity (Dec.-May) and that of low activity (June-Nov.). The synchronism between gonad growth and active feeding is the essential trait of life cycle of the surf clam, suggesting that resources from outside is directly used for the gonad growth without temporary reservation. The surf clam in Sendai Bay seems to concentrate its activity in winter through the process of adaptation to the environment.

    The present report deals with the growth of the tidepool gunnel Pholis nebulosa (Pisces: Pholididae) examined by the length-frequency data, based on 4, 243 individuals taken from Ago Bay during the period from April 1980 to May 1982. The length frequency distribution of each month could be analyzed into 0- to 3-age groups. The length growth curve Of the gunnel was described by fitting von Bertalanffy's model expanded by a periodic function as Lt=33.8(1-exp(-0.72(0.53t+0.099+0.075 sin(6.28t-8.47)))), where Lt=total length (cm) at age t, and t=the passage time (year) from December before hatching. The relationship between total length (TL, cm) and body weight (W, g) was expressed by the formula: log10 W=3.48 log10 TL-3.045. The length growth was intensive at the water temperatures between 10 to 17°C, and this species grew more rapidly in rather cool periods than in the warm seasons.

    Seawater adaptability of chum salmon Oncorhynchus keta fry, weighing 1.5-6.O g, was ex-amined after treatment with thyroxine or thiourea for various periods. Thyroxine or thioureawas administered either orally (250mg L-thyroxine Na salt or 100 mg thiourea/kg diet for 14 days)or by immersion (20mgL-thyroxine or 10g thiourea/100 Iiters for 14 days). The change in plasma Na was followed for 48 h after transfer of the fry to seawater. Although neither thyroxinenor thiourea had any consistent effect on seawater adaptability of the chum salmon fry, thyroxine appears to have effects, if any, on smaller fry.

    The influence of inoculative quantity of infectious pancreatic necrosis virus (IPNV) on mor-tality of rainbow trout fry Salmo gairdneri was studied. IPNV-Buhl strain which is the most dominant IPNV-strain in Japan was used for a challenge virus and a highly sensitive strain to IPNV (RT-101 strain, 0.13 g and 0.26g in the mean B. W.) was employed as an experimental fish in this study. The fry was immersed with 105.0, 104.0, 103.0, 102.0 and 101.0 TCID50/ml of IPNV at 15°C for 1h and incubated at 15°C for 28 days. As the results obtained, cumulative mortality in the fry weighing 0.13g was 92, 84, 80, 51 and 26% and in the fry weighing 0.26g was 61, 38, 26, 0 and 0%at 105.0, 104.0, 103.0, 102.0 and 101.0 TCID50/ml of IPNV, respectively. At 105.0 TCID50/mlof IPNV, the peak of daily mortality was on the 4-6th day after inoculation and the mortalitypattern was a typical acute type. The daily mortality became lower, the peak of the daily mortalitywas delayed and the death of fry continued until later, as tine inoculative quantity of IPNV wasreduced.

    Effects of 5 different pkotoregimes on otolith increment formation were observed in embryonic and larval rainbow trout. Fertilized eggs were incubated within 4h under photoperiods of 6-h light and 6-h dark (LD 6:6), LD 12:12, and LD 24:24 at about 10.3°C Other eggs at l week post-fertilization were incubated under constant light (LL) or constant darkness (DD). They were sampled at hatching and swim-up, and the number of otolith (sagitta) increments were counted under transmitted light. Sagittae first appeared as multiprimordia 1 or 2 days before the initia-tion of eye pigmentation and the initial increment was laid down at the eyed stage. The mean counts of otolith increments were highly correlated with each event of the light-dark cycles and expressed by the regression lines: Y=1.96X+29.25 for LD 6:6, Y=1.04X+14.39 for LD 12:12, and Y=0.53X+6.83 for LD 24:24 groups, where Y signifies the count of increments and X re-presents the number of days elapsed (X=O at hatching). Increments were also found under LL and DD, but the counts were variable for each individual. The mean rate of increment deposi-fion from hatching to swim-up was about 1.2 and 1.4 per day in the LL and DD groups, respec-tively. These results indicate that photoperiod works as a potent entrainer for the rhythmic formation of otolith increments in embryonic and larval rainbow trout. Some stimuli other than photoperiod may also be capable of inducing otolith increment formation.

    The growth increments in otolith were validated to establish the age determination method of the ayu Plecogdossus altivelis an amphidromous salmonoidei fish with 1 year life span.
    Newly hatched larvae have ca. 5 embryonic increments in a circular sagitta of ca. 15μmradius. No mark peculiar to the time of hatch was detected in sagitta. Sequential samplings of known age fish from a rearing pond and those released in a river demonstrated that the days after hatching and the number of sagittal increments outside the 15μm radius presented close linear regressions with the slopes of 1.007 (1-150 days), 0.950 (120-180 days) and 0.871 (215-355 days), suggesting a daily deposition of the increment in sagittae. This was supported by the tetracycline marking of otolith in both larvae and juveniles.
    Ages of fish over 200 days old tended to be slightly underestimated, because the earlier in-crements arround the focus became invisible due to thickening and brownish coloration of otolith.accompanied by fish growth, and further because the occurrence of conspicuous opaque bands (checks) corresponding to the tentative cessation of increment formation increased in older fish. The error, however, did not exceed 5 days (2%) till 300 days old, and the age determination method was applicable to the ecological study of this species for almost the entire life cycle.

    Aseries experiments was performed to find the fundamental aspects of the force acting on the ground rope in towing. Two types of models; rectangular plate and cylinder were used in the experiments. The motion of the sand on the bank which was formed by the ground rope in towing was observed. The volume of sand on the bank was calculated both with the results of measurements taken and a theoritical method based on the sand cone. Theoretical values agreed with calculated values. The force acting on the ground rope was almost proportional to the volume of bank formed by the rope.

    To investigate factors affecting development of rancid off odor or flavor in cooked fish meat during storage in a refrigerator at 5°C, rates of lipid oxidation of the cooked meats of various species of fish were compared to each other by using TBA test and then influences of water-extract of fish meat on the lipid oxidation were examined. Rates of increases in TBA values of cooked white muscles of fish examined here during storage were as follows; saury>mackerel>sardine>horse mackerel>flying fish>sablefish>halibut>dusky sole. White meat species such as sable-fish, halibut and dusky sole were essentially not susceptible to rancidity. The susceptibility of white muscle of mackerel to rancidity was reduced remarkably by repeated extractions with water prior to cooking. Further, the dialysed water-extract of mackerel meat showed much higher activity to cause rancidity of water-extracted mackerel meat than the diffused solution.
    From these results, heme pigment present in the fish muscle was suspected to be a major catalyst responsible for the development of rancidity after cooking.

    The frozen surimi of Alaska pollack was ground with NaCl and the salted meat paste was submitted to setting. When the setting proceeded, the decrease in myosin heavy chain (MHC) and the increase in jelly strength of the paste were investigated as a function of protein content, NaCl concentration, pH, and temperature. From the time courses, the rate (v1) and extent (e1) of decreasing MHC and those (v2 and e2) of increasing jelly strength were evaluated. As the decrease in MHC was accompanied by the formation of its polymer, the decreasing ability was presumed to manifest a cross-linking ability of MHC. On the other hand the increasing ability of jelly strength showed a gel-forming ability of the source. It was thus found that there is a positive correlation between the cross-linking ability of MHC (expressed as v1•e1) and the gel-forming ability (expressed as v2•e2) of the salted meat paste. As the protein content increases in the paste, the abilities given by v1•e1 and v2•e2 were both largely enhanced. At a fixed protein content, the same abilities (V1•C2 and v2•e2) Were largest When NaCl concentration Was at 0.5 M, pH at 7.0, and temperature at 25°C.

    Restructured sardine meat was produced from the mixture of sardine mince and sodiumalginate by the aid of dialysis against CaCl2 solution. In comparison of raw and cooked products From May-sardine-round (lipid 9.8%), the jelly strength did not increase and the breaking de-formation was slightly decreased by the cooking process. These results would be due to theprotein denaturation induced during the frozen-storage of sardine at -20°C for 2 months. In comparison of raw and cooked products from November-sardine-dress (lipid 16.6%), the jelly strength did not increase and the breaking deformation was slightly decreased by the cooking process. These results would be due to high lipid content in November-sardine-dress. In com-parison of raw and cooked products from December-sardine-round (lipid 11.6%) and December-sazdine-dress (lipid 10.4%), the jelly strength increased twice or more by the cooking process. However, upon cooking, the breaking deformation clearly increased in the case of round-product, but it remained unchanged in the case of dress-product. These results suggest that the proteins were kept in native state when sardine was frozen-stored at -40°C within 1 month. The cooked product from December-sardine-round possessed the most favorable texture with a juicy taste.

    Texture of material for imitation crab leg was characterized by creep/recovery behavior, and the relationship between sensory response and rheological property was discussed psycho-rheologically. The creep/recovery measurements were carried out on compression type parallel-plate viscoelastometer. The constant stress was applied for 300s to determine the creep, and the stress was removed to determine the recovery for additional 300s. The creep data were fitted to creep equation by regression analysis, and the theoretical recovery curve was predicted from the creep response. The requisites to material for imitation crab leg were great jelly strength in plunger penetrafion test and large strain in creep measurement. Material for imitation crab leg (high grade) exhibited small Young's modulus of Hookean body, small Young's moduli of Voigt body, and small viscosities of Voigt body. It also exhibited medium (moderate) retardation times and moderately large viscosity of Newtonian body. Since the strain was extremely large in creep behavior, perfect creep recovery was not observed due to the slight destruction of network struc-ture. In sensory evaluation, the requisites to material for imitation crab leg were large sense intensity of jelly strength and high degree of softness. There was reasonable agreement between sensory evaluation result and instrumental/rheological data.

    The concentrations of polyamines in the tissues of five live fresh-water fishes were determined by gas chromatograph equipped with a flame ionization detector. Spermine (Spn), spermidine (Spd) and putrescine (Put) were distributed in the tissues in general; Spn was largest, followed by Spd, and Put was smallest in quantity. Loach Misgurnus anguillicaudatus was lacking in Spn and Put, and contained cadaverine (Cad) in the muscle and skin. Cad was also detected in the skin of eel Anguilla japonica. A large amount of Spn was determined in the spermary of carp Cyprinus carpio (103mg/100g). During the storage at 5°C, the level of Spn in the muscle of carp decreased gradually as decomposition progressed and that of Spd was almost constant. The levels of Put, Cad and tyramine increased remarkably after initial decomposition.

    The salted fish meat paste prepared from frozen surimi of Alaska pollack with 3% NaCl was submitted to setting (suwari) at 20°C. Fish gels formed during the setting for up to 6h were completely dissolved in SDS solution containing urea and mercaptoethanol. The SDS-soluble protein was applied to analyze the protein subunits composition by SDS-gel filtration technique using Bio-Gel A 50m column, followed by SDS-polyacrylamide gel electrophoresis (SDS-PAGE).
    It was shown that myosin heavy chain (denoted as HC) decreased as the setting proceeded, and a group of oligomers of myosin heavy chain (correspond to (HC)n, n=2-6) was formed at an early stage of the setting. At a later stage of the setting, polymers of myosin heavy chain (correspond to (HC) n, n>10) were found by SDS-PAGE using 1.8% acrylamide gels. The SDS-gel filtration patterns of the other myofibrillar proteins were scarcely changed.

    Attempts were made to isolate toxins from the blue-green alga Microcystis aeruginosa which has been reported to form a toxic wafer-bloom in freshwater lakes and ponds in various countries. Aunialgal strain (M228) from Lake Suwa, Nagano Prefecture, was cultured, and the cells were harvested and freeze-dried. The water extract was subjected to high performance liquid chromato-graphy (HPLC) on a DEAE-5PW column, resulting in appearance of two toxic peaks. HPLC using an ODS-80TM reversed phase column made it possible to isolate two toxins, P-1 and P-2, with yields of 100μg and 700μg/g freeze-dried cells, respectively. The specific toxicity of the main toxin, P-2, was 70μg/kg mice (i.p.).

    The effect of three polymerized phosphates [sodium pyrophosphate (PPi), sodium tripoly-phosphate (TPi), and sodium polyphosphate (PolyPi)] on biochemical properties of carp myosin B was studied in the presence of MgCl2.
    When 1 mM PPi or TPi, or 0.03% PolyPi was added to myosin B in a medium consisting of 0.5M KCl, 40mM Tris-buffer (pH 7.O) and 1mM MgCl2, a decrease in viscosity and an enhanced liberation of inorganic phosphate (Pi) occurred. The mode of inactivation of Ca-ATPase changed from a single first order to a biphasic first order process. The change, however, was fully re-covered on addition of rabbit F-actin. On the other hand, determination of the liberation rate of Pi from those polymerized phosphates by myosin triphosphatase indicated that the conversion of TPi and PolyPi to PPi proceeded gradually.
    These results suggested that a large portion of F-actin in myosin B was rapidly denatured and the myosin-binding ability was lost upon the addition of PPi and MgCl2, and that the gradual production of PPi from TPi (or PolyPi) caused slow denaturation of F-actin in myosin B.

    The quantitative analysis of guanidino-compounds in a living body was made using high performance liquid chromatography with colorimetric method.
    The operations were performed by the operation system as shown in Fig. 7. Eiuate from the separation column (HITACHI•#2619-F, 4×80mm) was mixed with the developing solution S1, and heated in 5m reaction coil A (0.5mm inner diameter) at 70°C. The mixture was further mixed with the developing solution S2, and heated to color-develop at 70° in 15m reaction coil B (0.5mm inner diameter). Then, absorbance of the reaction mixture was measured at 535nm. As the result, ten kinds of guanidine-compounds were sensitively measured.

    Distribution of guanidino-compounds in various tissues of aquatic animals were studied. The results obtained were as follows. No more than creatine, creatinine and arginine were de-tested in various tissues of fishes analysed.
    In fishes, creatine contents were high in white muscle followed by dark muscle, brain, stomach, pyloric caecum and heart. Creatinine and arginine contents in fishes, were very little.
    In prawn and swimming crab, arginine alone was detected and its content was very high in muscle followed by brain, stomach, intestine, heart and liver.
    In cattle fish and pacific octopus, arginine content was high in mantle followed by heart, arm, brain, stomach, intestinal caecum, intestine, liver, pancreas, ovary and testis. And octopine was detected in its mantle and arm.

    A study was made of the induction of phenol-sulfate conjugating activity by exposure to some phenolic compounds and also the duration of its activity induced by pentachlorophenol (PCP) in the mid-gut gland of short-necked clam Ruditapes phlippinarum.
    In a preliminary experiment comparing the inductivity of phenol-sulfate conjugating activity in short-necked clam among PCP, resorcinol, p-cresol, p-chlorophenol, p-nitrophenol and phenol; PCP was the most effective inducer, resulting in a 2.4-fold activity in the control clam during the 3-day exposure period to 0.1ppm PCP. Therefore, a subsequent experiment was performed to elucidate the effect of long-term exposure to sublethal PCP (0.1ppm) for 5 weeks on the phenol-sulfate conjugating activity in clams, feeding on a unicellular green alga Dunaliella sp. by dipping the clams in its growing sea water-medium (ca.4×104 cells/ml) for 2-3h every other day, when the test PCP-water was replaced by a newly prepared one. The conjugating activity in the clam increased with the time of exposure to PCP and reached 42nmo1/min/g-tissue at 5-week period, corresponding to ca. 7-fold with that in the control. When the clams which were exposed to 0.1ppm PCP for 2 weeks and then transferred to PCP-free sea water, its induced activity of 24nmol/min/g-tissue decreased gradually, but it was still at a level of 1.5-fold with that in the control even after 3 weeks, although the biological half-life of PCP accumulated in the clam was only ca. 1h.

    A screening was undertaken to acquire bacteria with a high rate of β-1, 3-xylanase productivity. Such a strain was obtained from the bottom mud of the coastal sea in Fukuoka Prefecture. The organism needed β-1, 3-xylan as an inducer to produce β-1, 3-xylanase, and released a large amount of the enzyme into the culture fluid, when incubated with shaking at 25°C for 4 to 6 days in the following liquid medium; 0.1% peptone, 0.1% yeast extract, 3% NaCl, 0.2% K2HPO4, 0.05% KH2PO4, 0.05% MgSO4•7H2O, and 0.3% β-1, 3-xylan, pH 7.8. The enzyme exhibited a high activity at a range of pH 6.0 to 7.5. The enzyme seems to be an endo-type β-1, 3-xylanase which acts on the substrate to give various sizes of oligosaccharides and may be an essential com-ponent for the preparation of protoplasts from some groups of seaweed.
    According to the criteria of the Bergey's Manual of Systematic Bacteriology, Vol.1, the bacterium was assigned to the genus Vibrio. There was no listed species name for this organism.

    Aprotein fraction extracted from squid (squid protein fraction=SPF) was tested in Penaeus japonicus juveniles. SPF was substitued for purified fish protein in a mixed diet at a level ranging from 1.5 to 16% all diets being isolipidic and isoproteic. No change in food intake was observed, but weight gain and number of molts increased with SPF level and reached a plateau over 6% SPF. Food conversion was also improved by the addition of SPF. DNA content of whole shrimp did not change, but RNA and RNA to DNA ratio varied in a manner similar to weight gain suggesting that growth enhancement was due to hypertrophy of cells, not to hyperplasy. No clear effect of SPF on amylase or total proteases activities was found.
