    The chief of factors affecting body length-variations of a sand eel Ammodytes personatus were studied in Ise Bay. Mean body size and growth rate of first year sand eel in fourteen year-classes varied considerably among the year-classes. The relative variation in individual fish size, coeffi-cient of variation (CV), in each year-class was inversely related to population density and was thus density-dependent. The causes of density-dependent fluctuation of the variation were dis-cussed.

    Symplectoeuthis oualaniensis and Eucleoteuthis luminosa were caught with hand-lines under fish attracting light, and a study was made to investigate the depth of catching, water temperature and the relationship between the catch and the color of Jigs. The following results were obtained.

    1. At each fishing area, a large number of S. oualaniensis was caught above the thermocline (high temperature zone), their catch zone was shallower than that for E. luminosa. On the other hand, E. luminosa appeared to be caught in a large number in the deeper zone, within the ther-mocline (low temperature zone). This fact seems to indicate that rather than the brightness in water, the vertical distribution of the water temperature, is greatly related to the difference of the swimming zone (catch zone) of the two species.

    2. The chi-squares test was applied to the number of squids by jigs of various colors (red, white and green). The results supported a hypothesis that the catch of squids is independent of the color of a jig. The results are thought to indicate that squids do not have such a good sense of color.

    A survey cruise was conducted in the Western Pacific between August 26 and October 2, 1986 to determine the horizontal distribution of Anguilla japonica leptocephali.
    A total of 21 A. japonica leptocephali were collected mainly by 3m IKMT with 0.5mm mesh at night. During grid survey in the former leg, standardized IKMT oblique tows (130 min towing from the surface to 500m deep) showed that only 3 of the 35 stations were positive for A. japonica leptocephali: 2 specimens at Station 7 (lat. 18°56' N, long. 129deg;34' E), 3 specimens at Station 10 (lat. 15deg;56' N, long. 129deg;37' E) and 1 specimen at Station 31 (lat. 22deg;02' N, long. 122deg;25' E). Most of leptocephali (18 specimens) occurred in the more southeastern area (lat. 15deg;56' -19deg;03' N and long. 129deg;11' -37' E) than those reported in the previous studies. The direction of water flow in this area was mainly southwest.
    The size of specimens was roughly uniform and the mean (SD) was 43.1mm (2.8), which was ca. 10mm smaller than that of specimens ever collected. The minimum record of this species (33.9mm TL) occurred at Station 10.

    A total of 27 specimens of the anguillid leptocephali were collected by the R. V. Hakuho Maru, Ocean Research Institute, University of Tokyo, in the waters south of Okinawa and east of Taiwan and Luzon in September 1986. These leptocephali were identified as 21 Anguillia japonica Temminck et SchlegeI, one A. bicolor pacifica Schmidt, and five A. marmorata Quoy et Gaimard and/or A. celebesensis Kaup based on the segmental characters. The present A. japonica leptocephali were 33.9-47.0 (mean 43.1)mm TL, and were smaller than those of the previous collections. The location and month of the capture extended spatially and temporally the distribution of the Japanese eel leptocephali in the sea. The smallest specimen (33.9mm TL) in the present A. japonica leptocephali, the youngest larva hitherto known from the field, was collected in the waters east of Luzon. All the leptocephali of other Anguilla species were found in the waters south of 17°N except one specimen.

    In order to determine the most efficient sampling method of Anguilla japonica leptocephali, the diel change of vertical distribution of leptocephali and sampling efficiency of gears were examined in a fixed water mass marked by drifting buoy with drag in the Western Pacific in September 1986.
    Forty-min horizontal tows by 3m IKMT were conducted by night and day at 7 towing depth layers between 10 and 400m deep. Number of leptocephali taken by night was 2.6 times larger (60 fish) than those sampled during daytime (23 fish). Most of leptocephali (85 %) of night sample were taken at shallow layers, 10, 20, 50 and 80 m deep, while no leptocephali occurred at layers shallower than 80 m by day. All of 10 leptocephali of A. japonica were also taken at shallow layers between 10 and 50 m deep by night. Upper limit of daytime sample and the layers of maximum catch by night roughly coincided with a sharp thermocline observed at 70-80m deep. A. japonica leptocephali seem to be obtained most efficiently by a horizontal night tow at layer just up to a thermocline.
    Comparison of sampling efficiency between IKMT (8.7m2 mouth opening, 0.5mm mesh) and Hexagon Net (10.4m2 mouth, 1-8mm mesh) showed that Hexagon Net caused a bias in sampling leptocephali smaller than 25mm TL, while IKMT collected fewer leptocephali exceeding 40mm TL than Hexagon Net.

    The auditory sensitivity of the red sea bream, Pagrus major C. TEMMINCK et SCHLEGEL, was deter-mined by heart-beat conditioning using a sound coupled with a delayed electric shock. Sufficient care was taken in order to obtain a stable acoustic condition. Sound was projected from an air-speaker in a sound-proofed chamber in which the experimental tank was placed on an air spring.
    The audible range of the red sea bream extended from 50 to 1000Hz with a peak sensitivity of -14.2dB (0dB=1μbar) at 200Hz. On the basis of its responses to unconditioned sounds, it was suspected that the red sea bream was able to distinguish sound quality.

    Analyses of genetic relationships between parents and their diploid and triploid offsprings have made it possible to estimate rate of gene-centromere recombination at phosphogluconate dehydrogenase locus in the pacific abalone, Haliotis discus hannai. Differential survivals by genotype of thermostability variations of enzymes of animals against thermal stresses have been reported on some species under natural or experimental conditions of environments.
    The present work analyses parentage of genotype compositions at phosphoglucomutase (Pgm) thermostability locus in the normal diploid and triploid sib populations induced by genome mani-pulation, estimates rate of gene-centromere recombination at the locus, and discusses probable association between differential contribution of ootids/meiotic drive and their genotypes at Pgm thermostability locus in the pacific abalone, Haliotis discus hannai.

    Dynamics of Japanese surf clams as they come in contact with the teeth of hydraulic jet dredge, is analyzed and the optimum teeth angle is discussed. The teeth angle is consider to be a function of the weight of the shell in the sand, the drag force of the shell moving vertically, the static fric-tional coefficient between the shell and the teeth, and the horizontal component force acting on the shell by the teeth. The parameters were measured under laboratory conditions, and for the type of clams used in our experiments the teeth angle of the dredge (θ) can be simply expressed as a function of shell length (L);
    θ≤tan-1 {(0.058L2.0-0.025e0.39L)/(0.029L2.0+0.050e0.30L)}.
    The teeth angle θ for a shell of 10cm long was calculated by this formula, and the optimum teeth angle θ was found to he less than 40°C.

    Some properties of lysozyme activities of the skin mucus extract, serum and kidney extract in eel Anguilla japonica were examined. Lysozyme activity was measured by the turbidimetric method using acetone-ether dried cells of Micrococcus lysodeikticus as substrate. The optimum pH and temperature for the lysozyme activity of skin mucus extract were pH 6 and 30°C whereas those of serum and kidney extract were pH 8 and 50°C. Lysozyme activities of samples after heating in an acidic buffer (pH 4) were relatively stable but those in neutral (pH 7) or alkaline buffers (pH 9) were liable to be inactivated. The activities were inactivated by addition of chitin and bentonite, and inhibited by incubation with 1mM EDTA and iodine.

    Starch gel electrophoresis was carried out to survey the genetic variation in the haploid lavers of the Porphyra sp. claimed to be Porphyra renera. Eleven enzyme loci were resolved and scored. Eight of the eleven loci were monomorphic, while the three loci, catalase (CAT), glucosephosphate isomerase (GPI) and mannosephosphate isomerase (MPI) showed polymorphism. At the Gpi and Mpi loci, a number of lavers had electrophoretic phenotypes characteristic of heterozygotes.
    In order to compare with Porphyra yezoensis, the allelic frequencies were calculated on the assumption that the heterozygous phenotypes possessed two alleles. The genetic distance between Porphyra sp. and Porphyra yezoensis was 0.1511, indicating the differentiation of the subspecies level.
    Electrophoretic phenotypes characteristic of heterozygotes is unexpected, since Porphyra lavers are in a haploid phase. Our findings suggest that the plant is either in the diploid phase or that duplication occurs in one of the chromosome sets.

    In relation to the behavior of fish school encountered by the leader-net, Fridman's theory is as follows. The fish school of constant concentration is considered as a liquid stream of density ρ=const. Let the volume rate of the liquid in the incoming stream to the leader-net be q, the volume rates in the component of the same direction as yet which is parallel with the leader-net, and that of opposite side be q2 and q1, and the encounter angle be α, respectively, i.e.,
    q1/q2=(1-cosα)/(1+cosα).
    The author has put forth a new theory on the behavior of the fish passing through the mesh of leader-net.
    If the fish swimming speed when encountering the leader-net is ν, then the component of swimming speed in parallel with the leader-net is ν cosα, and that in right angle with the leader-net is ν sinα. The volume rate of the liquid passing through the mesh is q3, i.e.,
    q1=k1k1•sinα/2
    q2=k2•(sinα/2+cosα)
    q3=k3•sinα
    k1, k2, k3; const.
    This theory is more in conformity with the experimental values than Fridman's.

    The characteristics of digestive proteolysis and digestive proteinases in the crabs Geryon affinis and Chionoecetes japonicus were studied.
    For the two species, optimum pH of general protease activity was around 6 and optimum tem-perature was around 50°C General protease activity was stable at pH 6 for Chionoecetes japonicus and in the range of pH 5-8 for Geryon affinis, respectively. Effect of metal ions on the proteolytic activity was similar in two species. Using both synthetic and natural substrates, identification of proteinases in digestion was performed. Trypsin was found in two species but the activity was very low in Chionoecetes japonicus. Chymotrypsin was not found in both species. Activities of leucine aminopeptidase and carboxypeptidase A were found to be very low in the two species. No carboxy-peptidase B was found. Significant hydrolysis of collagen was found in both species. After gel filtration, a proteolytic activity was found in low molecular weight fractions.

    The thermal adhesion of various high polymers to Alaska pollack shio-surimi was ex-amined in order to clarify the mechanism of their gel-strengthening action. A pellet of the high polymers or their suspension was gently put on the shio-surimi and heated to adhere at 40°C for 1h or at 80°C for 40 min. The adhesive force measured by stretching the adherent high poly-mers was especially strong for the soybean protein Al and vital wheat gluten in the case of 40°C, and for the egg albumin besides these two proteins in the case of 80°C, but not strong for the agar, k-carrageenan and corn starch at both the temperatures. Considering that the last three poly-saccharides are effective gelling substance as well as the former three proteins, the contribution of the gelling substances to the muscular protein network structure seems to be not indispensable for the gel-strengthening action.

    Sarcoplasmic proteins were extracted from the ordinary muscle of twelve species of marine and four species of freshwater fish. Disc-gel electrophoresis in the absence of SDS resulted in species-specific patterns. Patterns of marine fishes widely differed from those of freshwater fishes.
    Polyacrylamide slab gel electrophoresis in the presence of SDS made it possible to categorize those fishes into three groups by patterns: marine white-fleshed, bottom fish (rich in 43K and 35K components), marine red-fleshed, pelagic fish (rich in 43K, 40K and 35K components), and freshwater fish (rich in 43K component). Red sea bream, Pacific mackerel and carp, as repre-sentatives of the three groups, were analyzed for relative amounts of 43K, 40K and 35K com-ponents in the total sarcoplasmic proteins: 12.6, 13.7 and 13.5% in red sea bream, 13.7, 18.7 and 11.6% in Pacific mackerel, and 19.5, 12.3 and 10.7% in carp, respectively.

    Attempts were made to identify the native proteins (“43K”, “40K” and “35K” components) whose subunits appeared as three bands in SDS-gel electrophoretograms of sarcoplasmic proteins from each of red sea bream, Pacific mackerel and carp ordinary muscle. Judging from gel filtra-tion behaviors on Sephadex G-150, along with subunit molecular weights and other properties, “43K”, “40K” and “35K” components were identified as creatine kinase, aldolase and glycer-aldehyde-3-phosphate dehydrogenase, respectively.
    Specific activity of creatine kinase ranged from 25.9-33.3 units/mg through the three fish species, whereas those of aldolase and glyceraldehyde-3-phosphate dehydrogenase were more species-specific, ranging from 12.4-32.3 and 28.0-39.4 units/mg, respectively. Molecular weights of isolated creatine kinase, aldolase and glyceraldehyde-3-phosphate dehydrogenase were deter-mined to be 86, 000-88, 000, 160, 000 and 136, 000-140, 000, respectively, regardless of fish species.

    Distribution and generic composition of denitrifying bacteria in the bottom sediments of Ise Bay, Kumano Nada, Nansei Shoto and the surrounding area were investigated from 1983 to 1986.
    Denitrifying bacteria were enumerated by the M.P.N. method. Their number in the surface layer (0-3cm) of the bottom sediments ranged from less than 2.0×101 to 7.9×104cells/g(wet). The average percentage of the occurrence of denitrifying bacteria to that of heterotrophic ones was about 1.64% in Ise Bay, Kumano Nada, Nansei Shotõ and the surrounding area, and about only 0.67% in the sand beach of Machiya Ura, Mie Prefecture.
    Most of the denitrifying bacteria were gram negative, asporogenous motile rods with polar, lateral or peritrichous flagella. About 64.8% of the denitrifiers required NaCl for growth and 86.9% were able to grow on the medium containing inorganic nitrogenous compounds (NO3- or NH4+) as a nitrogen source. Pseudomonas (40.1%) was the most dominant denitrifying bacterial genus. Pseudomonas, Alcaligenes and Bacillus comprised 77.8% of the denitrifying bacteria isolated from coastal and oceanic bottom sediments.

    Distribution and generic composition of lytic enzyme-producing bacteria in coastal and pelagic bottom sediments of Japan were studied.
    Bacteria capable of producing lytic enzymes were in the range of 101-104 cfu per 1g of wet bottom muds. The average percentage of the number of lytic enzyme-producing bacteria to that of heterotrophic ones was 12.1%.
    Out of 482 strains of lytic enzyme-producing bacteria isolated from bottom sediments of Ise Bay, Matoya Bay, Owase Bay, Kumano Nada, Nansei Shotõ and the surrounding area, Bacillus (87.3%) was the most dominant bacterial genus.

    The oxidative stability of sardine meal and oil mixed with the stick water browned by the Maillard reaction was compared with that of the meal and oil supplemented with synthetic anti-oxidants such as BHA and ethoxyquin. Browned stick water was prepared by the reaction be-tween added glucose and amino compounds in stick water at 80-120°C. The inhibitory effect was evaluated by measurement of oxygen absorption by fish meal or oil contained in a sealed flask and stored at 37°C in the dark. The ratio of oxygen/nitrogen in the head space of the flask was periodically determined by gas chromatography with a thermal conductivity detector. The antioxidative effect of stick water on fish oil and meal was remarkably enhanced with the progress of browning and nearly proportional to browning intensity. The oxidative stability of fish meal produced from press cake and the browned stick water with E4%1cm 0.28 at 420nm was comparable to that containing 0.01% ethoxyquin. After the Maillard reaction, the content of histidine, the most abundant free amino acid, and histamine in stick water decreased significantly. The decrease of these two imidazole compounds is preferable because these can produce gizzerosine which strongly induces Gizzard erosion in chicks.

    The effect of the ratio of mannuroic acid to gluronic acid on the solution properties of sodium alginate were examined by using two samples of which the M/G ratios were 2.7 and 0.5. The reduced viscosity could not be treated according to the Huggins' equation over a critical concentra-tion. The critical concentrations also corresponded to the critical point of the concentration dependence of the activation energy of flow, and were ca. 1.0g/dl for the M-rich sample and ca. 0.5g/dl for the G-rich sample. The results showed that the G-rich sample has a large chain ent-anglement in solution.

    The total protein obtained from sardine was succinylated and the modified protein (SP) was used to manufacture cheese analog foods.
    Emulsions stabilized by SP were curdled by lactic acid or glucono-ò-lactone (GDL), and pressed into cakes. The emulsions were stable during the acidification and pressing stages. Emulsification contributed to the whiteness and softness of the cakes. Rennin had a softening effect on the emul-sion cakes.
    Emulsion cakes were inoculated with Penicillium sp. and ripened under the conditions of RH 86%, at 15-20°C. P. caseicolum performed the most ripening of the curds, followed by P. camemberti and P. roqueforti. P. caseicolum made the cakes soft and brown because of its high protease and lipase activity, while in the case of P. camemberti and P. roqueforti, moderate ripening was observed. Nutritional value of curd protein ripened by P. camemberti was as high as sardine protein.
    It is possible that cheese analogs made from fish protein may become a novel fermented food.

    As part of the investigation on useful constituents in microalgae, we searched for biologically active compounds in the water-soluble and the fat-soluble fractions of twenty five species and strains of microalgae by means of the antimicrobial activity test and the inhibition test of develop-ment of fertilized echinoderm eggs. We found eleven and eight active species by the former and latter test, respectively, and five species to have both activities. The activity existed exclusively in the fat-soluble fractions. For example, a marine cyanophyte Oscillatoria sp.-2, a freshwater diatom Cyclotella sp., and a marine dinoflagellate Goniodoma pseudogoniaulax showed strong inhibition against the fungus Mortierella ramannianus. In particular, the fat soluble fraction of G. pseudogoniaulax inhibited the growth at a concentration of 0.1 mg/ml/. The fraction also had a strong inhibitory effect on the fertilized eggs of the sea urchin Hemicentrotus pulcherrius, stopping the first cleavage at a concentration of as low as 1 μg/ml.

    The seasonal variations with growth in the proximate compositions and mineral contents of cultured yellowtail, Seriola quinqueradiata, were examined during a 20 month period from Febru-ary, 1985 to November, 1986. The muscle of yellowtail was divided into four parts: dorsal, ventral, caudal, and dark muscles, and for each part the amounts of moisture, crude protein, crude lipid, ash, and 13 kinds of minerals were analyzed. The following results were obtained. The moisture content was higher in the caudal muscle than in the other parts. In all the muscles moisture contents tended to increase in the summer. Crude protein contents were slightly higher in one year old fish than those in two years old. In lipid contents, there was a remarkable variation according to growth, and the lowest level was found in the summer. A negative correlation was noticed (n=160, r=-0.692) between the contents of moisture and crude lipid. The seasonal variations in ash content in the dorsal, ventral, caudal muscles were relatively slight.
    Seasonal variations of Ca, Na, and K contents in the muscles were significant; values of P, Fe, K, Mg, Cu, and Mn in dark muscle were different considerably from those in other parts. Higher contents of Fe, Cu, and Mn were noticed in the dark muscle. Heavy metals such as Pb, Hg, and Cd in all the muscles tested were within the dietary safety levels. The mean value of the edible portion of all samples assayed during a 20 month period was as follows: moisture 55.3g, crude protein 23.6g, crude lipid 14.0g, ash 1.8g, Ca 3mg, P 265mg, Fe 1.1mg, Na 26mg, and K 336mg per 100g. These results indicate that several components differ in value from those listed in the standard tables of food composition in Japan.

    Methods for assaying the alternative complement pathway activity (ACH50) of rainbow trout Salmo gairdneri, ayu Plecoglossus altivelis, carp Cyprinus carpio, tilapia Tilapia nilotica, eel Anguilla japonica, yellowtail Seriola quinqueradiata, porgy Pagrus major and flounder Paralichthys olivaceus sera were investigated.
    The ACH50 values of the sera of all fish except eel could be measured by incubating each serum with rabbit red blood cells (target cells) at 20°C in the presence of 10mM EGTA and 10mM MgCl2. The pH values and incubation periods employed were: rainbow trout, pH 7.0, 1.5h; ayu, pH 7.0, 1.5h; carp, pH 7.5, 1.5h; tilapia, pH 7.0, 1.5; yellowtail, pH 7.0, 2.5h; porgy, pH 7.5, 2.0h; flounder, pH 7.5, 2.0h. The hemolytic activity of eel serum was not reduced even in the presence of 10mM EDTA, suggesting the participation of unknown factor(s) in the hemolysis. Therefore, titration of the ACH50 value of eel serum was not carried out.
    ACH50 values (unit/ml, mean±SD, n=3 to 24) of the above 7 species were as follows: rainbow trout, 456±88; ayu, 880±376; carp, 51±14; tilapia, 684±560; yellowtail, 1016±56; porgy, 328±188; flounder, 508±60. These values are 5 to 60 times higher than those reported for mammals, suggesting that the role of the alternative pathway is much more important in fish than in mammals.

    When carp myosin was denatured, its heavy chain was cleaved by calpain to give a major fragment of Mr=150, 000 (150k fragment). The same result was obtained from the denatured myosin which had been labeled at SH1 group of the heavy chain with a fluorescent probe 1, 5 IAEDANS. The following results indicated that the 150k fragment corresponds to a major part of the heavy chain of myosin rod segments: (1) the 150k fragment was insoluble at a low ionic strength; (2) it was not released by calpain digestion of heavy meromyosin; (3) it was larger in size than that of the rod prepared by α-chymotryptic digestion; (4) it did not contain the SH1 group.
    The remainder of the heavy chain including the SH1 group was further digested by calpain to lower molecular weight-fragments.
    On gel filtration at a high ionic strength where the denatured myosin aggregates and the 150k fragment were fully soluble, it was found that they were eluted together as an indistinguishable single peak, suggesting that they were strongly associated together by non-covalent forces.
    The significance of the 150k fragment in relation to changes in meat proteins during processing or storage was discussed.

    3'-Phosphoadenosine 5'-phosphosulfate (PAPS) synthesis from sulfate and ATP by the cell-free enzyme system of marine alga Porphyra yezoensis was demonstrated. PAPS formed in the reaction mixture was isolated by DEAE-Toyo Pearl column chromatography, and identified by the behavior on TLC, HPLC and UV spectra.
    PAPS synthesizing activity was inhibited by molybdate and selenate. PAPS was synthesized also from ATP and APS as substrates. These results suggest the presence of the PAPS synthesizing system involving ATP-sulfurylase and APS-kinase in the alga.
