    Using an energy components analysis of the 73 day continuous time-depth recording of a female northern elephant seal, we estimated the total daily energy requirement to be between 9.4 Mcal and 10.0 Mcal. Total prey energy consumed was estimated between 15.9 and 16.8 Mcal/day, yielding a diet of approximately 10.7 kg of mackerel or 21.0 kg of squid or hake per day. We estimate that at least 60 % of the dives must be successful to account for the mass gained during the period at sea.

    The spawning season of Japanese Spanish mackerel Scomberomorus niphonius lasts from the middle of May to the beginning of July in the Seto Inland Sea.
    The mean interval of individual spawnings ranged to 1.11-1.36 days, estimated by the ratio of females with hydrated oocyte. The relationship between batch fecundity (f) and body weight (w; g) was expressed as f =56.7w-5.71×104.
    The number of large vitellogenic oocytes mainly corresponded to tertiary yolk globule stage, compared with that of hydrated oocytes in the same ovary, led to an estimate that they were spawn-ed during approximately between four and eight consecutive days. The next large vitellogenic oocyte group seemed to be formed during the pause period of about one day.

    The morphology of the sense organs of the larvae and juveniles of the ribbonfish Trichiurus lepturus in Wakasa Bay was examined to obtain insights on changes in behavior and habitats with growth of the fish. Development of the sense organs proceeded in accord with the rest of the morphological development of the larvae and juveniles. Sense organs required for feeding were formed at the onset of exogenous feeding. The taste buds first appeared at the begining of the postlarval period and increased in number in association with the pharyngeal teeth and gill arches. During the late phase postlarval period, the lateral line canal on the head and the olfactory rescue were formed. The retinae became duplex and developed horizontal cells in three layers; this is inferred to be advantageous for a piscivorous feeding habit. The presence of well developed sense organs synchronized with the active migration of postlarvae from offshre to coastal waters.

    The sense organ morphology of wild flounder larvae caught in Wakasa Bay was examined and compared with that of reared ones, and the results were correlated with behavioral changes during growth of larvae in the sea. Mechanoreceptors developed earlier in the reared larvae, whereas the eyes and taste buds developed earlier in the wild larvae. Mechanisms involved in these differences were not ascertained in this study. The formation of the rod and twin-cone photoreceptors coincids, respectively, with the initiation of shoreward migration, and with the change from pelagic to benthic habit of the larvae. When the taste buds are formed, wild larvae exhibit selective feeding on the zooplankton Paracalanus parvus.

    From the results of experiments on foraging behavior in ten species of freshwater animals in clear water column measuring 60 cm deep and 100cm in diameter, the following information was obtained.
    Luring by prawn flesh bait began before its falling on water-surface in masu salmon Oncorhy-nchus masou, white spotted char Salvelinus leucomaenis, three spine stickleback Gasterosteus acule-atus, and goby Chaenogobius urotaenia. Hooking on watersurface northern dace Tribolodon ezoe rarely took part in addition to the above fishes. All fishes were lured to the sinking visible bait especially just before its arriving at bottom to begin with northern dace. However white spotted char or masu salmon predominatly foraged where the dominant among the other species was carp Cyprinus carpio, silver crucian Carassius auratus langsdorfii, northern dace, goby and three spine stickleback in order except lake prawn Palaemon paucidens being not a sight feeder.
    It is generalized that vulnerability to angling related with hooking order among freshwater fishes within each other's visible field are determined principally by interspecific dominance system and occasionally by quickness of foraging behavior.

    The variations in hemolymph ecdysteroids were traced during the molt cycle in immature female Kuruma prawn Penaeus japonicus. Ecdysteroids in hemolymph were separated using high performance liquid chromatography, and their levels were measured by a double antibody radioimmunoassay (RIA) method. The RIA was validated prior to assay. The competition curves for the serially diluted hemolymph extracts of the Kuruma prawn and two freshwater prawns, Macrobrachium nipponense and Macrobrachium rosenbergii, were parallel to the standard curve. The mean recovery rates for 20-hydroxyecdysone and ecdysone were 107 and 99.9%, respectively. The intraassay coefficients of variation were 8.2, 8.3 and 7.7% at 62, 48 and 19% binding, respectively. The interassay coefficient of variation was 12.0%at 46% binding. Ecdysone and 20-hydroxyecdysone showed identical cross reactivities. The minimum detectable level was 15 pg/tube. In the postmolt and intermolt stages, ecdysterioid levels in the hemolymph of P. japonicus were found at basal levels. The levels increased in the early premolt stage, and peaked at the late premolt stage. The predominant immunoreactive ecdysteroid in P. japonicus was 20-hydroxyecdysone. Small amounts of ecdysone and ponasterone A were also detected. In addition, high polarity and low polarity ecdysteroids were eluted, but could not be identified.

    Ovulation was induced in sexually mature female goldfish Carassius auratus maintained at 12°C under 15L-9D by raising the water temperature to 20°C under various photoperiods. The fish were checked every 2 or 3h, and those which had ovulated were blood-sampled to determine the timing of the GtH surge. In Experiment 1, 2 groups of fish (n=15 each) were placed under photoperiods fixed at 15L-9D. In Group A, lights on was 0400h, and in Group B, lights on was 1600h. Ovulation occurred during the dark-phase in both groups. In Experiment 2, the photoperiod was changed from 15L-9D (lights on 0400h) to continuous light (Group C; n=21) or continuous darkness (Group D; n=18) after the water temperature increase. In both groups, ovulations were observed at a time corresponding to the dark-phase of the acclimation photoperiod, however, in Group D, ovulation was slightly delayed when compared to Group C. In both Experiments, the GtH surge was observed only in ovulated fish. In Experiment 3, mature females were reared under 14L-10D (lights on 0430h) and either pinealectomized (PINX, Group E, n=19) or sham operated (Group F, n=21). The water temperature was increased one week following the operation. Fish from Group F ovualted during the dark-phase, whereas fish from Group E ovulated randomly. The GtH surge was observed in the ovulated fish from both groups.
    These results indicate that photoperiod plays an important role in determining the onset time of the preovulatory GtH surge. The pineal gland is somehow involved in the mechanism regulating this photoperiodic response.

    Immune responses in rainbow trout Oncorhynchus mykiss intraperitoneally injected with formalin killed Renibacterium salmoninarum cells (with or without Freund's complete adjuvant), were evaluated by monitoring serum agglutinating antibody and phagocytic activity of kidney leukocytes, and the challenge test. Serum antibody titers (range 8-16) were detected in the vaccinated groups 5 weeks after vaccination. The phagocytic activities in the kidney cells of vaccinated groups increased after 15 days and 25 days. However, experimental intraperitoneal challenge (107 or 106 cells/fish) of immunized groups showed no protective effects.

    In vitro efficacies of chemotherapeutics were tested against 20 strains of Mycobacterium sp., isolated from cultured yellowtail Seriola quinqueradiata in Kochi, Ehime and Wakayama prefectures in 1985-1986. Nine antibiotic agents, ampicillin (ABPC); streptomycin (SM); josamycin (JM); erythromycin (EM); lincomycin (LCM); rifampicin (RFP); oxytetracycline (OTC); chloramphenicol (CP), and one chemical synthetic agent, isoniazid (INH), were used in this study. The efficacies of chemotherapeutics to Mycobacterium sp. were determined as minimum inhibitory concentrations (MIC) to 20 strains, minimum bactericidal concentrations (MBC) to 10 strains and the ratios of MBCs to MICs.
    The strains showed high susceptibilities to RFP, SM and EM: 0.025-0.78μ g/ml (MIC), 0.025-0.78μ g/ml (MBC). The susceptibilities of isolates to ABPC were 1.56-3.13μ g/ml (MIC and MBC). The isolates showed relatively low susceptibilities to OTC, JM, CP, INH and LCM: 3.13-25μ g/ml (MIC), 3.13-50μ g/ml (MBC). SPM showed the lowest activity against the isolates: 25-100μ g/ml (MIC), 50->100μ g/ml (MBC). The range of MBC/MIC ratios were 1 for ABPC and SM; 1 ?? 2 for JM and LCM; 1 ?? 4 for EM, RFP and OTC; and 2 ?? 4 for INH.

    Computer simulations using compartment models were performed to examine the characteristics of stationary nets. Simulations showed that the front funnel affected the fish catching speed and the rear funnel primarily affected the number of fish retained in the front bag net.
    It was found that the fish density of a three compartment model and a two compartment model could increase by 2.64 times and 1.74 times the standard density of fish, respectively.
    In addition to these facts, Synergism was found between the functions of funnels (front and rear).

    Collecting low-fat meat from whole fish by use of cryo-separation which consists of cryo-shattering followed by cryo-sieving was studied in an attempt to pave the way for the acceptance of sardines and mackerels for human food. The segregated portions of mackerel, i.e., the light muscle, dark muscle, and viscera were shattered one by one at selected low temperatures. The particle size distribution of shattered tissues deeply depended on the temperature of shattering. At -60°C, the light muscle, the leanest among the portions, was ground into finer particles than that of the other portions. The relationship between the particle size and the fat content of particles, obtained through cryo-shattering of whole mackerels and sardines, showed various patterns depending upon shattering temperatures. Fortunately, however, a simple relationship in which the fat content of particles decreased with the reduction in particle size was obtained at -60°C for mackerel and at -80°C for sardine. The maximum recoveries of low-fat meat (light muscle equivalent) were 95% for mackerel and 89% for sardine.

    From the view point of effective utilization of fisheries by-products, a method for manufacturing bonito spermary extracts by autolysis was examined. The optimum lysing temperature was found at 60°C, pH 7.5 with two parts of water. The extract obtained from 20h autolysates showed favorable characteristic flavor. It was rich in free amino acid and 5'-nucleotide. Total amount of Arg, His, Glu, Ala, and Tau accounted for more than 70% of that of total free amino acid. Thus the products was expected to be used as characteristic and unique seasoning extract.

    Fresh round mackerel was frozen at -20°C, subsequently stored at the same temperature for 100 days, thawed in tap water, and stored again at -1°C for 30 days. During storage, VBN content, K value, TBA value, free amino acid composition, lipid class composition, and fatty acid composition of lipids in the white muscle of fish were determined at an appropriate interval and compared with those of the unfrozen fish which was stored under the same conditions, i.e. at -1°C. The results obtained showed that the freeze-thawing process slightly accelerated deterioration in freshness of fish during storage at -1°C. Oxidation and enzymatic degradations of lipids during storage were also accelerated by this processing during the subsequent storage. Contents of most free amino acid increased during storage, while that of histidine decreased. The rate of increase in the content of each of the free amino acids was higher in the freeze-thawed sample than in the unfrozen sample. These deterioration in the quality of freeze-thawed fish seems to be due to disruption of membranes and other tissue structures of the fish flesh caused by freeze-thawing process.

    This study was conducted to determine the effect of supplementation of Zn to fish meal diets on growth of Japanese eel. The feeding experiments were carried out using white fish meal diets supplemented with 0, 50, 100, and 200μ g/g of Zn as Zinc sulfate (ZnSO4•anhydrous). The results revealed that the fish on diet annexed with 50 and 100μ g/g of Zn were observed to have the high growth among these groups.
    Eel fed a diet without supplementation of Zn caused a high value of condition factor (19, 0) and the content of Zn in the bone was also significantly lower. Moreover, the analytical results revealed that the bones from cultured eels having abnormal vertebrae fractional group had a relatively low level of Zn.

    Myosin subfragment 1(S1) was prepared from the ordinary muscle of tilapia Oreochromis niloticus by α-chymotryptic digestion in the presence of EDTA. S1 was further separated by anion-exchange column chromatography into two isozymes, based on associating alkali light chains A1 and A2, S1(A1) and S1(A2). The two isozymes exhibited a similar pH-dependency of Ca-or EDTA-ATPase activity, with a minimum at around pH7 for the former activity and a maximum at around pH9 for the both. As a whole, however, both activities of S1(A2) were somewhat higher than those of S1(A1). In actin-activated Mg-ATPase activity, on the other hand, S1(A2) showed a lower apparent dissociation constant for actin and higher maximum velocity than S1(A1), in contrast to the counterpart of rabbit. All these results suggest that alkali light chains affect S1 ATPases both in the presence and absence of actin.

    Myosins were prepared from the ordinary muscle of 13 fish species such as skipjack, tuna, mackerel, and electrophoresed on SDS-polyacrylamide gradient gels. Resulting light chain regions were transferred to a GVHP membrane, and examined by immunoblotting technique for crossreactivity with the following anti-myosin light chain rabbit antisera: anti-skipjack A1, A2 and DTNB light chains, anti-round herring A1 and anti-Alaska pollack A1 antisera.
    Different blotting patterns were obtained depending on antiserum species used. A1 and DTNB light chains generally showed a strong cross-reactivity, while A2 light chain did a rather weak reactivity.
    It was suggested from these results that myosin light chains have primary to higher-order structures in common throughout the three types of light chain and the fish species examined.

    A new type fish meat for food stuff, tentatively named highly nutritional fish meat (NHFM), was processed from sardines by adopting a high-speed centrifugation of ground meat suspended in a small volume of alkaline solution. The gel-forming ability of HNFM was compared with that of ordinary surimi made from the same lot. HNFM was ground with 3% NaCl and submitted to the suwari process at 30°C for 90 min, then the material was heated at 90°C for 30min. Breaking strength and breaking strain of kamaboko thus obtained were measured with a rheometer; the whiteness was evaluated with a chromatic meter.
    The gelling properties of kamaboko formed from HNFM were superior to those from the ordinary surimi, i.e. breaking strength of the former ranging from 800 to 1, 000g was as much as 30% greater than that of the later, breaking strain was 12-13mm, and the whiteness was as high as 20%. The gel-forming ability of HNFM tended to reduced with an increase of its lipid content.

    Metabolites of five sulfa drugs in the bile and urine were identified after oral administration to rainbow trout of sulfamerazine, sulfisoxazole, sulfadimethoxine, sulfamonomethoxine, and sulfisozole.
    Bile samples were withdrawn at 48h after administration while urine samples were collected continuously up to 72h. Separation of metabolites in the sample fluids were carried out by two-dimensional thin-layer chromatography on Silica Gel 60 F254 HPTLC plate. Reactions of Ehrlich, Tsuda, and naphthoresorcinol were then observed.
    Unchanged forms, N4-acetylated forms, and glucuronides of the all drugs examined were detected. In the bile samples after administration of sulfisoxazole, two kinds of glucuronide were detected.
    It is suggested that the sulfa drugs are mainly metabolized to N4-acetylated forms and glucu-ronides in rainbow trout.

    Poly phosphates were widly used in surimi-based products in order to improve the quality. We analyzed the ortho-, poly-, and total phosphorus contents in the 28 kinds of surimi-based products collected in the commercial market in Tokyo.
    Ortho- and poly- phosphate were extracted with water from each sample and were determined separately by the modified method of Fisk-Subbarow.
    The total phosphorus content in surimi-based products was determined after the sample was burnt at 550°C.
    The results obtained are as follows: The average contents of ortho-, poly-, and total phosphorus in surimi-based products were 38.0, 16.0 and 96.5mg/100g, respectively. The values obtained were equivalent to 120 and 63.3mg/ 100g wet weight ortho phosphate and sodium triphosphate, respectively. The contents of these phosphates were quite different in each product analyzed.

    The degradation of heavy oil by two bacterial isolates from the sea water in oil-polluted Bisan Seto was investigated. Desulfurized “Class C” fule oil (the spilled fuel oil in “Mizushima Oil Spill”) before and after the degradation by coryneform Pl-1 and Moraxella Obl-1 was fractionated using silical gel columns. After 14 days at 25°C of incubation, the saturated fraction of the de-sulfurized fuel oil was more preferentially utilized than the aromatic fraction by both bacteria. When each fraction of the heavy oil was supplied as a sole carbon source, both bactria grew well on the saturated fraction but no growth was observed on the aromatic fraction. The results suggested that the saturated fraction of heavy oil was more suscetible to degradation than the aromatic fraction by these bacteria.

    Interstitial waters of marine sediments were separated into the two portions. A large interstitial water was extracted from sediment core samples by a centrifugation at 42×g for 30min, and then a small interstitial water was extracted at 1, 500×g from the residual sediment samples. The content of the large interstitial water was high in sandy sediment, while the small interstitial water was in silty and clayey sediment. Concentrations of polymeric carbohydrates were higher in the small interstitial water than in the large interstitial water, although no difference was found in monomeric carbohydrate. It was suggested that monomeric carbohydrates were uniformly distributed in marine sediments by diffusion, while polymeric carbohydrates were not.

    Cathepsin B (EC 3.4.22.1) was purified from the ordinary muscle of common mackerel by ammonium sulfate fractionation and successive chromatographies on Sephadex G-75, DEAE-Sepharose, CM-Sepharose, Q-Sepharose, and Sephacryl S-100 HR.
    The purified enzyme showed a single protein band on polyacrylamide gel electrophoresis, and its molecular weight was estimated to be 23, 000 by SDS-polyacrylamide gel electrophoresis and gel filtration on Sephacryl S-200 HR. The optimal pH of the enzyme for the hydrolysis of Z-Arg-Arg-NMec was 5.5. The enzyme was activated by sulfhydryl compounds, such as 2-mercapto-ethanol, cysteine, dithiothreitol, and glutathione. Among the compounds, cysteine was the most effective. The enzyme was moderately inhibited by pCMB and NEM, and strongly inhibited by TLCK, TPCK, antipain, leupeptin, E-64, Cu2+, and He2+. The enzyme hydrolyzed Z-Phe-Arg-NMec and Bz-Arg-NNap, but not Arg-NMec or Leu-NNap. The ratio of hydrolyzing activity against Z-Phe-Arg-NMec and Z-Arg-Arg-NMec was about 1:0.7.

    The fatty acid compositions of total lipids from various tissues of chum salmon caught on the coast and upstream at the spawning site have been investigated in detail with open-tubular gasliquid chromatography. At least 100 components were detected by GLC, of which 53 components were identified. Furan fatty acids could be detected in other tissues except for the flesh.
    In the skin, flesh and gonad, lipid contents and fatty acid contents decreased during the migration from seawater to freshwater. The distributions of positional isomers within 20:1 and 22:1 changed in this period. Although lipid contents and fatty acid contents in the liver and viscera almost maintained at constant levels, the fatty acid compositions changed during the migration. Especially, the contents of C20 and C22 monounsaturated fatty acids increased in this period. There were sex differences of fatty acid compositions in other tissues except for the liver of chum salmon caught on the upstream.

    We surveyed tetrodotoxin (TTX) production on 15 bactria strains isolated from the intestine of a puffer fish caught at Hakatajima, Ehime Prefecture, where the puffer fish is highly toxic. The mouse bioassay revealed that all culture broths were none toxic or very weakly toxic with the exception of one strain (code-named SA-4). The SA-4 strain was confirmed for its toxicity by the mouse bioassay, and mice died after showing the characteristic symptoms for TTX intoxication. The amount of toxin was 15 MU in 250ml of culture broth. After bioassays, all the remained solutions were purified through a Toyopearl HW-40 column, an Amberlite CG-50 column (NH+ form), and a charcoal column. Chromatography of purified toxins demonstrated that 2 strains in addition to SA-4 strain contained TTX and its derivatives in their broths. The cells of SA-4 strain were proved to be lethal to mice, and the existence of TTX was confirmed. The purified toxins and authentic TTX were hydrolyzed to 2-amino-6-hydroxyl-methyl-8-hydroxyquinazoline (C9-base), and converted into its TMS-C9-bases to be analyzed by a GC-MS analyzer. Mass fragment ions m/z 407 and 392, which were specific for TMS-C9-base, appeared in the same retention time. The results from detail examinations done on SA-4 strain led us to the conclusion that SA-4 strain was Schewanella putrefaciens.
