    The relation between the individual life history characteristics and the population growth in a rotifer Brachionus plicatilis is discussed, The population growth rates (r) in three different strains of the rotifer calculated theoretically from observed individual life history characteristics, embryonic developmental time, duration of pre-reproductive, reproductive and post-reproductive phase, and number of offspring, is presented. The observed characteristics varied between different strains. Due to these differences, the calculated population growth rate between 15 days and 20 days after the inoculation of one freshly laid egg varied from 0.30 to 0.46. The major factors which in-fluence the population growth rate seem to be spawning interval. Increasing the number of off-spring from one individals and the number of reproductive individuals would not bring about the desired increase in the population growth rate as well as the shortening of developmental time and spawning interval.

    In Uranouchi Bay, the external water density becomes higher than that of internal water in the summer stratification season, and the external saline water intrudes into the bottom of the bay at the spring tide. Much heat and oxygen is also transported into the bottom layer along with the water intrusion.
    Although the strong anoxic water is formed from the beginning of June in the bottom of Uranouchi bay, anoxic water masses on the bottom disappeared for a while after the intrusion, and the bottom water temperature suddenly rose about 2 to 3°C in a few days, but anoxic water was formed again at the next neap tide due to the high DO consumption and the upwelling of bottom anoxic water. These conditions killed a large number of cultured yellowtail fish, as occurred at the beginning of September 1988.

    Rotifer resting eggs can potentially be preserved in a manner similar to Artemia cysts and used as live feed in larval rearing. The potential for producing resting eggs from the most desirable rotifer stocks, the L-type (Japanese stock) and the S-type (Hawaiian stock), at different tempera-tures was investigated. Rotifers from these stocks were reared in 1000l tanks and fed Tetraselmis tetrathele. Salinities were controlled at optimum levels for resting egg production (8 ppt and 32 ppt for L- and S-type, respectively). At culture temperatures ranging from 23.1 to 30.6°C, the L-type rotifers produced more resting eggs at the lowest temperature (23.1°C) and the S-type pro-duced more at a higher temperature (28.2 and 30.6°C). The relationship of resting egg produc-tion to temperature is expressed by a logistic curve. The highest total number of resting eggs was 17.8 million in 8 days at 23°C for L-type rotifers and 101.5 million in 10 days at 28°C for S-type rotifers. The maximum number of resting eggs produced was 3, 400 and 14, 000 per 10, 000 L-type and S-type rotifers respectively. The maximum number of resting eggs produced per 1×108 cells of T. tetrathele was 2, 270 and 11, 100 for the L- and S-types respectively.

    Oxygen consumption rates of the young oval squid Sepioteuthis lessoniana (0.033-27.54g BW) reared from eggs and those captured from wild population were investigated on an individual basis by both water bottle and flow-through methods. Oxygen consumption rate (R in ml oxygen/ind./h) increased linearly with increase in BW (W in g) expressed as R=aWb. For squid smaller than about 2g BW, there was not a significant difference in the values of the exponent b and the coef-ficient a between R's measured respectively by water bottle and flow-through methods. The R was not significantly different from each other between squid reared from eggs and captured from the sea. The exponent b and coefficient a for young squids ranged from 0.741 to 0.944 and from 0.697 to 0.966, respectively. There was a tendency for the exponent to change with growth, such as b for the squids less than 0.5g BW being 0.944, while for the squids over 0.5g was 0.813, respec-tively. This suggests that R in very early life stage tends to increase more rapidly with growth than in older stages.

    Triploid red sea bream were induced by cold shocking the fertilized eggs. The gonadal develop-ment of 2- and 3-year-old triploid fish was studied histologically during the spawning season to clarify the sterility. All the 2- and 3-year-old triploid fish were males except for two specimens having bisexual gonads. The GSI value of some triploids was slightly increased, but histological appearance of the testes indicated that abnormal spermatogenesis was taking place. Only a few 3-year-old triploid fish produced milt when stripped. There were some spermatozoa with two heads and two or three flagella, and the density of these cells in the stripped fluid was very low. Based on these results, it was concluded that these spermatozoa would be not functional cells.

    Age at the first capture (xc) and the prohibition period of fishing (P) as important resource management measures were studied for abalone Haliotis (Nordotis) discus hannai fishery at the Kisakata fishing ground off the coast of Akita Prefecture. Abalone fishery has a long prohibition period and a knife edge selectivity, so a periodic pulse fishery could be assumed. Abalone was assumed to have a Ricker type or Beverton-Holt type reproduction curve. The biomass yield efficiency of parent stock and the recruitment efficiency that indicates its con-tribution to population were taken as objective functions. The sum of biomass yield efficiency and recruitment efficiency was calculated, and the optimum xc and P were investigated. It was found that the current xc=5.6 yr, and P=1 yr. were not the optima. On the other hand, because abalone is a slow-growing and long life species, time-preference and discount rate are seen to have significant effects in its fishery management. Dynamic optimization method in the Beverton-Holt model for a single cohort and Faustmann formula for multiple cohorts were used to determine the optimum xc and P respectively. Considering the results, fishery situation and market value of abalone, it was concluded that the xc=current xc (5.6 yr.), but P=2 yr. are optimum measures for resource management at Kisakata.

    The swimming behavior of the loggerhead turtles Caretta caretta was studied by ultrasonic telemetry. Three adult female loggerhead turtles, captured as they crawled onto Moto Beach. Kochi, Japan, were equipped with 50KHz ultrasonic depth transmitter and released. The swim-ming speed of the turtles was faster at night than in the day. The vertical movements of the turtles were irregular at night and in the morning, but the turtles usually swam near the sea surface in the afternoon. The movements of the turtles could be divided into six swimming patterns.

    Water temperature and the number of Gymnodinium nagasakiense cells in Gokasho Bay were monitored for six years. The times at which cell densities of G. nagasakiense reached 1cell/ml (early growth phase) and 1×103cells/ml (red tide phase) were dependent on the mean water tem-perature in the winter (December-March). The correlation coefficient between the Julian days on which cell densities first reached 1cell/ml and the mean winter water temperature was -0.98, and that between the Julian days on which cell densities first reached 1×103cells/ml and the mean winter water temperature was -0.91. These results suggest that the timing of the early growth and red tide phases of G. nagasakiense is predictable by monitoring the winter water temperature.

    Distribution of bigeye tuna Thunnus obesus in the Indian Ocean was discussed by using the catch-and-effort data of Japanese tuna longline fishery from 1967 to 1985. According to the average numbers of catches in individual 5-degree squares in the Indian Ocean, the catch distribution was recognized in a wide area between Lat. 25°N and 40°S. The higher catch tended to be observed in the western and eastern tropical regions and in the southern higher latitude regions. The areas with low catches were observed in the mid-latitude area between the tropical region and the north high latitude region, the Arabian Sea, north of Bengal Bay, and in the high latitude areas extending from east to west between Lat. 35°S and 50°S. These results were similar to the catch distribution patterns of bigeye tuna caught by longline fishery obtained in the Pacific and Atlantic Oceans, with some minor differences in the longitudinal direction.

    The promoter elements of the-100 region, CCAAT box, and TATA box were found in the 5'-flanking region offish globin genes. The 5'-flanking and 5'-untranslated regions of the yellowtail Seriola quinqueradiata a-like globin gene were recombined with the chloramphenicol acetyltrans-ferase (CAT) gene to determine whether or not these sequences possess the function of a promoter. The recombinant plasmids were introduced into fish cell lines (GF, TO-2, and GCT), and signifi-cant CAT activity was detected. In the GF cell line, CAT activity was enhanced. This result suggests that the sequences from the fish globin gene possess promoter activity and play an impor-tant role in the regulation of gene expression in cultured fish cells. SV40 early promoter activity was also detected in the fish cell lines.

    The data base of digital images of good catch conditions is not sufficient, but that of poor catches is indispensable to apply the vertical distribution of water temperature to fishing information system in order to decide the position of squid jigging in the Japan Sea. The 63 stations yielding less than 200 squids in more than 4 hours consecutive swings were sorted out, as the examples of poor catch conditions among 416 jigging records from 1982 to 1988. In this report, the data base of digital images of poor catches was examined by the group average method of cluster analysis.
    As for the results, the digital images of poor catches having mutually high similarity were pigeonholed into 15 small groups (=clusters), although there remained 17 independent images, and they were then divided into 2 large groups, which were in relation to the area division in the Japan Sea. Some small clusters consisted of the digital images within the same year, their component images were from one of the two areas and within 30 calendar days. Other clusters consisted of those in the different years, but the individual images in each of them were also from the same area and within 30 calendar days. This tendency is helpful in quasi-real time application of the data base of the fishing information system on board.
    On the basis of the results, a general flow chart for fishing information system was proposed.

    The effect of the kind of surimi, NaC1 content, and pH on meat adhesion to casing of retort-sterilized fish sausage was investigated. A poly vinylidenechloride-vinylchloride co-polymer tube was used as casing. The degree of meat adhesion was expressed by adhesive strength and the amount of meat which adhered to the casing.
    The results are as follows;
    1) The degree of meat adhesion increased wth the increase in NaCl which was added between 0.5% and 4%, while the jelly strength of the sausage reached a maximum at 3% NaC1 and was slightly low at 4% NaC1.
    2) When the pH of the sausage was lowered by the addition of glucono delta lactone (GDL), both the degree of meat adhesion and the jelly strength of the sausage decreased with the lowering of pH.
    3) The sausage which had a stronger jelly strength gave stronger meat adhesion strength.

    The active compound was water-soluble and had a molecular weight of more than 10, 000 Da.
    The organism producing the active compound was similar to the genus Moraxella.

    In order to show the usefulness of Cypridina luciferin analogues, 2-methyl-6-phenyl- and 2-methyl-6-(p-methoxyphenyl)-3, 7-dihydroimidazo [1, 2-α] pyrazin-3-ones for the specific detection of active oxygen species, chemiluminescence rates, and intensities, including their quantumn efficiencies were measured and determined by using various oxidants in aqueous conditions. The oxidant-specificity of their chemiluminescence to both singlet molecular oxygen and the superoxide anion radical was shown experimentally for the first time.

    The effects of tertiary sulfonium compounds such as dimethyl-β-propiothetin (DMPT), dimethylacetothetin (DMT), and vitamin U and quaternary ammonium compounds such as glycine betaine and choline on the growth and molt of striped prawn were examined. The results indicated that 0.1mM solution of DMPT most highly stimulates the growth and molt of prawns among various concentrations of DMPT. Though to a lesser extent, DMT solution at 0.1mM also ac-celerated the growth and molt of prawns. However, 0.1mM solutions of vitamin U, glycine betaine, and choline, especially the latter two remarkably inhibited the growth and molt of prawns. Survival rates of prawns in the DMPT, DMT, vitamin U, and control solutions showed similar tendencies of gradual decrease, whereas the glycine betaine and choline solutions brought about rapid death of prawns at the later stages.

    This study was undertaken to find the salt tolerant histamine-forming bacteria in commercial rice-bran pickles of sardine. Eight isolates of histamine-forming bacteria were found in the rice-bran pickles of sardine and shown to be salt tolerant bacteria and mesophiles. Five out of the 8 isolates were nonhalophiles. Especially, 2 out of these 5 isolates produced large amounts of histamine. All isolates were cocci, Gram positive, and non-motile bacteria. They were identified as Staphylococcus biologically. A remarkable histamine producer, strain No. 7 was mixed into a histidine broth added with 10% and 12% NaCl. The histamine level reached 100mg/100ml after 3 days at 37°C. Strain No. 7 seemed to be related to the accumulation of histamine in commercial rice-bran pickles of sardine.

    The alga causing a fresh water red tide in the Ananai-gawa Dam-controlled Lake was identified to be Peridinium bipes fo. occulutatum. The effects of light intensity, temperature, pH, and nitrogen on the growth of this alga was examined.
    The growth yield was greatest at a light intensity of 10, 000lx, a temperature of 20°C and a pH of 7.3-8.0. The growth rate was highest at a light intensity of 10, 000 lx and was not affected so much at temperatures from 10 to 25°C or a pH range of 6.0-8.7 as tested. The alga can use NO2--, NO3--, NH4+-, and urea-N under their low concentrations as a sole source of nitrogen and cannot use three amino acids (glycine, alanine, and glutamic acid) effectively. The half-saturation constant for nitrogen was 50-65μgN/l and specific growth rate was saturated at 0.5-1.3mgN/l, but a higher concentration of nitrogen (5mgN/l) was needed for reaching maximum cell yield. The minimum cell quota of nitrogen was calculated to be 0.32ng N/cell. Under the saturated condition of nitrogen, the maximum specific growth rate was 0.17-0.21 d-1.
    Among freshwater phytoplankton this alga was found to have a rather higher Ks value of nitrogen for growth and to have a smaller growth rate.

    Ayu Plecoglossus altivelis of 12cm in body length were raised in concrete tanks at four different water velocities (4cm/s, 25cm/s, 35cm/s, and 45cm/s) for 60 days. Rearing of the fish in water velocities 35 and 45cm/s exerted favorable growth and feed efficiency. Rearing in current water, however, failed to depress lipid content of muscle, liver, and intraperitoneal fat body when com-pared to that in the standing water (4cm/s).
    Following the feeding experiment, the fish were starved for 13 days in indoor tanks. The body weight loss was suppressed in the fish reared in water velocities of 35 and 45cm/s. Rearing in high water velocity (<45cm/s) could improve lipid metablolism, especially in activation of utilization of lipid reserves for energy requirements.
    In ayu culture, water velocity in the rearing tank should be controlled to 45cm/s or more.

    In order to elucidate a mechanism responsible for the development of the odor characteristic of cooked squid flesh, squid mantle flesh was extracted with various solvents and the resulting extracts were evaluated organoleptically after cooking. As a result, 80% aqueous methanol extract was found to produce a cooked squid-like odor. The extract was fractionated by means of ion exch-ange column chromatography and each of the fractions obtained was subjected to cooking, followed by organoleptic evaluation. However, any fraction did not produce a cooked squid-like odor. On the basis of the composition of 80% aqueous methanol extract, an artificial extract was prepared with reagents. The artificial extract thus obtained closely resembled 80% aqueous methanol extract in respect of the cooked odor. To elucidate the role of individual components, the artificial ex-tracts from which certain components alone or as group were omitted were subjected to organoleptic evaluation after cooking. All of the neutral, acidic, basic, and sulfur-containing amino acids were involved in the development of the cooked squid-like odor. The odor of artificial extract cooked without addition of either octopine or ribose was lacking in the characteristics of cooked squid odor. Zinc ion as well as phosphorus compound accelerated the development of the cooked squid-like odor.

    Cyclic amines, pyrrolidine (Pyr) and piperidine (Pip), can form the corresponding carcinogenic N-nitroso compounds. The present study established a gas chromatographic determination method for these amines contained in food samples. Both amines extracted from samples were purified by cation-exchange chromatography, converted into the N-n-amyloxycarbonyl deriva-tives and then determined by gas chromatography on a mixture of 5.5% OV-17 and FFAP (10: 1 w/w). Aniline was used as an internal standard in gas chromatographic analysis. When the established method was applied to carp muscle and 12 kinds of canned fish products, Pyr was detected in all samples at concentrations ranging from 0.10 to 6.63μg/g while lesser amounts of Pip were in a few canned products only. It was noticeable that the canned dark muscle of tuna and skipjack contained especially high amounts of Pyr as compared to other samples. A part of Pyr in the canned products was assumed to originate from raw material.

    We have characterized and purified recombinant salmon growth hormone I (rSGH I). The primary structure of rSGH I confirmed by N-terminal sequence analysis, amino acid composition analysis, and tryptic and thermolytic mapping was identical to that deduced from SGH I com-plementary deoxyribonucleic acid except for an additional Met at N-terminal. Peptide mapping also revealed that rSGH I had two disulfide bonds, Cys49-Cys161 and Cys178-Cys186, as in mam-malian GHs. Recombinant SGH I, as well as natural SGH I, was classified as an alpha-helix rich protein from its circular dichroism spectrum. Recombinant SGH I was equipotent with natural SGH I in receptor binding assay. Gel filtration and sodium dodecyl sulfate polyacrylamide gel electrophoresis indicated that rSGH I exists as a monomer like natural SGH I.

    We found another type of protinase-independent modori phenomenon different from the ordinary type which we reported previously. In the case of this type of modori, gel-degrada-tion occured more severely the greater the temperature rise from 50°C to 85°C, while in the ordinary type it occurred evenly in a temperature range between 50°C and 85°C. These two types of modori seemed to be related to the magnitude of post mortem fall in muscle pH of the fish, because sardine muscle causing an extraordinary type of modori was found to have higher gel-forming ability, lower lactic acid content, and a higher value of muscle pH than those causing an ordinary type of modori. The presumption was proved positively by the fact that sardine mince in prerigor state treated by soaking in acidic saline water (0.3% NaCl, pH 5.8-6.0) had the ordinary type of modori, while the mince treated with neutral saline water (0.15% NaCl, pH 6.8) had the extraordinary type of modori.

    The development of antioxidative effect due to the Maillard reaction between L-tryptophan and D-glucose was found out to be closely related to the degradation of fructose-tryptophan (FT), an Amadori compound formed from these substrates. Therefore, FT was synthesized and used as a starting material for the Maillard reaction in this study. The antioxidative effect was developed by the decomposition of FT itself, while the extent of the antioxidative effect development was ac-celerated by the addition of free tryptophan. During the Maillard reaction of FT with tryptophan or glucose, FT degradation followed the first order reaction, whereas developments of brown color and fluorescence were the zero order reaction. The Maillard reaction products (MRP), which had the most potent antioxidative effect in this study, were used as an antioxidant for the preven-tion of sardine lipid oxidation. It was revealed that MRP effectively inhibited the oxidation of sardine lipid and had a synergistic effect with α-tocopherol.

    Various concentrations of sodium pyrophosphate (0-1.0mM, PPi) together with 1.OmM MgCl2 were added to walleye pollack myosin B (MB) in 0.5M KCl containing 40mM Tris-Buffer (pH7.0).
    Immediately after addition of PPi-MgCl2, measurements of viscosity and ultracentrifugal sedimentation pattern indicated dissociation of a considerable amount of myosin from MB.
    The first order rate constants (KD) for inactivation of Ca-ATPase of the PPi-MgCl2 treated MB were estimated at2-25°C and comparison was made with those of myosin. The results showed that the inactivation rate of Ca-ATPase of the PPi-MgCl2 treated MB was largely influenced by the concentration of PPi and temperature for the treatment. As the temperature was increased along with the concentrations of PPi-MgCl2 for the treatment, the formation of myosin from MB and the rate of denaturation of Ca-ATPase also increased.

    The thermal gelation of cross-linked oval filefish myosin with 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) was studied by means of SDS-polyacrylamide gel electrophoresis, sedimentation equilibrium, jelly strength, and chymotryptic digestibility. The cross-linked myosin with EDC formed a polymerized complex. The molecular weights from sedimentation equilibrium of EDC-myosin (4mg/ml) increased rapidly up to 1mM EDC, and slowly above the concentration of 1mM. The thermal gelation of myosin by heating at 30-50°C were enhanced in the presence of up to 25mol EDC/105g protein and the EDC more than 25mol effectively depressed “himodori”. The chymotryptic digestion of EDC-myosin suggested that there are two cross-linking sites with EDC above or below 1mM.

    Denaturation of carp myofibrils induced by changing pH in association with KCl con-centration was studied.
    Myofibrils were mixed with glycine-NaOH buffer to set the pH between 9.73 and 11.04 in the presence of varied concentrations (0.1 ?? 1.0M) of KCl and stored at 2°C. Alkali-induced denaturation of myofibrillar protein such as myosin and actin was investigated by means of changes in various ATPase activities and in the digestibility by α-chymotrypsin.
    The results showed that under the alkali treatment, the majority of myofibrillar protein was slowly denatured in a complexed form as actomyosin in the presence of a low concentration (0.1M) of KCl, whereas it was rapidly denatured through dissociation into myosin and actin in the presence of a high concentration (over 0.5M) of KCl.
    It was also shown that under the alkali pH, a fall in pH of myofibrils suspension brought about slow denaturation of actin together with myosin, while a rise in pH caused rapid denaturation of both actin and myosin.
