    To estimate the growth of swimming crabs Portunus (Portunus) trituberculatus in Osaka Bay, the carapace widths of juvenile crabs in shallow areas at Tarui and trawl-caught crabs at Izumisano fishing port were examined. Juvenile crabs inhabited the shallow areas from April to November. There were three groups of crabs. In one group, crabs appeared in July, grew rapidly and migrated offshore in September. In the others, crabs appeared in August or September and those that grew quickly migrated offshore in about October or November (second group) while those that grew less quickly migrated the following July or August (third group). The recruitment period to the trawling area was from July to November and the recruited groups corresponded to the groups in shallow areas. The average carapace width of each group in November was estimated to be 181-182mm (recruited in July-August), 169-172mm (recruited in September, female), 148-152mm (same, male), 144-152mm (recruited in October), and 105-115mm (recruited in November). The growth of crabs is well expressed by a logistic equation or Tanaka's ALOG growth curve.

    This paper describes the influence of water tank shape and size on the schooling behavior of fish. In this experiment, we used two dimensional water tanks of two different shapes, i.e. circular and square, of various diameters and side lengths between 50 and 125cm. Fish schooling behavior was expressed mathematically in terms of the Sannomiya and Matuda model which comprises of seven linear forces influencing fish behavior. The results showed that all forces except the propulsive force were independent of tank size and that schooling and attractive forces against the walls were strongest in the circular tanks. The random force was greatest in magnitude, accounting for approximately 60-75% of the total, followed by the repulsive force to the walls and the schooling force which affected fish schooling behavior. Fish schooling in square tanks tended to behave irregularly and remain in the tank corners.

    The effect of low light intensity on thallus and rhizoid growth of the germlings of Myagropsis myagroides and Sargassum horneri was studied. The germlings were cultured for 8 weeks at 20°C under two different light intensities with 12L/12D and total darkness. In both species, the thalli and rhizoids of the germlings grew well at 2000-2800lx. In S. horneri, furthermore, under this condition many adventitious branches were formed from rhizoids of individuals. In M. myagroides and S. horneri, the thalli and rhizoids of the germlings grew very slowly under the conditions of total darkness and 200-300lx. In both species, however, after 8 weeks under these conditions no individuals detached from the substratum or died. Moreover, the germlings of these two species had been cultured for 2, 4, and 6 weeks under these conditions, after which they were transferred to the condition of 2000-2800lx and cultured for 2 weeks. These germlings showed rapid growth after the transfer. In both species, the germlings which were cultured in this study were attached to the substratum by rhizoids. Staining with alcian blue revealed that an adhesive substance surrounded the rhizoids.

    We observed the behavior of two false killer whales Pseudorca crassidens in response to a variety of underwater pulsed sounds. The whales were kept in a net enclosure. We employed 15 different kinds of sounds and carried out 68 experiments. The behavior of the false killer whales after the sounds were transmitted was divided into three categories: “effective”, “somewhat effective”, and “not effective”. Effective behavior indicates that the false killer whales swam directly away from the source of the sound soon after transmission. At the beginning of the experiments, the sound pressure level of the effective category was above about 170dBrelμPa. This level rose as the number of experiments increased. A single pulse did not affect the behavior of the false killer whales. Pulseinterval and pulse-duration modulated sounds seemed to be more difficult to acclimate to than stable pulse sounds. To repel false killer whales, unexperienced sounds are thought to be more effective than experienced sounds.

    Water movement caused by a kyucho, a sudden swift warm current flowing into a bay, was surveyed in Yusu Bay in Ehime Prefecture, Japan. The kyucho flowed into the upper layer of the bay forcing two thirds of the bay water out of the bay through the bottom layer. Following this event, cold water from outside the bay flowed into the bottom layer and some bottom water flowed back again out of the bay. As a result, about five-sixths of the bay water was displaced by water from outside the bay. During one cycle of the kyucho event, the water on the bottom of the bay moved three times.

    The reproductive cycle of the sand snail Umbonium (Suchium) giganteum was investigated on the basis of samples taken monthly during the period from December 1986 to June 1988. Seasonal changes in the gonadal index, size-frequency distribution of the ovarian egg, and the results of a spawning experiment in the laboratory were analyzed to determine the spawning season.
    The gonadal index was low in August and September, but high during the period from December to June; the index values show a bimodal type of distribution with peaks in November-December and April. This is highly correlated to the seasonal changes in seawater temperature. As indicated from the period of maximum ripeness of gonad and the spawning observed in the laboratory, it is concluded that the spawning of U. (S.) giganteum occurs twice a year, i.e. in winter (December and January) and in late spring to early summer (April, May and June), and that the latter is the main spawning season.

    In an attempt to find suitable fishing gear solely to trap whelks Neptunea arthritica, three different trap designs were used to carry out a preliminary survey. Trap A had two entrances from the side, while traps B and C had one entrance on the top. A string of thirty traps set in order of trap A, trap B, and trap C and baited with about 900g of sardine were set at depths between 21 to 23m.
    In the four samplings conducted between July to December 1991, the soak time varied from 19h to 23h. The catch number differed significantly between the traps. The average catch number per trap/day in trap A was 43.9, in trap B 62.0, and in trap C 37.1. Mean catch rates in terms of weight varied from 1.40kg/trap in A, 2.22kg/trap in B, and 1.18kg/trap in C. The mean shell length taken by traps A, B, and C was between 61mm and 63mm. The overall results obtained from traps A, B, and C revealed that trap B was the most efficient for catching whelks. The present traps used in our survey were non-selective in catching whelks.

    Three species of the family Scombridae, viz. Auxis thazard, Katsuwonus pelamis, and Thunnus thynnus were studied for their karyotypes and cellular DNA contents. The first species is unknown as to its karyotypes or its cellular DNA content, which are described below. The diploid number of these three species is invariably 48, and composed of acrocentric chromosome in Auxis thazard and Katsuwonus pelamis, but in Thunnus thynnus the karyotype is composed of one pair of metacentric, two pairs of subtelocentric, and 21 pairs of acrocentric chromosomes. The DNA contents are 1.8pg/cell in Auxis thazard and 2.00pg/cell in Katsuwonus pelamis, these values are being included in the range of typical ones as members of the teleost group. Relative chromosome size ranges from about 1.2% to 3.3%, showing almost unimodal composition in size. The karyotypes of these three species of the family Scombridae are characterized by a smaller variability in size of chromosomes.

    The 5'-upstream regions from the carp Cyprinus carpio α-globin gene family were fused to the bacterial reporter gene, the chloramphenicol acetyltransferase gene. The transcription role was examined in a transient expression assay after transfection into fish nonerythroid cell lines and human erythroleukemia cell lines. Our studies demonstrated that carp α-like globin genes have functional promoter regions. In the fish GF cell line, CAT activity was enhanced. There was no preferential expression of the carp α-globin promoter-CAT constructs in the human erythroleukemia cell lines, K562 and HEL, compared with that in fish non-erythroid cells.

    A simple and secure method for growth estimation of blades has been devised where punching-holes along the midribs in blades are applied as growth markes of the blades in Undaria pinnatifida. The growth of blades in the apical direction was dependent upon the restricted regions which were contiguous to the lowest margin of the blades. The restricted regions expanded according to the growth of the blades, and the maximum distance from the lowest margin to the restricted growth regions measured 39.53cm. Exact growth estimation of the blades has been accomplished by measuring the distance moved by the punching-holes along the midribs at 40cm or over 40cm from the lowest margin of the blades in the present organism.

    Fish behaviour surveys in the Nayaura set-net fishing ground off Mie prefecture were carried out in relation to the fish leading effect of the natural bank.
    The topography of the fishing ground was surveyed by a side-scan-sonar and characterized by a long bank surrounded by large and small rocky reefs, which extend parallel to the leadernet from the shore to the mouth of set-net. A large number of fish schools, identified as Japanese anchovy Engraulis japonica from the catch data, were observed by the scanning sonar and recorded mostly in the daytime. The size of fish schools tended to be larger inside the natural bank area than outside. The natural bank in front of the leader-net affected the migrating route of fish schools. The migrating behaviour of fish schools were classified into seven categories. Of them, about 8% of the fish schools were seen to approach and move along the leader-net.

    We have attempted to promote the decomposition of organic matter deposited on the marine bottom below fish net pen culture by the biological activities of a deposit feeding capitellid polychaete, Capitella sp. 1. The organically enriched sediment was reproduced in the tub in the laboratory by the addition of moist pellets for fish culture on the sediment. We compared the chemical characteristics of the sediments between the experiments with and without Capitella. In these experiments, the Capitella colonies increased rapidly, and the biological activities such as feeding, reworking etc. efficiently decomposed the organic matter added on the sediment. The levels of organic matter of the sediment with Capitella fluctuated stably around the initial level throughout the period of the experiments for 60 days, while the levels in the experiments without Capitella gradually increased. The promotion of organic decomposition of the sediment in the experiments with Capitella also prevented the development of reduced conditions in the sediment. If we can re-establish a dense Capitella population on the bottom below the fish net pen quickly in the environmental recovery process from autumn to winter by releasing artificially cultured colonies of the worms, the potential of organic decomposition on the bottom will be significantly enhanced.

    The growth and maturation of 5- and6-year-old plants of Eisenia bicyclis were investigated by the leaf-marking method from June 1989 through June 1991 at Iwaki on the Pacific coast of northeastern Honshu, Japan. The length, diameter, and weight of each stipe was 50-60cm, ca. 3cm, and 400 ?? 600g throughout the investigation periods, respectively. The arm length was 13 ?? 14cm in 5-year-olds and 16 ?? l7cm in 6-year-olds. The weight of the lateral blades showed a seasonal peak in August or June, and a bottom in December or January, according to the seasonal changes in the weight and length of the lateral blade, the number of bladelets per lateral blade, and the number of lateral blades. Lateral blades with sori were observed throughout the year. The number of lateral blades with sori increased from August through January, when lateral blades without wrinkles on their surface showed an increase. Plants of both 5 and 6 years of age were found to produce 42 lateral blades and to discard 42 blades in a year. From the wet weight and the number of lateral blades falling off each year, it is estimated that the annual fallout product of both 5- and 6-year-old plants is ca. 2.1kg in wet weight.

    In chromosome manipulations of fish, duplication of genomes has been achieved generally by inhibiting the nuclear division. Recently, it has been also reported that genomes can be duplicated through fertilization with fused spermatozoa. In this study, fusion conditions of polythylene glycol (PEG) and motilities of fused spermatozoa were examined by using rainbow trout and nigorobuna spermatozoa. A comparison between two molecular weights of PEG indicated that a molecular weight of 4000 was superior in fusion ability to that of 1000. Fusion rate [(total number of spermatozoa participating in fusion/total number of tested spermatozoa) × 100] of rainbow trout spermatozoa was higher than that of nigorobuna. In a molecular weight of 4000, fusion rates of rainbow trout spermatozoa at concentrations of 30, 40, and 50% were 28.4, 40.2, and 44.4% respectively, while those of nigorobuna were 5.0, 18.6, and 34.8 respectively. When the diameter of the micropyle is taken into consideration, fused spermatozoa capable of entering an egg are restricted to those consisting of two spermatozoa. The maximum fusion rates between two spermatozoa in rainbow trout and nigorobuna were respectively 18.50% (concentration: 40%) and 15.9% (concentration: 50%). The duration of the motility of PEG-treated spermatozoa did not vary significantly depending on the three treatment concentrations, and compared with the control ones, the motion times of those of rainbow trout and nigorobuna were shortened by 2/3 and 1/2, respectively.

    A method is described for preparing a nonbiodegradable copolymer pellet containing luteinizing hormone-releasing hormone analogue (LHRH-a), des G1y10-[D-Ala6] LHRH ethylamide, polymerized by ultraviolet rays to produce sustained hormone release. The pellet was applied to induce gonadal development and maturation in red sea bream Pagrus major during the non-spawning season, from November to December.
    Single implantation of a copolymer pellet containing 100 μg LHRH-a induced gonadal development and spawning in completely immature fish 18 days after implantation, about six months prior to the nutural spawning season. Fish implanted with a 50 μg LHRH-a copolymer pellet or a control pellet were not induced to spawn. The histopathological findings of muscular tissues surrounding the implant showed that no inflammatory response to a foreign body appeared even one month after implantation. Results from the present study indicate that the LHRH-a copolymer pellets prepared by us have several features: (1) the pellets can be prepared easily and mass-produced at one time, (2) the pellets are useful for long-term hormone therapy in inducing sexual maturation in fish requiring a series of hormone treatments, and (3) the copolymer has good biocompatibility. However, the fertility rates of eggs produced were low. Further investigation is necessary to devise an effective method for obtaining more viable eggs by the use of copolymer pellets.

    Hijiki Hizikia fusiformis, brown alga, was steamed for 6h, and kombu Laminaria ocho-tensis, brown alga, and nori Porphyra spp., red alga, were boiled in a seasoned solution for 2h by imitating the manufacturing method of boiled and dried hijiki, and tsukudani (seasoned and cooked seafood) of kombu and nori, and changes in dietary fiber in these seaweeds were studied. The water soluble dietary fiber of all seaweeds increased steadily during heat processing. However, the molecular weight of the main components in the soluble dietary fiber decreased during heating, and especially in the case of nori it diminished markedly to 15% of the unheated sample after 1h. There were some differences in the changes of insoluble dietary fiber content and, as to kombu, the insoluble type was reduced to 40% of the unheated sample by boiling for 1h. Amounts of carbohydrates dissolved from hijiki increased gradually during steaming, and the molecular weights of the main components in this fraction were comparable to those of the soluble dietary fiber in the hijiki.

    Juvenile rainbow trout, weighing 0.12g on average, were fed on diets containing 0.2 and 2mg/kg of peptidoglycan (PG) derived from bifidobacterium thermophilum at a ratio of 3% body weight (BW) for 56 days, so that fish were given 6μg or 60μg PG/kg BW per day. On days 28 and 56, fish were challenged by immersing them in Vibrio anguillarum suspension, and the mortality was monitored for 21 days. PG did not influence the growth of fish at these doses. On day 28, PG-administrated groups showed a significantly higher survival rate compared with the control group. On day 56, however, no significant difference in the survival rates was observed between the PG-administrated group and the control group at either dose. These results indicate that the oral administration of PG enhances the resistance of rainbow trout against V. anguillarum infection, and a longer administration of more than 28 days may lead to a decrease in disease resistance.

    Kamaboko prepared with a combination of NaCl and using the suwari treatment (setting incubation) was used for the measurement of dielectric properties the state of bound water in dehydrated kamaboko. The apparent dielectric relaxation times of hydrated water were varied according to the combination of NaCl content and the suwari treatment. The hydrated water in the dehydrated kamaboko was divided in two groups with distinct regions, which are separated at the apparent dielectric relaxation time of 10μsec. A 2.5% NaCl-suwari-heated sample with an 8.7% water content showed low-and smaller high-frequency dispersions, and each relaxation time was about 634μsec and2.5μ sec, respectively. The mobility and amount of bound water seem to be strongly related to the gel strength and network structure of kamaboko. The present findings reveal that not only the amount of bound water but also the extent of water mobility, calculated as the dielectric relaxation time, is important for the analysis of the state of hydrated water in kamaboko.

    Activities and substrate specificities of crude enzymes of dimethylthetin-homocysteine (DMT-Hcys) and betaine-homocysteine (Bet-Hcys) S-methyltransferases from livers of fish (12 species), cattle, rat, and chicken were examined. Specific activities of DMT-Hcys enzyme were higher than those of Bet-Hcys enzyme in red sea bream and yellowtail, but vice versa in rainbow trout, flounder, and rat. In carp, cattle, and chicken, specific activities of both enzymes were almost the same. Substrate specificities of the DMT-Hcys enzymes from the liver of red sea bream were examined on a number of structurally similar compounds to DMT. Of all the test compounds, dimethylpropiothetin proved to be the most efficient substrate for the enzyme.

    A feeding experiment was conducted to determine the effect of various types of phosphates on zinc (Zn) availability to rainbow trout. One of the varieties of phosphates (Ca(H2PO4)2, CaHPO4, Ca3(PO4)2, NaH2PO4, Na2HPO4, KH2PO4, K2HPO4, H3PO4) or bone meal prepared from fish meal was supplemented to semipurified diets up to the same phosphorus (P) level as in the white fish meal based diet. Growth was reduced by the supplement of various kinds of phosphates and bone meal, but the supplementation of CaHPO4, did not affect growth. The supplementation of Ca(H2PO4)2 induced short body dwarfism, while Ca3(PO4)2 and bone meal supplement caused eye lens cataracts. Dietary Ca3(PO4)2 and bone meal lowered the Zn content in vertebrae, and moreover Ca(H2PO4)2 reduced both Zn and Mn contents. Zn absorption was affected by calcium phosphates, being lowest in fish fed the diet supplemented with Ca(H2PO4)2 and Ca3(PO4)2.
    The results of the experiment demonstrate that the excess amount of P in the dietinduced poor growth and suggest that the portion of P in tricalcium phosphate contained in white fish meal might mainly inhibit Zn availability, and that the proportion of Ca to P influenced Zn bioavailability to rainbow trout.

    The effect of culture conditions on L-histidine decarboxylation activity of halophilic hista-mine-forming bacteria (Photobacterium histaminum (C-8) and P. phosphoreum (N-14)) were studied, and the results obtained with both strains were shown as follows.
    The decarboxylation activity of the cells of both strains was in general the highest at the beginning of the stationary phase of the growth and decreased gradually during the stationary phase. The culture conditions of both strains for temperature and initial pH value which showed the highest decarboxylation activity were 15 ?? 20°C and 4.5, the lowest pH value for supporting growth, respectively. The development of activity in bacterial cells of both strains was shown in the presence of more than 10-3 M of L-histidine. The decarboxylation activity decreased in the presence of histamine in the culture medium.

    The effect of extrusion processing by a twin screw extruder on the nutrient utilization, and the availability of defatted soybean meal (SBM) as partial protein source were studied with rainbow trout of about 4-5g. In Expt. I, fish were fed the control and 30% SBM diets with and without extrusion processing for 6 weeks. In Expt. II, fish were fed ordinary steamed dry pellets (DP) containing 30% SBM or 30% extruded SBM (ExSBM) as a partial substitute for fish meal, and extruded soft-dry pellets (SDP) with 30% SBM for 8 weeks.
    Expt. I: There was no marked difference in growth or feed efficiency between the control and 30% SBM diets. Their dietary values and growth rate were not significantly improved by the extrusion, although the digestibility of starch was markedly elevated. The digestibility of dietary protein was not influenced by extrusion processing, ranging between 88 to 91%. Expt. II: Of the 30% SBM diets, SDP with a higher energy content produced higher growth, PER, protein, and energy retention than DP. Feed consumption was slightly lower in SDP with the higher energy level, leading to improved feed efficiency. Replacement of 30% SBM with the same level of ExSBM did not result in any nutritional improvement of DP.

    The potential of defatted soybean meal (SBM) as a protein source in high energy diets (extruded soft-dry pellets, SDP) was studied with rainbow trout of about 5g. The SDP contained respectively 0, 20, 30, 40, and 50% SBM, or 25% SBM plus 15% corn gluten meal (CGM) as a substitute for fish meal. The digestibility of SBM nutrients in SDP was determined using a reference diet and test diets which were formulated to contain both the reference diet and SBM in a ratio of 70:30, 60:40, and 50:50, respectively.
    The combination of 25% SBM with 15% CGM produced the best growth, while inclusion of SBM up to 50% did not reduce growth or protein utilization efficiency compared to the control diet. However, feed efficiency was slightly lower in SBM supplemental diets caused by the undigestible carbohydrate portion of SBM. The digestibility of protein, starch, and energy of SBM was 91, 54, and 80%, respectively, rather unaffected by the SBM levels in the formulation.
    It was concluded that SBM can be included up to 50% in SDP, replacing up to 91% of fish meal, supporting excellent growth and feed performance. A combination of 25% SBM and 15% CGM was confirmed to be an effective alternative protein source for rainbow trout.

    The effects of condensation of KCI solution by freezing on the denaturation of tilapia myosin B in 600mM KCl medium were estimated by measuring its Ca2+-ATPase activity in the presence of different KCl concentrations at 25°C. The KCl concentration was increased by freezing at a concentration from 600 to 3007mM at -11°C near the eutectic point; the Gibbs free energy changes ( ?? G) for this solution obtained from the heat of the solidification was 0 at -2.3°C and -163.1J/mol at -11.0°C.
    The Ca2+-ATPase activity was decreased as the KCl concentration increased at a concentration from 600 to 3007mM. It was found that ?? G for the Ca2+-ATPase system was strongly correlated to that for the solidification of KCl solution.
