Med., 1985, 146 (4), 379-383 - Mothers of offspring Balb/c mice were stimulated after birth by two substances, a bacterial lysate (LAB) and a chemical, diethyldithiocarbamate (DETC). Anti-sheep red blood cell (SRBC) antibodies were studied after immunization of stimulated mothers or offspring. An increase of anti-SRBC was observed in LAB-stimulated mothers, but these antibodies were decreased in their offspring before weaning. Sometimes, these antibodies were increased in LAB-stimulated newborn mice. DETC stimulation of mothers induced an elevation of antibody response in mothers and newborns. The same results were obtained in previous investigations where the pregnant mother was stimulated with the same agents.

Effect of Carteolol, Indenolol and Metoprolol on the Thyroid Hormone of Hyperthyroid Patients. Tohoku J. exp. Med., 1985, 146(4), 385-390-Thirty - four hyperthyroid patients were divided into four groups: placebo, carteolol (20mg/day), indenolol (60mg/day) and metoprolol (120mg/day) groups. The heart rate was determined before and two weeks after the treatment with each drug. Serum levels of T4, T3 and rT3 were measured before, and one and two weeks after the treatment. Both indenolol and metoprolol significantly reduced the heart rate (p<0.01 and p<0.05, respectively), whereas carteolol was ineffective. Carteolol significantly decreased T4 from 23.4±4.4μg/100ml to 20.1±3.2μg/100ml (p< 0.01) in one week and to 20.0±3.7μg/100ml (p<0.01) in two weeks. T3 and rT3 were also decreased significantly (p<0.05 and p<0.01, respectively) two weeks after the treatment with carteolol. After two weeks of indenolol treatment, T4 was decreased significantly from 26.2±8.5μg/100ml to 23.9±8.2μg/100ml (p< 0.05) and T3 from 789±391ng/100ml to 592±340ng/100 ml (p<0.02). Metoprolol had no effect on serum thyroid hormone levels. The decrease in the serum level of thyroid hormones by carteolol and indenolol but not by metoprolol in patients with hyperthyroidism would be due to the blockade of β2-adrenoreceptors concerning with thyroid hormone secretion.

Med., 1985, 146 (4), 391-400 -Human urinary chondroitin sulfate isolated from the cetylpyridinium chloride-complex of the non-dialyzable fraction of the pooled urine was subjected to ethanol fractionation, successive enzymic digestion with neuraminidase and mucopolysac-charidases, and anion exchange chromatography. The gas liquid chromatographic analyses of the acetyl and butaneboronic acid ester derivatives of the reduced terminal sugar units after treatment with sodium borohydride plus hydrolysis revealed that 42% of the urinary chondroitin sulfate was bound to peptide through xylose. The reducing terminal sugar units of the peptide-free form consisted of 34.6% of xylose, 22.4% of galactose, 16.4% of glucose of unknown origin and 26.6% of glucuronic acid. These observations showed that the xyloside, galactoside and glucuronide linkages at non-terminal sites of carbohydrate chains of chondroitin sulfate were cleaved in tissues. It was thus suggested that the endo-types of β-xylosidase, β-galactosidase and β-glucuronidase, which act on proteochondroitin sulfate are present in tissues.

    IIJIMA, T. and TAIRA, N. Abolition by Nicorandil of Spontaneous Action Potentials of the Small Atrio-Ventricular Node Preparation of the Dog. Tohoku J. exp. Med., 1985, 146 (4), 401-403 - Effects of carbachol and nicorandil on spontaneous action potentials were examined in the small atrio-ventricular (A-V) node preparation of the dog. Carbachol (3×10-6M) definitely hyperpolarized the membrane potential and abolished the spontaneous activity. Nicorandil (3×10-4 M) also abolished the spontaneous activity but the membrane potential became quiescent at -65mV, which was less negative than the maximum diastolic potential. From these results it appears that nicorandil may increase the potassium conductances but not hyperpolarize the membrane potential because the background potassium conductance is little developed in the nodal cells.

Med., 1985, 146 (4), 405-417 -Specific antibodies to fibronectin and type I collagen in rat livers were used to demonstrate these matrix proteins with direct immunoperoxidase method in paraffin sections of normal liver, DAB-induced hepatoma and CCl4-induced fibrotic liver. In normal livers, the immunoreactive products of both matrix proteins were found in the periportal regions, while the hepatocytes and most of the interstitial matrix remained unstained. The specimens obtained from DAB-treated liver showed a more intense reaction with fibronectin antibody in the perisinusoidal space including proliferated cholangiolar cells, as compared to no reaction with type I collagen antibody. In CCl4-fibrotic liver, apparent reactions were also found for both matrix proteins in the periportal interstitium and in progressing fibrotic area with severe fatty metamorphosis. These findings suggest that these matrix proteins have an advantage in the attempt to distinguish different patterns of neoplastic alteration in experimental rat livers.

Changes in the Immunological Parameters after a Single Dose of Forphenicinol, a New Small Molecular Immunomodifier. Tohoku J. exp. Med., 1985, 146 (4), 419-427 - Forphenicinol [L-2-(3-hydroxy-4-hydroxymethylphenyl) glycine, M.W. 197.19] is a derivative of forphenicine, an inhibitor of alkaline phosphatase discovered by Umezawa. In order to find an optimal dose, a single dose of the drug ranging from 10 to 600mg per body was orally administered to a total of 55 patients (36 cancer, 13 tuberculosis, and 6 others). The possible changes in the percentages of the peripheral T and B lymphocytes, natural killer (NK) activity, and the proliferative response to phytohemaglutinin (PHA) were studied as immunological parameters. A significant effect of forphenicinol was demonstrated in restoring the normal proportion of T and B cells, especially in those who had 'low'-T and/or 'high'-B before administration of the drug. No difference was found between cancer and tuberculosis. The mean percentage of T cells increased from the low initial level of 68.0 to 75.6 in cancer (n=12, p<0.05) or from 63.1 to 78.5 in tuberculosis (n=7, p<0.05), while that of B cells decreased from the high initial level of 34.6 to 26.9 in cancer (n=7) or from 34.9 to 14.3 in tuberculosis (n=6, p<0.025). The effect of a single dose of the drug tended to disappear by day 8, a peak response being found on day 3 in most cases. With respect to this parameter, an optimal dose was found in a range from 60 to 100mg. Forphenicinol was rather inhibitory on the NK activity, while it exerted diverse effect on the lymphocyte proliferation. Med., 1985, 146 (4), 429-437 - Analysis of endometrial carcinoma of 15 cases and the normal endometrium from 12 control cases was made using flow cytometry. The proliferetive activity and the ploidy level of the tumors were determined and a comparative study of the tumors classified by the nuclear grade of the tumor was made. In comparison with normal endometrium, endometrial carcinoma tissue had a significantly larger S-phase fraction of the cell cycle. Moreover, among the endometrial carcinoma cases, the percentage of S-phase cells was found to increase with increases in abnormalities of nuclear morphology. Endometrial carcinoma cases had higher levels of G2+M phase fractions than normal endometrium, but not significantly so. The proliferation index of endometrial carcinomas was significantly higher than that in normal endometrium, and it was found that proliferative activity became more robust the more irregular the nuclear morphology. With regard to the ploidy level, all of the normal endometria and Nuclear Grade 1 or 2 endometrial carcinoma cases showed a DNA distribution between diploid and tetraploid. In contrast, Nuclear Grade 3 cases could be subdivided into two groups, one which had a DNA distribution between diploid and tetraploid with an extremely high proliferation index and the other which had an aneuploid DNA stem line.

Med., 1985, 146 (4), 439-447 - In order to investigatee the effect of gastrointestinal hormones upon the secretion of extrapancreatic glucagon, tetragastrin, secretin, caerulein and cholecytokinin-pancreozymin octapeptide (CCK-octa) were administered during saline or arginine infusion in pancreatectomized dogs. Intravenous administration of tetragastrin (7μg/kg) did not elicit any changes in plasma glucagon during saline infusion, while the plasma glucagon increased significantly following tetragastrin infusion during arginine infusion. The administration of secretin (3U/kg) did not affect the plasma level of glucagon during saline or arginine infusion at all. Plasma glucagon did not change after the administration of caerulein (0.5μg/kg) during saline infusion, whereas it increased significantly following caerulein administration during arginine infusion. Intravenous administration of CCK-octa in a dose of 20U/kg did not affect the plasma level of glucagon during saline infusion but exerted a significant rise of extrapancreatic glucagon during arginine infusion. It is concluded from the present experiment that the administration of tetragastrin, caerulein or CCK-octa enhances the release of extrapancreatic glucagon stimulated by arginine infusion while secretin infusion does not affect the secretion of extrapancreatic glucagon.

Circulating Immune Complex-Like Materials Which Bind to Heat Inactivated Clq Interfere with the Clq Solid Phase Assay for Immune Complexes. Tohoku J. exp. Med., 1985, 146 (4), 449-456, - Clq solid phase assay (Clq SP) was devised based on the fact that immune complexes (IC) and aggregated human globulin (AHG) bind to Clq. Neither IC nor AHG was found to bind to heat inactivated Clq. On the other hand, circulating immune complex (CIC)-like materials in patients' sera were able to bind to heat inactivated Clq, indicating that these CIC-like materials are not true CIC. Gel filtration analysis showed that molecular size of such CIC-like materials was almost the same as monomeric IgG, while Lure CIC were in heavy fractions. True CIC did not bind to heat inactivated Clq but bind only to native Clq. The CIC-like activity is not due to rheumatoid factors. About 2/3 of CIC positive sera by Clq-SP are not really CIC positive but are due to interference by the CIC-like materials.

    MUNAKATA, H., ISEMURA, M. and YOSIZAWA, Z. Glycosaminoglycans in Brush Border Membranes from Rabbit Small Intestine. Tohoku J. exp. Med., 1985, 146 (4), 457-460 - A brush border membrane fraction was prepared from mucosae of rabbit small intestine by differential centrifugation. The membrane fraction was digested with pronase and a glycosaminoglycan fraction was isolated from the digest by fractionation with cetylpyridinium chloride. Individual glycosaminoglycans were quantitated by specific mucopolysaccharide-degrading enzymes and nitrous acid. The glycosaminoglycan composition was then calculated to be as follows: Hyaluronic acid, 34%; chondroitin sulfate A/C, 16%; dermatan sulfate, 11%; heparan sulfate, 39%.

Tohoku J. exp Med., 1985, 146 (4), 461-467 - The middle ear effusion specimens were obtained by myringotomy and aspiration from 4 children of 4-7 years old, who had been diagnosed as patients with secretory otitis media on the basis of conductive hearing loss and tympanogram. In cases 1 and 2, their ear fluids were macroscopically serous, while those of cases 3 and 4 were mucous. These ear fluids were digested with pronase and the digests were analyzed by cellulose acetate membrane electrophoresis with alcian blue and high-iron-diamine stainings. All samples were found to contain glycopeptides possibly derived from sulfated mucin-type glycoproteins with small amounts of glycosaminoglycans. The glycoconjugates from cases 3 and 4 were further examined after hyaluronidase and chondroitinase ABC treatments, followed by heparitinase digestion. The resultant glycopeptide fractions appeared to be electrophoretically homogenous and their chemical compositions suggested that they were typical mucin-type glycopeptides. Furthermore, they contained sulfates. The data suggest that in secretory otitis media, one of the major components of middle ear effusions is sulfated mucin-type glycoprotein.

Med., 1985, 146 (4), 469-478 - A case of primary extramedullary plasmacytoma (PEMP) with local amyloid deposition arising in the left parotid gland of a 78-year-old woman is described. The present case was studied with immunologic techniques with results confirming that the neoplastic cells were of monoclonal nature, producing λ-light chains alone. PEMPs of the salivary glands are rare, with only 8 cases reported in the literature. The salient features of these 9 cases are summarized. Problems connected with the clinical, pathological and therapeutic aspects of PEMP with regard to the rare parotid location are discussed.

Med., 1985, 146 (4), 479-480 - A technique for indirect measurement of blood pressure by a combination of commercially available apparatuses was devised. There were good correlations in both of systolic and diastolic blood pressures measured by the indirect and direct methods.

    TSUKAMOTO, T., DIRINGER, H. and LUDWIG, H. Absence of Autoantibodies against Neurofilament Proteins in the Sera of Scrapie Infected Mice. Tohoku J. exp. Med., 1985, 146 (4), 483-484 - Autoantibodies against neurofilament proteins were not detected in any of the sera from the following scrapie infected mice: 19 mice infected with scrapie agent 139A in pre-clinical stage, 32 histologically confirmed scrapie mice and other 12 clinical scrapie mice infected with various strains. The test sera were assayed against acetone-fixed central neuron cultures from fetal mice by indirect immunofluorescence and immunoperoxidase techniques. The negative result suggests that autoantibodies against neurofilament proteins do not play a role in the pathogenesis of scrapie.
