In vitamin E deficient rats, increase of cysteine protease inhibitor (CPI) level was observed not only in plasma, but also in other organs. Concerning this phenomenon following results were obtained. 1. Plasma (or serum) cysteine protease inhibitor (α-CPI) level increased markedly in vitamin E deficient, muscular dystrophic rats, while serine protease inhibitor level was almost unchanged. 2. In vitamin E deficient rats, plasma α-CPI level closely correlated with plasma pyruvate kinase activity. 3. In testis of vitamin E deficient rats, ficin inhibitor level increased markedly, but trypsin inhibitor level was unchanged. 4. In liver, heart, spleen, lung, kidney and muscle, CPI level was also increased by vitamin E deficiency. 5. Plasma α-CPI and tissue CPI increased by vitamin E deficiency were immunologically homologous to T-kininogen. 6. By antioxidant ethoxyquin, increase of the plasma α-CPI level in vitamin E deficient rats were suppressed. From these results. roles of CPI and T-kininogen in vitamin E deficient rats were discussed.

Ascorbate oxidase is a copper-containing blue enzyme that occurs in plant tissues. Ascorbate oxidase expression in pumplin was studied. Ascorbate oxidase activity and protein rapidly increased during formation of pumpkin callus. Ascorbate oxidase was secreted into the culture medium in the cell suspension cultures. supporting the idea of cell wall localization of ascorbate oxidase. In addition, calcium joins markedly stimulated the secretion of ascorbate oxidase. A full-length c DNA clone for ascorbate oxidase was isolated. The amino acid sequence of pumpkin ascorbate oxidase is homologous to those of the other blue multicopper oxidases. Northern blot analyses showed that ascorbate oxidase was induced by auxin, suggesting that ascorbate oxidase is involved in cell growth. In pumplin callus, ascorbate oxidase activity and protein could be markedly increased by adding copper, a prosthetic metal of the enzyme. However, Northern blot analyses showed that ascorbate oxidase mRNA did not increase by adding copper. This suggests that copper controls ascorbate oxidase expression at translation or at a site after translation. A genomic clone for ascorbate oxidase was also isolated.

The body weight, liver weight, contents of thiobarbituric acid-reactive substances, flavins and glutathione in the liver, activities of superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase and glucose-6-phosphate dehydrogenase in the liver cytosol fraction, and activities of GOT and GPT in the serum of mice were measured at 24 hours after administration of ethanol as parameters of the acute alcoholic liver injury. In the groups of administration of 86.0 or 103.2 mmol/kg of ethanol, the acute alcoholic liver injury was recognized. When administered 0.19 mmol/kg or riboflavin, flavin mononucleotide, or flavin adenine dinucleotide (FAD) to mice suffered from the acute alcoholic liver injury caused by the administration of 86.0 mmol/kg of ethanol, the suppressive effect of each flavin was recognized and FAD worked most effectively. We considered that the acute alcoholic liver injury was related to the lipid peroxidation and that it was suppressed by the administration of FAD.

Vitamin D and provitamin D in 89 kinds of Japanese foods were assayed by an HPLC method. The method was confirmed to be useful for the determination of the compounds in various kinds of food samples, e. g., fishes and shellfishes, meats, eggs, milks, fungi, cereals, potatoes, pulses, vegetables, fruits, algae, confectoneries, fats, oils and seasonings. Vitamin D contents were generally high in fishes (especially flatfish, bastard halibut, eel, red sea bream, yellowfin tuna, mackerel, skipjack, yellowtail, Japanese pilchard and horse mackerel : higher than 500_<IU>/100g) and fungi (especially dried Kikurage and dried Shiitake : higher than 500_<IU>/100g). Although cow's and human breast milks contained the active vitamin D metabolites, the biological activities were very low (approximately 13_<IU>/100g in both milks). Eggs also contained low contents of 25-hydroxyvitamin D besides vitamin D. Vitamin D in meats including liver and the other foods were under the limit of detection. Provitamin D contents were high in fungi. Vitamin D in foods was not decreased by solar-drying and cooking, e. g. baking, boiling and frying.

Effects of dietary calcium intake on bone mineral contents, density and strength in marginally vitamin D-deficient rats investigated. Vitmin D-deficient rats were fed ad libitum one of the diets containing various calcium concentrations (0.22, 0.44, 0.88 and 1.20%) prepared by L-calcium lactate with oral supplementation of a low level of vitamin D_3 (0.35 IU/rat, twice a week) for 28 days. During and after feeding, concentrations of calcium, phosphate, 25-dhyroxyvitamin D_3, 1 α, 25-dihydroxyvitamin D_3 and alkaline phosphatase activity in plasma were measured. Furthermore, femoral ash weight, mineral density and strength were also measured. In spite of marginally vitamin D deficiency, increased calcium intake provided increased plasma calcium level, bone growth, mineral density and strength. However, the magnitudes of increased bone mineral contents, density and strength in marginally vitamin D-deficient rats were significantly lower than those in vitamin D-sufficient rats reported previously. These results suggest that sufficient amount of vitamin D supplementation with an appropriate calcium intake are essential for completing normal bone growth, although increased calcium intake increased some extents of plasma calcium level and bone mineral contents, density and strength even in marginally vitamin D deficiency.
