Nuclear receptors belonging to the NR1H and NR1I subfamilies, including vitamin D receptor (VDR, NR1I1), control cholesterol and bile acid metabolism. Bile acids are detergents essential for the digestion and intestinal absorption of hydrophobic nutrients, such as triacylglycerol, cholesterol and lipid-soluble vitamins, including vitamin D.Primary bile acids are generated from cholesterol and are secreted in bile as glycine and taurine conjugates. The intestinal microflora convert the primary bile acids to the secondary bile acids, including lithocholic acid (LCA). VDR, a receptor for 1,25-dihydroxyvitamin D_3(1,25(OH)_2D_3), acts as a bile acid receptor with specificity for the secondary bile acid LCA and its derivatives. VDR activation by 1,25(OH)_2D_3 or LCA induces the xenobiotic metabolism of bile acids. Synthetic LCA derivatives induce tissue VDR activation without inducing hypercalcemia in mice. VDR acts as a bile acid sensor as well as an endocrine receptor for vitamin D signaling.

Evidence has accumulated that collagen cross-links play important roles in bone strength. We have demonstrated that the quantitative and qualitative deterioration of lysyl oxidase controlled and non-enzymatic cross-links (Advanced glycation end products, AGEs, Pentosidine) of collagen in patients with osteoporotic femoral neck fracture cases might be affected by hyperhomocysteinemia, oxidative stress and B-group vitamins insufficiency. Recently, Shiraki et al. demonstrated that a functional polymorphism in methylenetetrahydrofolate reductase (MTHFR) polymorphism, T allele (C677T), may be a risk factor for future fracture in addition to the traditional risk factors. In addition, we have reported that a higher urinary pentosidine was an independent risk factor, for vertebral fracture in a 5-year prospective study in Japanese post-menopausal women. If confirmed in large, prospective trials, measurement of serum homocysteine and serum or urinary excretion of pentosidine might be characterized as markers reflecting bone collagen deterioration.

Tetrahydrobiopterin (BH_4) is an essential cofactor for biosynthesis of monoamine neurotransmitters including dopamine and serotonin. Here, we report the levels of pteridines, i.e. BH_4, dihydrobiopterin, biopterin, dihydroneopterin, neopterin, in the cerebrospinal fluids (CSF) of patients with neurological diseases including sporadic Parkinson's disease (sPD), frontotemporal dementia (FTD), multiple system atrophy (MSA), and meningitis. Each pteridine was directly measured by a post-column oxidation method. Determination of reduced form of pteridines by a post-column oxidation method in biological fluids, including CSF, serum, and urine, would be useful to monitor the level of oxidative stress in vivo.
