    Adenovirus types 6 and 31 were isolated from an outbreak of epidemic diarrhea in an infant home in late December 1964. Infection of adenovirus type 6 was confirmed by the antibody response. The antibody response to adenovirus type 31 was proved in the sera six months, but not two months, after the infection. Prevalence of adenovirus type 31 among children near Tokyo was confirmed by examination of neutralizing antibody to the virus. The possible association of adenovirus infection with the outbreak of infantile diarrhea is discussed. Some properties of adenovirus type 31 isolates are described.

  • II. QUANTITATIVE DETERMINATION OF THE HEAT-LABILE TOXIN BY SKIN REACTION

    To establish a quantitative method for titration of the heatlabile toxin of Bordetella pertussis by the skin reaction, various conditions were examined and their relative merits assessed.
    The skin reaction of the rabbit was characterized by a pale, ischemia-like spot with or without petechial hemorrhage, while hemorrhage was prominent in the guinea pig and suckling mouse. Mean diameters of the pale spot in the rabbit and of the hemorrhagic spot in the other species provided a linear regression on log dose over a relatively wide range. The regression co-efflcient was not affected by endotoxin or the lymphocytosis-promoting factor, though endotoxin intensified the reaction in the guinea pig and suckling mouse. These facts enabled us to construct a statistically valid assay method. Animal variations were controled by expressing the toxicity relative to that of a reference.
    An absolute unit of the toxicity was defined. Normalization of toxicities determined under different conditions is possible.

    Differential centrifugation of a sucrose homogenate of tuberculous mouse spleens was conducted to obtain sediments of cytoplasmic granules at 1, 000 g, 3, 900 g, 8, 400 g, 15, 000 g, and 25, 000 g for 20 min. Acid phosphatase activity of the Triton-extract of each sediment indicated that the 15, 000 g and 25, 000 g sediments were rich in lysosomes. A large-molecular mycobactericidal fraction associated with cathepsin and acid phosphatase activities could be separated from the detergent-extracts of the 8, 400 g, 15, 000 g and 25, 000 g sediments by gel-filtration on Sephadex G-150 column, but not from the 3, 900 g sediment. From the lungs of the same group of mice, such a mycobactericidal fraction was not obtained.
    The active fraction from the spleens was further treated with one of the following three agents; 90% acetone, ethyl ether, and 2M urea. The mycobactericidal activity was found in the 90% acetone-soluble fraction and also in the small-molecular protein fraction released by ether-or urea-treatment. Both subfractions were free from the enzymatic activities. Their mycobactericidal activity was heat-stable at 100 C for 5 min. The difference between the two was that the 90% acetone-soluble subf raction was more effective with less Folin value and with less exposure time to the bacilli, but more dependent on acidic environment than the protein fraction.

    To find the distribution and the favourite site of localization of Toxoplasma in the cases of latent infection occurring naturally, the organs and other materials from 14 cats, 26 dye test-positive and 11 dye test-negative dogs were examined by inoculation into mice. Ten cats and 9 dogs were found to have been infected latently. In the cats, the organisms were isolated frequently from the diaphragm, brain, lungs, abdominal muscles, and heart muscles, and less frequently from the stomach, small and large intestines, mesenteric lymph nodes, spleen, and bile bladder. In the dogs, the parasites were demonstrated frequently in the diaphragm and abdominal muscles, and occasionally in the heart muscles, brain, small and large intestines, spleen, and uterus. The musculature particularly the diaphragm was found to be the most favourite site for the parasites in latently infected cases. A larger number of virulent strains killing mice within 1 or 2 weeks were found among the cat strains than among the dog strains.

    A study is presented of the successful application of guinea pig sera as a source of the accessory factor to the toxoplasma dye test. A total of 209 serum samples were obtained from guinea pigs of different ages from 10 to 185 days. The sera were tested for possible usefulness as an accessory factor with the aid of Alsever solution. Without addition of Alsever, all the sera deprived the toxoplasma cytoplasm of its affinity for methylene blue. To the contrary, when Alsever was added, approximately 20 % of the serum samples were proved to be useful as a source of the accessory factor. Three-fifths of these qualified sera produced titers higher by 1 tube or 4 fold for a positive control serum than that estimated by using a human accessory factor serum, while remaining two-fifths showed the same titer as that with the human serum.
