  • 梅酒熟成に関する研究 (第1報)

    梅酒の熟成中における糖, 酸度, pH, 色調の変化について, 30℃および常温保存の試料を, 原料混合から1年にわたって追跡し, つぎの結果を得た.
    1) 漬け込み後4日目くらいまでは固体のショ糖の溶解のみが起こって全糖度が増加するが, ショ糖の転化は起こらない.
    2) ショ糖の転化は4日後ころから起こり, 10~90日くらいまでは急激に, 以後は徐々に進行してほぼ200日後に完了する.
    3) 全酸度は30℃および常温保存試料について, 漬け込み直後6.1および4.4であったものが30日後まで急増してそれぞれ20.0および19.6となり以後徐徐に増加して365日目にはそれぞれ25.4, 21.6となった.
    4) pHは最初3.2くらいで4日目には3.0に下がり, 以後熟成中あまり変化は認められなかった.
    5) 色調の変化は日数の経過とともにほぼ直線的に着色が進行した.これは糖の褐変が関与しているものと考えられる.
    以上の結果にもとついて梅酒熟成中の成分変化について考察した.

    Recently, fine granules have widely been used for dispensing, because they are free from defects peculiar to powder and granule. Twenty-four commercial preparations of fine granules stored in the Department of Pharmacy, Osaka University Hospital, were evaluated for their properties in compliance with J. P. IX and the standard of fine granules for dispensing proposed by Aoki. Sense tests were also made in order to determine whether the fine granules are fit to dispensing. The preparations which did not comply with J. P. IX and Aoki's standard were deemed unqualified for dispensing. Some preparations did not pass the sense tests, though they bore the above examinations. Efforts should be made to minimize granules which pass through the 200-mesh filter.

    The relation between dry and moist states of potato starch and change in physical properties of the powder (agglomeration, dispersibility, apparent specific volume and angle of repose) by the addition of various lubricants (talc, magnesium stearate, calcium stearate, synthetic aluminum silicate and hydrous silicic acid) were studied. Following results were obtained: 1) Effect of hydrous silicic acid and synthetic aluminum silicate added to dried potato starch was most potent, and that of magnesium stearate was weakest. 2) When added to moist starch, hydrous silicic acid and synthetic aluminum silicate were most effective, but it was notable that calcium stearate, which was not so effective when added to dried starch, proved effective in moist starch. Almost all of the lubricants proved more effective in moist starch than in dried starch. 3) The effect of hydrous silicic acid added to dried starch was not changed even after the starch was moistened. All other lubricants reduced their effect after moistening. Especially, the lubricant effect of talc and magnesium stearate was lost when the water content of starch exceeded about 13%.

    梅酒の香気成分について, 減圧水蒸気蒸留法とヘッドスペース法で得られたにおい濃縮物を用い, GC-MS によって成分の同定, 定量を行った. また 1, 2, および 5 年間貯蔵した試料について貯蔵中の香気成分の変化を研究した.
    その結果, 減圧水蒸気蒸留法ではアルデヒド類, 酸類, アルコール類, エステル類などを含めて 38 成分, ヘッドスペース法では減圧水蒸気蒸留では認められなかった低沸点成分 6 種を含めて 22 成分を同定, 定量した. 成分の数は貯蔵に伴って増加し, とくにエステル類は成分の数, 含有量ともに顕著に増加した. これは貯蔵中に梅の実中の有機酸が順次溶出され, 多量に存在するエタノールとエステルを作るものと考えられ、た. 最も含量の多い成分は benzaldehyde であったが, その含有量は貯蔵中に顕著に減少し, 同時に benzoic acid, 化を受け, さらにエステル, アセタールへと変化するものと考えられた.
    以上により, 梅酒の香気成分は, はじめ比較的単純な組成であったものが貯蔵により複雑な組成となり, マイルドな風味に変化すると考えられた.官能検査の結果もこの推定と一致した.

    Improvement on the physical properties ot the tollowing 4 samples, highly adhesive and agglutinative, was tried: Adsorbin, calcium lactate, Laxatol powder, and yokuinin extract powder. After adding mgnesium stearate to the 4 samples, their packaging errors and recovery of weight after packing were determined with packaging machines having different valves (Konishi KC-747-K Type 12 with a plastic valve type 12 and with a stainless steel valve).
    In all the 4 samples, the packaging error was smaller with the machine having the plastic value than with a stainless steel. By addition of 0.4% magnesium stearate, the physical properties, packaging error and recovery of weight of Adsorbin were improved, thus suggesting that the method is usuful for drug dispensing. An improvement on physical properties was also found in the remaining 3 samples, but no improvement on the packaging error and recovery of weight.

    Phenytoin (PHT) fine granule, which has the same bioavailability as the tablet, has recently been marketed. We projected a change of PHT dosage form from usual powder to fine granule. In view of the fact that 97% of PHT products prescribed and marketed are available in fine granules, 50% PHT preparations were produced in hospital pharmacy. 5 kinds of preparations using various lactoses as diluents were prepared and tested for their quality. Among them, the preparation from Aleviatin fine granule passing through a 42-mesh sieve and EFC lactose showed the best results in the mixing property and various sense tests. The change to this dosage form and its bioavailability tests were carried out by a cooperation of doctors and pharmacists. Serum PHT levels of all patients, assayed by EMIT method, showed a 20% to 30% increase after the administration of the new preparation (50% PHT fine granule with EFC lactose).

    In order to examine the efficiency of dispensing work in the stage of dividing and packing of powders, by new automatic dividing and packing machines introduced, Monte Carlo simulation was carried out by a computer. At the same time, the new machines were tested for error of dividing weight, the accuracy of dividing weight in relation to the number of subpacks, and the rate of loss. The machines tested were TOKYOSYOKAI OMP-90 type (OMP) equipped with a linear vibrator to feed powders into the division holes by alternating motion, and YUYAMA YS-63R type (YS) with a linear vibrator to feed into the revolving disk (80rpm), which was formed by gap of V-like. Fine Lactose, SM®, powder, Crystalline Lactose, MIYA-BM®, Fine Granule, and Berizyme®, Granule were selected as the sample for the studies.
    As a result of simulation, YS was more efficient than OMP in the service system. YS showed smaller variations of dividing weight than OMP tested. In case of OMP, dividing weight of the third and the third last packages at the turn of linear vibrator, was much heavier than that of other packages. In some of these drugs, errors in dividing weight increased with a greater number of subpacks.For the rate of loss, YS was slightly smaller than OMP tested.
    As for the automatic dividing and packing machine, the accuracy of dividing weight is contrary much to efficiency of dividing and packing speed. Therefore, these results suggest that this method is much useful to select a machine by using the two kinds of contrary factors from the practical point of view.
