    Tumor-infiltrating lymphocytes (TIL) were tested for cytotoxicity against autologous tumor cells in a study utilizing a chemically induced cancer ofthe bladder (transitional cell carcinoma),
    BC
    -47, in inbred ACI/N rats. From tumors grown after subcutaneous implantation of
    BC
    -47 in the rats TIL were separated by density gradient centrifugation and incubated in plastic dishes for separation of non-adherent from adherent cells. The non-adherent cells were further fractionated into T and
    B
    cells by the panning method using anti-rat F(ab')2 antibody. The cell fractions were each added to
    BC
    -47 in culture to be assessed for antitumor effect by the crystal violet dye exclusion method and 3H-thymidine incorporation inhibition assay. Peripheral blood mononuclear cells (PBMC) were also tested as described above. TIL expressed significantly higher cytotoxicity against
    BC
    -47 with the mean % cytotoxicity of 56.6±5.6% and 87.5±

    One method currently being considered for the disposal of high level radio-active waste is burial in the seabed. When a hot object is buried in soil, the temperature of the soil adjacent to the object is raised and there is transient heat flow away from the object. The rise in the temperature of the soil causes excess pore pressures to be generated and transient pore water diffusion also takes place. A finite element formulation of the coupled heat flow and consolidation problem is presented. The computer implementation of this formulation is found to compare favourably with analytical solutions based on the same physical principles.

    This study aimed to determine the prevalence of fecal carriage of extended spectrum β-lactamase (ESBL) and/or plasmidic AmpC β-lactamase (pAmpC) producing Escherichia coli among dogs (n=428) in Turkey. Polymerase chain reaction (PCR) and sequencing were used to characterize genes encoding β-lactamase and plasmid mediated quinolone resistance (PMQR). Antimicrobial susceptibility testing and PCRs for virulence genes and phylogenetic groups were also performed. Cefotaxime resistant

    The Itoigawa-Shizuoka Tectonic Line (ISTL) in central Japan is a complex 150km-long fault system consisting of north-trending east-dipping reverse, northwest-trending left-lateral strike-slip, and north-trending west-dipping reverse faults. To help to resolve segmentation and to estimate the magnitude of future shocks on the ISTL, we conducted four trench excavations across the Hakushu fault, the Shimotsuburai fault, and the Ichinose fault group in the southern part of the ISTL, where no paleoseismological data for the surface faulting had been available. On the trench walls at the Hakushu fault, we found the evidence for the most recent surface-rupturing event occurring sometime between 6, 650 and

    In the first part of the present work the interaction of glycophorin with dimyristoylphosphatidylcholine (DMPC) is studied by freeze fracture electron microscopy, densitometry, calorimetry, and 90° static light scattering. An exothermic lipid/protein interaction energy of WP=190 kJ•mol-1 was found by application of the well known Van Laar relation for the displacement of the freezing point and the Gibbs-Duhem relationship. Secondly, the effects of Ca2+ on the lipid/protein interaction were studied. Following Ca2+ addition a remarkable decoupling of the interaction of the glycophorin head group with the bilayer surface was revealed by densitometry and gold-labeling electron microscopy. It is estimated that about 80% of lipid once disturbed by the adsorption of glycophorin head groups is decoupled after addition of Ca2+. Thirdly, the selective interaction of glycophorin with binary lipid mixtures was studied, including the mixtures of DMPC with dimyristoylphosphatidylserine (DMPS) and dilauroylphosphatidylcholine (DLPC), and the mixture of dipalmitoylphosphatidylcholine (DPPC) with DLPC.

    Archaeomagnetic field intensity in Finland (Lat. 61°N, Long.

    ) has been determined for the past 6400 years with the Thellier technique of bricks, potsherds and baked clays. The normalized archaeointensity curve for Finland, based on 23 mean values, shows an increase from ∼4360
    BC
    to the maximum at
    AD
    500, after which it decreases to the present value. The high intensity at
    AD
    500 as found in Finland, but not in Bulgaria and Japan, is not a consequence of the applied Thellier technique since we are able to reproduce the known field intensities in the laboratory, and some of the dated bricks yield values which are in broad agreement with Nurmijärvi Magnetic Observatory data. We also show that variations in grain size of the magnetic carriers, cooling rate or magnetic refraction are unlikely to cause systematic errors in intensity larger than a few percent. Based on results of self-made bricks we show that the archaeointensity, however, depends on the angle between the laboratory and ancient field directions. If these are not aligned errors up to ±10% in archaeointensity can occur and are probably caused by anisotropy of remanence or by other factors not yet understood. The high intensity at
    AD
    500 in Finland can be modelled by a radial nondipole field source at the core-mantle boundary producing enhanced field at higher latitudes (Finland) and relatively weaker fields at lower latitudes (
    e
    .g. Bulgaria and Japan). Extrapolation of the present field (IGRF 1990) back in time shows similar field behaviour.

    BACKGROUND

    Atopic dermatitis (

    AD
    ) and allergic contact dermatitis (ACD) are both forms of eczema and are common inflammatory skin diseases with a central role of T cell-derived IL-
    22
    in their pathogenesis. Although prostaglandin
    E
    2 (PGE2) is known to promote inflammation, little is known about its role in processes related to
    AD
    and ACD development, including IL-
    22
    upregulation.

    OBJECTIVES

    We set out to investigate whether PGE2 has a role in T cell-derived IL-

    22
    induction and development of ACD, which has augmented prevalence in patients with
    AD
    .

    METHODS

    T cell cultures and in vivo sensitization of mice with powerful haptens (oxazolone and dinitrofluorobenzene) were used to assess the role of PGE2 in IL-

    22
    production. The involvement of PGE2 receptors and their downstream signals was also examined. The specific effects of PGE2 during ACD pathogenesis were evaluated by using the oxazolone-induced ACD mouse model. Gene expression of PGE2 and IL-
    22
    signaling pathways was also investigated by using genomic profiling in human lesional
    AD
    skin biopsies.

    RESULTS

    PGE2 promotes IL-

    22
    production from T cells through its receptors,
    E
    prostanoid receptor 2 (EP2) and EP4. This is mediated by its downstream cAMP-PKA signaling and probably involves the transcription factor aryl hydrocarbon receptor (AHR). Selective deletion of EP4 in T cells prevents hapten-induced adaptive IL-
    22
    production in vivo. Importantly, blockade of endogenous PGE2 production by a COX inhibitor indomethacin or deletion of EP4 in T cells limit atopic-like skin inflammation in the oxazolone-induced mouse ACD model. Moreover, both PGE2 and IL-
    22
    pathway genes were coequally upregulated in human
    AD
    lesional skin but were down-regulated after treatment with betamethasone or ultraviolet
    B
    (UVB) radiation, both common therapies for
    AD
    .

    CONCLUSIONS

    Our results thus define a crucial role for PGE2 in promoting ACD by facilitating T cell-derived IL-

    22
    production.
