Kunitz-type chymotrypsin inhibitor CIb1 of silkworm
Bombyx mori is a basic peptide consisting of 62 amino acid residues. To investigate the physiological roles of this peptide, we cloned it for genomic structure analysis. CIb1 cDNA was used as a probe to screen a BAC sub-library. One positive clone containing the upstream sequences was isolated and the sequence result showed that CIb1 gene consists 3 exons spaced by 2 introns. Consensus TATA and CCAAT boxes were identified in the 5'-flanking region. Other binding sites for transcription factors such as NF-kB, GATA, C/EBP, COUP-TF/HNF-4, SRY, and HOXA3 were also detected. Southern blotting suggested a single copy of CIb1 gene in the silkworm genome. Northern blotting indicated that the expression of CIb1 gene is transcriptionally regulated during development and apparently tissue specific. Together our data, the transcription factors putatively responsible for the roles of CIb1 in silkworm hemolymph were discussed.

Seasonal morph determination of the butterfly
Eurema hecabe is controlled by photoperiod experienced during larval stage, but its hormonal regulation has been unknown till recent years. Recently, Ishii et al. (2002) demonstrated that summer moph producing hormone, which is a neuropeptide, is involved in the determination of seasonal wing morph. Therefore, we tested whether diapause hormone (DH) of
Bombyx mori is effective for the summer morph production of
E. hecabe As a result, diapause hormone failed to induce summer morph when applied to autumn morph producing pupae, but unusual and curious melanization was induced on the forewing upperside of the adults obtained. Furthermore, DH may have a possibility to stimulate ovarian maturation in diapause-destined autumn morph females although we must do further experiments. This is the first report where DH effect is found in other insect species except
B. mori.

PCR against genomic DNA from individuals of
Bombyx mandarina distributed in three areas of east Asia, i.e., Japan, Korea and China, was performed with the inverted terminal repeats of the
Hyalophora cecropia MLE as a primer. Each specimen gave several bands with different sizes. One individual from China had a discrete 4.2-kb band containing a
BMC1 retrotransposon, which is a unique fragment detected in almost all of the silkworm races and strains. Thus, the 4.2-kb unit might provide a clue for addressing the issue about the segregation of
B. mori genome. From the phylogenetic analysis, MLE (
BmTNML ) of 4.2kb band and 2.5kb band from the specimen of Fukuoka had high similarity each other supporting the idea that this type of MLE is an ancient type before segregation of
B. mandarina among different chromosome numbers, i.e.,27 and 28.

Ultra-fine transparent silk Fabrics were woven using 10 denier silk filaments produced by a strain of silkworms breed at the Okaya branch of NIAS. Using these ultra-fine filaments, fabrics having a weight/area ratio of 12-13g/__m2__ were able to be produced. Several other silk fabrics were woven with synthetic filaments that were removed by heat treatment, leaving only the silk. The characteristics of these fabrics will be discussed.

Except for hemocytes and fat bodies, insect immune tissues have not been carefully determined. To identify the organs that express cecropin B in silkworms, we constructed a transgenic silkworm that was transformed with the green fluorescent protein (GFP) gene under the control of an 800-bp CecB2 promoter region and followed by the 540-bp 3'-UTR of CecB2 using a transposon, piggyBac. In the transgenic silkworms, little GFP was detected in normal larvae, while a strong GFP signal was observed in the fatbodies, hemocytes, and tracheae in the head and testis after bacteria injection. Our finding that infection induces an antibacterial peptide in generative cells in insects is new.
