Abstract
Diflunisal, a new salicylate analogue, is a potent, long - acting anti - inflammatory drug that reduces serum uric acid level (SUA). We investigated the action mechanism of hypouricemic effect of diflunisal in terms of the 4 compartment model system to clarify different actions on uric acid metabolism between diflunisal and aspirin. Diflunisal(500mg)induced a significant decrease in SUA(0.7±0.4mg/dl)with a concomitant increase in urinary urate excretion(170±30mg)for 6 hours. This diflunisal induced uricosuria was markedly inhibited by 4 g of pyrazinamide, but was not additive to that by 100 mg of benzbromarone. These findings suggested that low doses of diflunisal reduce SUA through its uricosuric effect, which results from inhibition on postsecretory reabsorption without suppressing secretion.
