Abstract

As regards pharmacologic actions of promin and related preparations on leprosy, the author obtained from experiments made in leprosy partients and animals, following results:
1) Antibacterial effects of promin, promizole, diasone and methyl-promizole In vitro diasone, promizole and methylpromizole showed nearly equal bacteriostatic action upon tubercle bacilli (Typus humanus H 37 R V), while promin is in its effect a little inferior to them. No difference was seen between promin and diasone in their bacteriocidal action. Promin seemed to be more effective than others on rat leprosy.
2) Absorption and excretion: When intravenously or orally administered in man, all of these preparations are absorbed and excreted in urine rapidly, although the excretion rate only of diasone is somewhat lower. Promizole is a little more acetylated in human body than others.
3) Cbncentrations in orga-tissues: The concentrations of each preparation in nerve-tissues are Iower than those in skin. tissues. promin accumulates in organ-tissues somewhat slower than others. Contents of promizole and diasone in various organ-tissues are higher than those of promin
4) Influences on blood: Though each drug caused anemia, influence of promin was somewhat slighter in comparison with those of others. Resistance of erythrocytes decreases after giving each drug continously for 1 to 2 weeks.
5) Influences on circulatory organs: No change was seen in blood pressure. The capillary permeability of skin has a tendency to increase after administration of all drugs, while microscopic findings of skin capillaries and skin temperature showed no change.
6) Influences on reticuloendothelial system: In animal experiment promin activates the function of this system slightly. No definite result was obtained in experiments with other preparations.
7) Influences on metabolism: No change was seen in the basal metabolism, while protein-, hydrocarbon-, calcium- and cholesterinmetabolism are all disturbed temporarily.
8) Influences on vitamin metabolism: Decrease of blood level of vitamin B1 was observed after administration of all drugs.
9) Influences on liver; No remarkable change were seen in Takada's reaction, hepatosuiflein-test and other examinations,
10) Influences on skin reactions: Q. R, Z. and Mituda's reaction are not influenced.
11) Influences on acid-base balance: CO2 combining power of blocd decreases sometimes and it leads to acidosis.
12) Influences in phagocytosis of leucocytes and on electric mobility of erythrocytes The temporary increase of phagocytic funtion of the fomer was seen, while no remarkable change was to be found according to the latter.
13) Estimated from the results of slide-cell-culture, antibacterial effect of serum on tubercle bacilli is found to be not proportinal to the blood level of each, previously administered preparation.
14) The serum of patient treated with promin inhibits the Oxygen. consumption of liver of rat.
15) Diamino-diphenyl-sulfon and its derivates seemed to be generally antagonistic to P A B A-
16) Diamino-diphenyl-sulfon inhibits the oxygen consumption of emulsion of leproma:
