Renin Content in Subcellular Fractions of Normal Rat Kidney Extract

Abstract
Normal rat kidney of Wistar-King strain was cut into cortical, corticomedullary and medullary portion. The cortical extract was divided into subcellular fractions by the step-wise centrifugation method.
Enzyme activity of acid phosphatase, cytochrome C oxidase and glucose-6-phosphatase was determined in order to ascertain the nomenclature of each fraction.
Renin content in these extracts as well as the extract of whole kidney was determined. In the subcellular fractions, renin content in lysosomal fraction was 1.10±0.25μg, mitochondrial fraction 0.60±0.12μg, mitochondrial-microsomal fraction 0.18±0.01μg, microsomal fraction 0μg and soluble fraction 0.62±0.11μg of Ang. II per mg of protein. The study indicated that renin was located mainly in the lysosomal fraction in renal cortex.
