Abstract
This study was designed to investigate the response of one bolus injection of [Sar-1, Ile-8] angiotensin II under negative sodium balance.
Male Wistar-King hypertensive rats were made by constriction of the left renal artery with a silver clip. Two-kidney Goldblatt hypertensive rats were given either a regular or a low salt diet for 8-9 week after clipping.
The effects of arterial pressure by one bolus injection of 100 /μg/kg of [Sar-1, Ile-8] angiotensin II were studied about agonistic action and depressor response. Plasma renin concentration (PRC) was determined by radioimmunoassay.
In two-kidney Goldblatt hypertensive rats with a low sodium diet, PRC was increased significantly with a blunted agonistic action, and a marked depressor response.
The present study shows that the pharmacological effects of this angiotensin II analo-gue was influenced by the sodium balance and one bolus injection test was clinically feasible.
