Abstract

 We found that skin keratinocytes generate active cortisol through induction of 11β-hydroxysteroid dehydrogenase 1 (11β-HSD1) and clarified that they regulate skin aging and wound healing by inhibitory experiment using 11β-HSD1 specific inhibitors.

 We also reported that UV irradiation, hapten induced-skin inflammations or barrier disruption with acetone treatment were negatively regulated by 11β-HSD1 using whole body or keratinocyte specific 11β-HSD1 knockout mouse. We would like to review the recent advances in the regulatory mechanisms of maintenance of skin homeostasis by new homeostatic regulator, 11β-HSD1 in consideration of pharmacological and anti-inflammatory action of corticosteroid in allergic skin diseases including atopic dermatitis.
