Abstract
Spheroplasts of Plasmodiophora brassicae Woronin were successfully obtained from resting sporangia by digestion with Novozym 234 and/or chitinase. Digestion of the resting-sporangial wall by these enzymes was enhanced by a series of pretreatments with EDTA, 2-mercaptoethanol, protease and lipase. Electron microscopy of the spheroplasts showed that outer and middle wall layers of resting sporangia were removed by these pre- and post-treatments.
