Abstract

Nasal drops of emedastine difumarate using poloxamer (Pluronic) were evaluated in vitro and in vivo. The mixture of Pluronic showed the appropriate gelation temperature and viscosity in comparison with the single use of Pluronic F-88, F-98 or F-127. The release rate of the drug from the mixture of Pluronic F-88 and F-127 was affected by the Pluronic concentration and mixing ratio. The higher concentration of Pluronic mixture and higher ratio of Pluronic F-127 in the mixture indicated the lower drug release rates from the Pluronic mixture. The residual drug amounts in the nasal cavity, Tmax and AUC0-∞, using the mixture of Pluronic F-88 and F-127 (ratio 1:1, concentration 20-30%) were about 4-5, 1.7-3.5, and 1.7-2.5 times higher than those using the solution, respectively. From these results, the Pluronic mixture would be a useful base for nasal drops of emedastine difumarate.
