Synthesis and Radioprotective Activities of Water Soluble Vitamin E Derivatives

Abstract
[Back Ground] Tocopherol-Mono Glycoside(TMG) is a water soluble Vitamin E Derivative,and it has High Bio-defensive activity. However its cost of Synthesis is quite expensive.
[Purpose] Lower cost of the synthesis of water soluble Vitamin E Derivatives, and their radioprotection activities are reported.
[Materials]Tetra Methyl Chromanole Carboxilic Acid(TMCCT:Trolox) Inorganic Alkaline metal Hydroxide ,Alcohol were used for the experiment.
Synthesis (1) TMCC-Salt:Uni-molecular mixture of TMCC(2g) and NaOH were wormed up,and TMCC-Na Salt(2.6g) was obtained.
Synthesis(2) TMCC-Ester:Uni-molecular mixture of TMCC and alcohol such as Glycerol was heated at 65 degree/40min,and obtained TMCC- Glycerol Ester.
[Solubility in Water]
TMCC: 0.02%
TMCC-Na: 0.13%
TMCC- Glycerol Ester: 0.625%
Radio-Protective Activities: The radiation induced damage(ring open) of Plasmid DNA irradiated 25 Gy, TMCC-Na(1mmol), TMCC-Glycerol Ester(1mmol) were measured by Comet method as follows:
[After 25 Gy of irradiation] no TMCC Derivatives: DNA damage (46%), Protection (54%)
TMCC-Na:
TMCC-G Ester: DNA damage (5%), Protection (91%)
[Conclusion] 1.Radiation Induced Plasmid DNA Damage were Protected by TMCC-Na,or TMCG Ester
2.Radioprotective Activity of TMCC-G Ester was the highest.
