Relationship among Three Electrophoretic Characteristics of β-Amylase in Soybean and Wild Soybean Seeds

Abstract
β-amylases in soybean seeds can be separated by gel electrophoresis into two(Sp1a and Sp1b) variant bands which are controlled by two codominant alleles, and by gel isoelectric focusing into seven isozymes which belong to two zymogramatical types (high and low pI type). However the relationship between both characteristics is not clear. Therefore crude seed extracts from soybean and wild soybean were subjected to SDS-polyacrylamide gel electrophoresis and β-amylases were renatured and detected on SDS-gel in order to compare their molecular size. Soybean and wild soybean β-amylases were separated into high mobility(HM) and low mobility(LM) groups by SDS-gel electrophoresis. Under these experimental conditions, sweet potato β-amylase used as a standatd was once dissociated into subunits and then renatured into the tetrameric active from. twelve lines of soybean and wild soybean were examined for three electrophoretic characteristics of the enzyme. The results showed that the Sp1a variant band consisted of high (H) pI type isozymes with a low mobility(LM) and the Sp1b band was composed of low (L) pI type isozymes with a high mobility(HM).
