Abstract
We observed plasma cell neoplasms or dysimmunoglobulinemia in BALB/c mice after intraperitoneal injections of Freund's complete or incomplete adjuvants.
The mice, 8 weeks of age, received three times of intraperitoneal injections of 0.5ml of the adjuvants. Development of ascites and swelling of intraperitoneal lymph nodes were seen soon after the treatment. In 16-20 weeks after the initial injection of the adjuvants, we observed by immunoelectrophoretic analysis pathological changes in immunoglobulins of mouse serum. The findings in immunoglobulins are as follows: bifurcation of 7Sγ1 and 7Sγ2 precipitate lines is strong or a split of the arc of γ-precipitate line is seen. These changes are seen often in mice 20 weeks after the initial injection of the adjuvants.
When these changes in immunoglobulins appeared, proliferation of plasma cell was histologically found on intraperitoneal tumor tissue. This histological change is not of plasmocytoma, but a reacting, inflammatory change and a lipogranulomatous reaction. In 42 weeks after the initial injection of the adjuvants, abnormalities in immunoglobulins of mouse serum were found in most of the animals. This is marked in BALB/c mice injected with Freund's complete adjuvant. Of the pathological findings in immunoglobulins of mouse serum by immunoelectrophoretic analysis, we observed the typical monoclonal pattern of 7Sγ1 and 7Sγ2. The mice which showed a monoclonal pattern and other changes in serum immunoglobulins excreted Bence Jones protein in urine.
When the changes in immunoglobulins of mouse serum developed as a monoclonal pattern, mice were sacrificed and intraperitoneal tumors were examined microscopically. These tumors were plasmocytoma consisting of undifferentiated, immature and abnormal plasma cell.
For examining the process from the polyclonal pattern to the monoclonal one, changes in serum immunoglobulins of mice, 30 weeks after the initial injection of the adjuvant, were observed. At this time, changes in immunoglobulins of mice showed variable findings. These findings were all found in 7Sγ1 and 7Sγ2. Especially, a strain of 7Sγ1 precipitate line appeared.
In addition, for examining the changes in immunoglobulins of the same individual, 7Sγ1 precipitate line was strong and bifurcation of 7Sγ1 and 7Sγ2 were marked in 16 weeks after theinitial injection of the adjuvant. Afterwards, in 30 weeks, 7Sγ1 precipitate line was strained, and at last in 42 weeks, a monoclonal pattern appeared in 7Sγ1 area.
Other immunoglobulins of mice, i. e. γA and γM, did not develop a particular change.
At present, we are going to study on the possibility of transplantation of these plasma cell tumors induced by the adjuvants and on the activity of antibody of these myeloma proteins. Especially, the activity of antibody of myeloma protein induced by the adjuvants is important with regard to the problem whether the myeloma protein is an abnormal protein.
The above experiments showed changes in immunoglobulins of mouse serum from the polyclonal to monoclonal pattern, in accordance with histological changes from inflammatory to tumorous plasma cell proliferation.
