Abstract
The determination of serum glucose with o-toluidine using a glacial acetic acid medium is sensitive, precise and accurate enough for the requirements of a clinical laboratory. It is not convenient for a routine work because g acial acetic acid has irritant vapor.
Therefore, a new method of determination of serum glucose with o-toluidine using no glacial acetic acid (TGBM method) is presented. The procedure employs a mixture of glycollic acid, benzyl alcohol and methyl cellosolve instead of glacial acetic acid.
The new reagent (TGBM reagent) is non irritant, colorless, and stable for at least several months when stored at 4° in a brown bottle.
The coefficient of variation was ±2% by the TGBM method; recovery experiments gave a mean yield of 100%. Good agreement was observed between the TGBM method and the UV method using hexokinase-G-6 PDH or OTB method.
