Abstract

 The purpose of this study was to analyze environmental DNA(eDNA) of ayu (Plecoglossus altivelis) at the Miomote river and the Kuma river and to obtain knowledge about whether the habitat and a spawning place of ayu can be evaluated by environmental DNA and gained the following findings. (1)As a result of comparing the environmental DNA concentrations of Ayu in Riffle, run and pool, environmental DNA concentrations in riffles are the highest, followed by in runs and pools. From the previous study, it is known that the population density of Ayu is higher in the order of in riffle, run and pool, and environmental DNA also showed the same tendency. In addition, since the environmental DNA concentration tends to increase in places where the flow velocity is high, it is possible to more accurately evaluate habitat of ayu by combining environmental DNA concentration and hydraulic physical quantity . (2)The environmental DNA concentration of the spawning bed is very high and it is possible to differentiate it from the environmental DNA concentration caused by inhabitation and to evaluate the local spawning bed.
