Abstract
We investigated the regulating mechanism of LAK cell induction in terms of perforin production by a semiquantitative method using universe microphotospectrometry. Only CD 8+ lymphocytes showed perforin production after IL-2 stimulation. Perforin productions of CD 8+ lymphocytes were markedly up-regulated by addition of monocytes, TNF-α, or transferrin to culture medium with IL-2. These findings indicate that up-regulation of LAK activity is due to enhanced production of perforin, and suggest that study of perforin production in cancer patients may be important in immunotherapy using IL-2 and LAK.
