Abstract
Rapid determination methods for natural cytokinins in the plant was developed by LC/MS/MS. An analysis was done about the solvent which 80% extracted by the methyl alcohol solvent from cress which grew for 7 days in light and dark condition. Mobile phase constituted 0.1% Acetic acid and acetonitrile. Cytokinis was eluted using a linear gradient from 5% acetonitrile to 50%(35 minutes, 200 ml/min). Benziladenine was used for the internal standard, and a good straight line was shown about zeatine(Z) and zeatine riboside(ZR) in the absolute quantity in the range of 10 pg from 2000 pg. These determination limit were 9.0 pg(Z) and 10 pg(ZR), limit of detection(S/N<3) was 4.5 pg(Z), 5 pg(ZR).As the result, the existence of zeatine in cress(light condition) was 1.5 ± 0.018 ng/freshweight g which determination had by this procedure.
