Abstract
Understanding of the mechanisms of anther development and its abortion under chilling is important to solve some agricultural problems, including chilling injury in rice. We used proteome analysis for characterisation of rice anther proteins that would be involved in the mechanism.
Rice anthers collected at booting and heading stages were homogenized with Tris buffer (pH7.4). The extracts were applied to IEF and SDS-PAGE. Lectin blotting analysis using 7 kinds of lectins (ConA, LCA, PNA, PHA-E, RCAI, UEAI, WGA) showed mannose-binding lectins (ConA and LCA) strongly reacted with a lot of proteins from anthers. Number of the Con-A reacted proteins were much in anthers at heading stage rather than microspore stage. Among them, 80kD and 44kD proteins accumulated specifically in microspore stage, moreover each protein significantly decreased and increased respectively by the chilling treatment. We will report the structural analysis of the 80kD protein in this meeting.
