Abstract
AP2/EREBP family consists one of the largest groups of transcriptional factors in plants. Recently we isolated AtEBP by functional screening of yeast. Objective of this study is to understand the role of AtEBP in plants.
As a first step, we investigated the cellular localization using GFP-AtEBP. Results indicated that the fusion protein located in the nucleus in plant cells. We generated transgenic tobacco overexpressing AtEBP and these expressing GUS gene fused with a promoter containing GCC-box, respectively. The resulting GUS activity of the crossed lines indicated that AtEBP can function as an transcriptional activator through GCC-box in vivo. By using Arabidopsis plants overexpressing AtEBP, we could suggest that AtEBP regulates several defense genes.
In order to understand the position of AtEBP in ethylene signaling, we carried out Northern analysis using ethylene-related mutants. The results suggested that the position of AtEBP in ethylene signaling is different from that of ERF1.
