Abstract
To identify the genes concerning the organic and/or inorganic substances which are contained in xylem sap, we are trying to identify the genes which express in the tissues around the root vascular bundle by a gene trap procedure using Arabidopsis. We made 51,000 gene trap lines which are randomly inserted by T-DNA including the GUS reporter gene in the genome of Arabidopsis. Within these lines, 35 lines expressed GUS in the steles of roots. 26 lines expressed GUS only in steles, 1 line expressed in endodermis and stele, and 8 lines expressed in various tissues of roots. Only 2 lines expressed GUS specifically in the steles of the roots. These root-stele-specific lines might be inserted by T-DNA in the genes related to the biosynthesis and/or loading of the xylem sap substances.
