Isolation of PS II complex from Acaryochloris marina

Abstract
A chlorophyll d dominated cyanobacteria, Acaryochloris marina was recently isolated from a species of colonial ascidians and thalli of the red algae. The PS I RC was isolated from A.marina, however, PS II complex has not yet been isolated. The nature of the primary donor of PS II of A.marina is under debate, because it is difficult to isolate RC complex of PS II. In order to analyze the mechanism of primary charge separation, we attempted to isolate PS II RC. Thylakoid membrane of A. marina was solubilized using detergent of Triton X-100. The supernatant after the solubilization was loaded onto hydoroxylapatite column, and then PS II fraction were reloaded onto an anion-exchange column. After loaded to anion exchanged column, CP47-RC and RC complexes were eluted by NaCl gradient. The Qy spectrum of CP47-RC and RC peaks at 700nm and 704nm, respectively. The further characterization is in progress.
