Abstract
Paper quality is affected by the cell wall quality in wood tissues. We needed to expand our understanding of the cell wall formation process; therefore we used the oligo-microarray analysis and have been identifying transcription factors involved therein. We are now trying to characterise the factors in detail, using a modified yeast one-hybrid system.
In our experiment, we used the promoter region itself (approximately 2.0 kb in length) of the gene rather than tandem copies of a cis-acting element to construct the reporter vector, since the properties of the promoter regions of Eucalyptus genes remain unknown. Our results revealed the possibility of detecting protein-DNA interaction. We are currently using this system to detect interactions between promoters of Eucalyptus cell wall formation related genes and their transcription factors.
