Abstract
Arabidopsis ASML1/WRI1 is an AP2-type transcription factor identified from wrinkled 1 mutant with defects in seed oil accumulation and from asml1 mutant with enhanced expression of sugar-inducible model reporter gene. Microarray analyses with several mutants suggested that ASML1/WRI1 promotes expression of genes involved in sucrose metabolism and fatty acid synthesis. In fact, ASML1/WRI1 trans-activated expression of reporter genes under the promoter of various genes involved in fatty acid synthesis in protoplast transient assay. In this study, we found that sequences 300 bp upstream from ATG in SUS2, Pl-PK, KAS1, BCCP2 promoters are required for trans-activation by ASML1/WRI1. We also found that recombinant ASML1/WRI1 specifically binds to these DNA fragments. From these results, we suggest that ASML1/WRI1 directly activates genes involved in fatty acid synthesis.
