Abstract
TMBP200 of tobacco BY-2 cells is a member of the highly conserved family of microtubule associated proteins including Xenopus XMAP215, Human TOGp and Arabidopsis MOR1/GEM1. To know functional domains of TMBP200, we are attempting to examine subcellular localization of truncated polypeptides of TMBP200 in BY-2 cells. Previously, we reported that GFP tagged full-length TMBP200 (1-2029 aa) decorated MTs throughout the cell cycle and GFP::TMBPN1 (1-269 aa) and GFP::TMBPC1(1402-2029 aa) were weakly associated with spindle/phragmoplast MTs but did not decorate cortical MTs. Here, we have established cell lines that express GFP tagged TMBPN2-C1 (270-2029 aa), TMBPN3-C1 (586-2029 aa), TMBPN4-C1 (838-2029 aa), TMBPN5-C1 (1101-2029 aa), TMBPΔPMBD (Putative MT-Binding Domain)(1-1787 aa) or TMBPN1-N5 (1-1401 aa). Currently, we are examining subcellular localization of the above GFP tagged polypeptides and will discuss the MT-binding domain(s) of TMBP200.
