Microscopic detection of individual microbial cells in soil was done by using fluorometry and image analysis. A derivative of the fluorescein diacetate was used for staining all species of microorganisms in soils, and a method of image analysis conjunct with spectrofluorometry was applied to the detection of methane-producing bacteria in the soil. Merits and demerits of these method were compared with those of the molecular biological stainings such as (fluorescence) in situ hybridization.
