Many proteins including 30 kDa protein, storage protein, chymotrypsin inhibitor and so on had been shown that they were synthesized in the larva stages of fat body of silkworm, secreted into hemolymph, and then transferred between different organs and cells with the physiological development. Protein profiles of fat body in the developing silkworm larvae were analyzed by means of two-dimensional gel electrophoresis (2-DE) and mass spectrometry (MS). The analysis of fat body gave rise to a protein map comprising approximately 1000 spots detectable by silver staining following high 2-DE with a pH range of 3-10 and a mass range 12-100 kDa. To obtain landmarks for fat body, 142 spots were identified by peptide mass fingerprinting using MALDI-TOF-MS. Spot matching, alignment and quantitation was performed with PDQuest software. Comparison of temporal profiles of fat body protein in the developing silkworm larvae revealed significant changes in the expression of proteins during development.
