Abstract
Three cherry trees (Prunus × yedoensis Matsumura. ; Somei-yoshino) in Miyota, Nagano Prefecture, were applied for the core sampling using the increment borer. Three core samples were divided into 6 pieces, respectively, and genomic DNA was extracted from each small piece. Internal transcribed spacers of basidiomycetes were amplified by PCR with the obtained genomic DNA as a template, and the signals of amplified DNAs were observed on agarose gel. When the amplified DNA fragments were analyzed by denaturing gradient gel electrophoresis, several basidiomycetes were identified. Especially, Fomitella fraxinea (Synonym, Perenniporia fraxinea ) was detected in most of samples, suggesting that the fungus is the dominant wood rotter against cherry trees in this area. The constitution of fungal community differed according to the piece in the core sample. The fact clearly indicates the localization of fungi in the stem of cherry tree.
