Abstract
Bone-like hydroxyapatite/collagen nanocomposite sponge (H/C sponge) was prepared. MG63 cells were seeded on/in the H/C sponge and cultured under a pressure/perfusion condition. After harvested the cells, histological evaluation, total DNA content measurement, alkaline phosphatase (AlkP) protein analysis and osteogenic gene expression analysis by real-time PCR were carried out. Collagen sponge was used as a control. Total cell number on the H/C sponge was 1/2 of the collagen sponge, because pore size of the H/C sponge was larger than the collagen one and surface of the H/C sponge was rough. However, AlkP protein activity and gene expression of cells on the H/C sponge was better than those of cells on the collagen sponge. Subsequently, the H/C sponge could be useful for bone tissue engineering scaffold.
