Diversity of Fimbriae from Bacteroides (Porphyromonas) gingivalis and Reactivities to Periodontitis Patients, Sera

Abstract
Fimbriae isolated from Bacteroides (Porphyromonas) gingivalis strain SU 63 which was different from fimbriae of B. gingivalins Srain 381 in the antigenicity of their autoclave extracted polysaccharide, was characterized and reactivity to sera of periodontitis patients were investigated.
The fimbriae of B. gingivalis SU 63 was eluted with Tris-HCl buffer containing 0.5 MNaCl on a DEAE Sepharose CL-6 B column chromatography. The purified fimbriae of B. gingivalis SU 63 produced a single band of a 70 KDa protein on SDS -PAGE and was observed as a 5 nm width fibroid structure on electron microscopy by negatively staining with 1% uranyl acetate. B. gingivalis SU 63 fimbriae did not react with monoclonal antibody against B. gingivalis 381 fimbriae on Westernblotting and enzyme linked immunosorbent assay.
Ten of twelve sera of periodontitis patients strongly reacted to B. gingivalis SU 63 fimbriae more than to B. gingivalis 381 fimbriae and the mean of reactivities was 2.5 times higher than that to B. gingivalis 381 fimbriae on enzyme linked immunosorbent assay.
These findings suggested that fimbriae of B. gingivalis SU 63 was different from fimbriae of B. gingivalis 381 in terms of molecular weight, antigenicity and reactivities to the sera of periodontitis patients.
