Abstract

Phosphonylation of inosine (Ino) has been achieved using inorganic diphosphonate (DP) in aqueous solution. The optimum condition for the phosphonylation of Ino with DP is Ino : DP = 1 : 10, pH 11 and 10 °C. 2´-Phosphonylinosine and 3´-phosphonylinosine were synthesized with the total yield of more than 95 %. The phosphonylated products of Ino were stable at 10 °C and pH 9. The reaction mechanism of Ino with DP was discussed.
