Most velvet proteins were composed of components derived from blood when they were electrophorically detected and compared. The N-terminal amino acid sequences on proteins specific for whole velvet and the skin parts of velvet after detected by SDS-PAGE were determined, and proteins specific for whole velvet were identified as fibrillarin (K-Q-P-G-G-P-N-G-P-G-G-R), cuticular collagen and collagen (K-Q-P-G-G-P-G-G-P-G-G-G-G-G). Also, a protein specific for skin parts of velvet was identified as a hypothetical protein (S-L-P-N-A---L-Y). The velvet major proteins, which were detected by denaturing micro two-dimensional electrophoresis (Type IV 2-DE), were analyzed by Matrix-assisted laser desorption/ionization-time of flight-mass spectrometer (MALDI-TOF MS). As a result, velvet major proteins were identified as keratin and serotransferrin. Protein components specific for velvet were identified as collagen alpha 2 (I) and keratin.
